Peripheral blood from 15 young (20–30 years) and 15 aging (65–80 years) subjects was analysed for the proliferative response of T cells upon stimulation with non‐T cells in the autologous mixed culture reaction (AMLR) and allogeneic MLR, and for the proportion of monoclonal antibody‐defined lymphocyte subsets and monocytes. No significant difference was observed in the AMLR or allogeneic MLR between T and non‐T cells in aging and young subjects. However, when non‐T cells were further fractionated into adherent monocytes and non‐adherent B cells (B cells and null cells), the AMLR between macrophages and T cells was significantly (P<0.05) higher in young subjects than in simultaneously studied aging subjects. In contrast, the AMLR between T cells and B cells was significantly (P<0.05) higher in the aging subjects than in the young group. In the T‐T AMLR (using phytohaemagglutin‐stimulated T cells as stimulators), aging subjects had a significantly (P<0.05) lower proliferative response than simultaneously studied young subjects. In vitro addition of purified interleukin 2 reconstituted the T‐T AMLR to the base‐line T‐T AMLR in young humans. Two hundred and seventy‐seven patients with a broad spectrum of neoplastic diseases, including 10 classes of solid tumours and three classes of haematologic malignancies, were retrospectively surveyed, and from the same sample of plasma or serum their concentrations of serum amyloid A (SAA), serum amyloid P component (SAP), C‐reactive protein (CRP), and carcinoembryonic antigen (CEA) were measured. SAA levels varied from 105 ng/ml to 105,000 ng/ml, and mean SAA levels were higher in patients with metastatic tumours than in those with limited disease (P<0.001). Similarly, CRP levels varied from less than 8 μg/ml to 328 μg/ml and were significantly higher in the metastatic disease category. In contrast, SAP levels varied from 32 μg/ml to 120 μg/ml and showed no difference in patients with limited or metastatic disease, although an overall slight elevation was present. CEA levels were available in 150 patients and were significantly higher in patients with advanced lung or breast cancer than in patients with limited disease. The correlation between mean SAA and CRP levels was significant (r=0.74,P<0.001), suggesting that SAA originates as an acute‐phase protein rather than as a tumour cell product. Autologous rosette‐forming cells (ARFC) have been considered to be post‐thymic precursor cells. Since thymocytes and peripheral T lymphocytes differ considerably in lactate dehydrogenase isoenzyme pattern and in activity of the enzymes of the purine metabolism, we investigated the enzyme profile in the ARFC. The L‐lactate:NAD+oxi‐reductase analysis showed an isoenzyme pattern that closely resembled the pattern found in peripheral T lymphocytes and was totally different from the thymocytes. The levels of adenosine deaminase and purine nucleoside phosphorylase were identical to those found in the peripheral T lymphocytes and different from thymocytes. In our hands, the ARFC‐enriched suspension contained predominantly OKT4+and Tμ+lymphocytes. Rat spleen coll cultures exposed for 24 h to concanavalin A (Con A‐CM) contain, in addition to inierleukin 2 (IL‐2). factors that promote colony formation in vitro by mouse T cells (TCPA) and B cells (BCPA). TCPA and BCPA are separable on a Sephadex G‐75 column. TCPA has a molecular weight of 15,000 daltons and shows the same elution profile as IL‐2. Absorption studies with Con A‐activated T cells suggested that TCPA and IL‐2 are the same entity. BCPA has an apparent molecular weight uf 45.000 daltons and stimulates colony formation by B lymphocytes seeded at very low cell density (104–5 × 104cells/ml). In contrast to TCPA, BCPA can only be demonstrated in gel‐filtered material owing to the presence of B colony suppressor activities in crude Con A‐CM. Two B colony inhibitory activities were demonstrated by AcA 34 chromatography of crude Con A—CM with molecular weights of 80,000–130.000 and about 50,000. respectively. In a previous clinical trial we demonstrated that. by increasing the postoperative administration of methylprednisolone from 1.0 to 3.5 mg/kg/day. ihe onset of the first inflammatory rejection episode was significantly delayed and the size of the inflammation was reduced. The ‘high initial’ steroid treatment specifically depleted blast cells and macrophages from thein situinflammatory infiltraie. In ihis trial we demonstrate that the ‘high initial’ glueocorticosteroid administration significantly improves 1‐year cadaver allograft survival from 44% to 68% (P = 0.00.3) without increasing the number of complications. Although the ‘high initial’ steroid administration only partially overcomes the impact of HLA‐AB incompatibility, it seems to overcome entirely the impact of absence of blood transfusions. The ‘high initial’ steroid administration also makes the first episodes of inflammation easier to overcome: less steroids are needed to counteract the first rejection, and, as a consequence, only 30% more steroids were used in the ‘high initial’ versus the ‘low

Mice were immunized against fluorescein isothiocyanate (FITC)‐labelled human gamma globulin (HGG) in the absence or presence of different adjuvants. The immune response was assayed every other day with regard to both total Ig‐secreting cells and FITC‐specific plaque‐forming cells (PFC). The adjuvants influenced the type of immune response induced to the same antigenic determinant. Thus, addition of Freund's complete (FCA) or incomplete (FIA) adjuvant preferentially led to the secretion of IgG1 PFC of an average high affinity. Most newly appearing IgG‐secreting cells were also detected as FITC‐specific PFC. The use of lipopolysaccharide (LPS) as an adjuvant resulted in the induction of both IgM and IgG, particularly of the IgG3 and IgG2b subclasses. However, these antibodies had relatively low affinity, and a large number of total IgG‐secreting cells induced by LPS had no detectable FITC specificity. The FCA/FIA‐ and LPS‐induced responses to FITC‐HGG were additive when injected together, indicating that they act on distinct subpopulations of B lymphoid cells. The adjuvant response to LPS, but not the response to FCA/FIA, was totally absent in mice of the C3H/Hej strain, which are non‐responders to the polyclonal activating properties of LPS. Amyloid P component (AP/SAP), a glycoprotein, precipitated with purified snail galactans fromHelix pomatiaandAriania arbustorumin a dose‐dependent manner. Radiolabelled AP hinds to human peripheral blood mononuclear cells (PBMC), erythrocytes, and cells derived from human non‐T, non‐B acute lymphocytic leukaemia. The AP cell binding is specific in that it is dose‐dependent and can be blocked both by excess cold AP and byHelix promatiagalactan. although it cannot be blocked by an equal amount of the monosaccharide galactose. In vitro studies of human PBMC immune responses demonstrated that AP inhibits PBMC proliferation responses to mycobacterial purified protein derivative and to phytohaemagglutinin and the humoral, antibody response to pokeweed mitogen. The AP‐induced suppression of non‐specific antibody production by human PBMC was dependent on the time at which AP was added to the culture. AP was suppressive if added in the first 48 h of the 7‐day culture, and the suppression could not be reversed by washing the cells after the exposure to AP. It has been reported that lactale dehydrogenase (LDH) purified from testes (LDH C) of homotherms is not immunochemically cross‐reactive with somatic forms of LDH purified from heart (LDH B) or muscle (LDH A). On the basis of this premise, LDH C has been considered for use as a contraceptive vaccine. Data presented here indicate that mouse anlisera to either mouse or rat LDH C are cross‐reactive with LDH A and B purified from muscle and heart tissues of mice. However, rabbit antisera to mouse LDH C are not cross‐reactive with either mouse LDH A or B. Thus, the degree of cross‐reactivily is dependent on the species from which the immunogen LDH is purified, the antisera are derived, and the LDH used in the assay is purified.
