A prospective study of influenza infection was carried out on 90 blood donors deficient for serum IgA as tested with double immunodiffusion. Half of them lacked IgA even by radioimmunoassay (RIA). A correlation existed between serum haemagglutination‐inhibiting (HI) antibody and resistance to infection, suggesting that the serum HI antibody was an important determinant of protection. The rate of infection as evidenced by a fourfold or greater rise in HI titre, was about the same in the RIA‐negative and RIA‐positive donors and only slightly higher than the corresponding rate in pregnant women and in a ship's

IgA‐deficient blood donors and their age‐ and sex‐matched controls were compared for the occurrence of complement‐fixing antibodies in serum against several viruses. The level in the IgA‐deficient persons was slightly higher against several respiratory pathogens (adenoviruses, type B influenza virus, parainfluenza virus, and respiratory syncytial virus) that give rise to localized infections, and against coxsackie B group of viruses. No corresponding difference was observed in mumps, varicella, and cytomegalovirus infections, where viraemia is a characteristic feature, or inMycoplasma pneumoniae

β2was extracted from human lymphocytes with nononic detergent and separated by affinity chromatography on concanavalin‐A‐Sepharose. The retarded part β2‐micorglobulin is assumed to be associated with HLA antigens. Among 54 human monoclonal IgA proteins, 5 inhibited the binding of labeled human monoclonal IgM (IgM Se and IgM Ba) to protein‐A‐containing staphylococci, whereas 48 did not have this ability. One protein (IpA Ha) showed discordant findings in the IgM Se and IgM Ba inhibition assays. Among the 54 IgA proteins, 49 were of the IgA1 and 5 of the IgA2 subclass All of the 5 IgA2 proteins inhibited the binding of IgM Se and IgM Ba to staphylococci, whereas the 48 proteins that reacted negatively in the two inhibition belonged to the IgA1 subclass. These findings show that the ability to react with staphylococci is a properly of the IgA2 subclass. IgM proteins may also be divided into two groups based on different reactivities with protein A. Lymphocyte subpopulations in the peripheral blood of 36 sarcoidosis patients and 36 age‐ and sex‐matched controls were determined, using the sheep erythrocyte rosette (SRBC‐R) technique and immunofluorescence staining with whole rabbit anti‐human immunoglobulin antisera. In the patient group the total number of lymphocytes, SRBC‐R lymphocytes, and IgM‐positive lymphocytes was reduced in comparison to in the control group. The Cγ2 homology region of rabbit IgG does not behave like a domain. Thus, there is no trans‐interaction between the two Cγ2 regions: instead there is an unusual cis‐interaction between Cγ2 and Cγ3 regions. The observations were made on the plasmin digestion products Facb (IgG minus the Cγ3 region) and pFc’(Cγ3 region), which did not dissociate under neutral conditions but dissociated in 3M guanidine solution (that is. cis‐interaction between Cγ2 and Cγ3). We describe a method enabling the identification of both lymphocyte class and morphology from a single microscopic slide. As a marker for B cells we used surface immunoglobulin. The surface‐Ig‐carrying cells were rosetted after polyvalent anti‐Ig treatment withStaphylococcus aureusstrian Cowan 1 (StaCw) and the cells were cytocentrifuged onto a microscope slide. The lymphocytes forming rosettes with StaCw were identical with cells expressing surface Ig studied by fluorescein‐isothiocyanate‐conjugated anti‐Ig. As a marker for T cells we used the acid α‐naphthyl acetate esterase (ANAE) histochemical marker. The cell smears were first stained for ANAE and subsequently counterstained to distinguish also cell morphology. The ANAE‐marker‐carrying cells were all included in the population of lymphocytes forming rosettes with sheep erythrocytes. Blood lymphocytes from the inbred rat strains AS and BN differ in the magnitude both of their in vitro proliferative response to different mitogens and of their in vivo antibody response to the mitogenic fraction of phytohemagglutinin (PHA). We have examined the segregation of in vitro responsiveness to PHA in (AS × BN)F1× BN backcross rats and have tried to correlate it with other characters that vary in backcross rats. In vitro responsiveness regulated by one or a few loci, is linked to the in vitro responsiveness to B lymphocyte mitogens and the in vivo antibody response to the mitogenic fraction of PHA, but is not linked to the major histocompatibility locus (Ag‐B) nor to the frequency of short‐lived, small Ig‐negative lymphocytes in blood. Lymphocytes from high‐responder rats have a shorter lag period before the onset of DNA synthesis in vitro than low‐responder rats, and possibly also a higher number of in vitro responding cells. To explain our findings, that the same gene og genes regulate in vitro responsiveness to different mitogens and in vivo antibody response to the mitogenic fraction of PHA, we suggest that the gene or genes act in an immunologically unspecific manner on regulation of lymphocyte proliferation in vitro as well

Phytohemagglutinin (PHA‐P; Difco) contains four immunogenic components. The antibody response in rats to two of these components is shown to be genetically determined; AS rats are high responder and BN rats are low responders to both antigens. Responsiveness to the two components segregates independently as autosomal, dominant traits in a manner that is compatible with a one‐gene hypothesis for both responses. The antibody response to the mitogenic fraction of PHA and to other mitogens. These responses are not linked to the major histocompatibility complex (MHC) of the rat.
