The ultrastructure of the prolamellar body and its gradual development into a thylakoidal system have been studied in etiolated seedlings of Salvia splendens L. exposed to continuous light for various periods of time (from 60 seconds to 36 hours).

Autoradiographic study of the incorporation of precursors of polysaccharides and lipids in microsporocytes and tapetum of « Rhoeo discolor » Hance.OnRhoeo discolor, the localization and the level of polysaccharide and lipid synthesis were determined in the evolving pollen mother cells (PMC) and plasmodial tapetum by applications of3H-glucose,3H-myo-inositol,3H-acetate,3H-choline and14C-meva lonate on excised anthers during variable time intervals.

摘要:

The application of a C-banding technique to mitotic chromosomes of Rosa rugosa ‘Scabrosa’ resulted in poor resolution of bands, but facilitated the detection of a B-chromosome. The presence of a B-chromosome may be associated with the long period of flowering exhibited by this clone.

Cytology of the tetraploid cytotype ofAllium chinenseG. Don is described. This species has 32 chromosomes in its somatic cells. The male meiosis is characterized by the formation of multivalents, bivalents and univalents resulting in aberrant segregation of chromosomes at anaphases. This and the heteromorphicity in the somatic chromosomes is a pointer towards its allotetraploid nature. Pollen stainability is extremely low and the pollen grains have varying number of chromosomes.

Chromosomes of unfixed root tips hydrolysed for 7 minutes and stained by the Feulgen method show band patterns paralleling in reverse style those shown by Q-staining inScilla sibirica, Ornithogalum montanum, Vicia fabaandBrimeura amethistina: Q-bands of reduced fluorescence appear deeply stained inScilla sibirica, while brightly fluorescing Q-bands appear as colourless gaps inOrnithogalum montanum, Vicia faba and Brimeura amethistina.Whether such phenomenon is caused by condensation or loose packing of DNA at band level or simply to differential reactivity of DNA to the Feulgen stain is briefly discussed.

Meiosis in a double-ditelosomic plant of Pearl Millet with 2n=12+4 telocentric chromosome was studied at PMC meiosis. The telocentric chromosomes were found to be corresponding to the 5th chromosome of the normal complement and were designated as 5L and 5S. The mode of transmission of these telocentrics was studied through reciprocal crosses. Origin of the double-ditelosomic condition and the possible use of these telocentrics in chromosome mapping were discussed.

A critical analysis of karyotype and meiosis of the different varieties of the species Petroselinum crispum (Mill.) A.W. Hill collected from India and abroad showed normal bivalent formation with considerable irregularities in meiosis. The probable causes of the chromosome instability at meiosis have been discussed. The chromosome study of these varieties showed that the proportion of sub-metacentric and metacentric medium chromosomes in the complement is higher in comparison to the longer ones. The most remarkable feature of its karyotype is that it is graded. Depending on the gross morphological similarity the number of chromosome types has been found to be common to all of them (A-F). Type A (chromosomes with supernumerary constriction) is entirely absent in the imported variety. In spite of the basic similarity, in pattern of chromosome morphology the varieties differ in minute details of karyotype indicating that the structural alteration in the chromosomes have played an important role in their evolution.

This study gives the results of investigations made on the ultrastructure ofChenopodium amaranticolorCoste & Reyn foliar trichomes. These hairs, when localized on subapical leaves, have a pigmentation which is due to the presence of betalains. This investigation has given information both on the ultrastructure of plastids in pigmented hipsophylls and on the ultrastructure and development of this trichome, which for many aspects results similar to the « hair of salt ». PTAC staining has evidenced, both on stalk and bladder cell, the presence of plasmalemma invaginations that seem to originate vesicles that maintain PTAC positivity. PTAC positive vesicles are therefore present in the cytoplasm of both hair cells; some of them take connection with the bladder cell tonoplast and others are close to the cell wall. Cytochemical pyrantimonate localization of cations has also been employed: reaction products are found in cytoplasm, mitochondria, chloroplasts and vacuole. The presence of hairs with gland-like ultrastructure, of vesicles behaving like plasmalemma and of ion localization are discussed.
