Monocyte‐regulated Hyporesponsiveness of Human Cord Blood Lymphocytes to OKT3‐monoclonal‐antibody‐induced Mitogenesis |

OKT3 monoclonal antibody recognizes surface antigenic structures present on all human mature T lymphocytes and is mitogenic for resting peripheral T cells. Recent reports suggest that these structures are linked to the specific antigen receptor of the T cells and play an important rule in T‐cells activation. We have tested the mitogenic action of OKT3 on resting lymphocytes from human newborns, their mothers, and unrelated adults. We found that the prohlerative response of cord T cells to OKT3 is significantly lower than the response of maternal and adult cells at all doses of the antibody tested (5–1000 ng/ml). This difference was not dependent on culture conditions (source of serum, kinetics induced by the OKT3 antibody, or different proportions of adherent cells in peripheral blood mononuclear leukocytes), and could only to some extent be accounted for by differences in the proportions of OKT3‐binding cells between these populations. Removal of adherent monocytes largely diminished the OKT3‐induccd proliferation of maternal and adult cells, by an average of 70–80%. This report describes an effective method for the cloning ofMycobacterium lepraereactive T lymphocytes with Epstein‐Barr‐virus transformed autologous B cells as antigen‐presenting cells‐ The two advantages of this method are that it drastically reduces the number of autologous peripheral blood mononuclear cells I (<107cells) needed to obtain and propagate these T‐cell clones (TLC). and that it enables us to expand individual TLC to large numbers of cells (<108) Thus she major obstacles for the cloning of T lymphocytes‐especially important with regard to patients‐are bypassed. Thus far. TLC Srom three leprosy patients have been established. These TLC are HLA class H restricted in theirM. lepraedirected response. Monoclonal antibody K2H recognizes a human glycoprotein complex that is not rcstricted in haematopoietic lineages but is preferentially expressed on early haematopoietic cells, T cells, and monocytes This glycoprotein complex is made of constant ini 120,000–140,000M1, subunitnoncovalently associated at the cell surface with subunits of higherM1.ranging from 150,000 to 200,000 on different cell types. Internal labelling with [35S]methionine and pulse‐chase experiments revealed that in the cell the 120,000M, glycoprotein of this complex is also noncnvalently associated with a 100,000Mtglycoprotein, and that both glycoproteins are independently biosynthesized. Sendai virus vesicles were used as vehicles for the insertion of various cell membranes into different ceil lines. The transplantation efficiency was measured by using FITC‐labelled concanavalin A (Con A) or monoclonal antibodies against the T‐cell marker Lyt 2 and the major histocompatibility complex product H‐2Dhin a fluorescence‐activated cell sorter. Results indicate that it is possible to transplant mitogen responsiveness to certain cell types. Con A stimulation of T‐cell membrane transplanted BW 5147 showed that it is possible to induce a mitogen dose‐dependent T‐cell growth factor production. Soluble heterologous immune complexes (IC) were used to study the mechanism of IC binding to rat liver in vivo. Binding of IC to the various cell types of the liver, endothelial cells, hepatocytes. and Kupffer cells, was only inhibited by aggregated swine immunoglobulins. Binding was not inhibited by the absence of complement components. Intravenous injection of asialoglycoproteins. to block the galactose receptor, could not prevent IC binding. We conclude that Fc receptors play an important role in the binding of soluble heterologous IC to hepatocytes. endothelial cells, and Kupffer cells.

Supernatants from concanavalin A (Con A)‐stimulated chicken spleen cells were used to generate a long‐term cultured cell line from antigen‐primed chicken peripheral blood leukocytes. This line has been kept in continuous proliferation in vitro for more than 25 weeks. Morphologically these cells were lymphoblastoid and expressed class I and class II antigens of the major histocompatibility complex as well asT‐cell (but not B‐cell or macrophage) antigens. In addition they contained no peroxidase or non‐specific esterase activity, neither were they phagocytic. Proliferation of the line was totally dependent on exogenous T‐cell growth factor (TCGF) activity provided by the Con‐A‐stimulated spleen cell supernatant, comparable with the proliferation of Con‐A‐induced T‐cell blasts. TCGF activity from the supernatant was absorbed both by the long‐term cultured T cells and by Con A blasts, demonstrating the presence of receptors for the same TCGF species on the two populations. We have used the long‐term cultured cell line to characterize chicken TCGF further The molecular weight of the biologically active fractions found by gel filtration on Sephadex G‐100 was approximately 13,000 and isoelectric focusing showed chicken TCGF to have a pI of pH 5.9.
