摘要:
A new method permits direct densitometric quantitation of different molecular‐size components obtained by thin‐layer Sephadex gel filtration. A transparent plate suitable for protein staining and transmission densitometry is prepared by over‐layering agarose after the filtration. Analysis of commercial γ‐globulin preparations with this method give better resolving capacity than ultracentrifugation or column gel filtration. Furthermore, this method is easily adapted by means of immunogel filtration to specific characterization of the molecular size categories. As many as nine IgG‐related components differing in size were demonstrated. Component categories containing as little as 0.06 γg could be detected. The error of the method varied between 1.9% and 1.3% for the different size categories. Good reproducibility was observed when the method was used for analysis of preparations with known qualitative and quantitative IgG

One hundred and five rabbits were immunized with a streptococcal group A or C vaccine About 50% of the rabbits died during the immunization, mainly of un‐known causes. The amounts of precipitating and agglutinating antibodies, antibody restriction, and γ globulin allotypes within the a locus were determined and correlated with one another. The mean precipitin and agglutinin titers of the rabbit antisera with some kind of restricted response in almost all case were higher than the titers of the antisera from the rabbits with a heterogeneous response, but the difference was significant in only a few cases No significant correlation was found between allotypic markers and the other variables mention

One hundred and five rabbits were immunized with a streptococcal group A or C vaccine. An anti‐antibody production in relation to the antistreptococcal antibody production was measured by two methods, the latex fixation test and the sensitized sheep erythrocyte test (the Waaler‐Rose test). The anti‐antibody production was correlated to the amount of precipitating and agglutinating antistreptococcal antibodies, antibody restriction, and γ‐globulin allotypes within the a locus. Anti‐antibodies measured by the Waaler‐Rose test showed no correlation with any of the variables mentioned. The anti‐antibody production measured by the latex fixation test showed a correlation with antibody restriction; that is, a positive latex reaction was found in a higher percentage of rabbits with a homogeneous or restricted heterogeneous antibody response to streptococcal carbohydrate antigens than in rabbits with a heterogeneous antibody response. Rheumatoid anti‐Gm(a) antibodies were shown to react with native Gm(a‐) IgG. This reaction takes place with the CH3 domain (pFc' fragment) of IgG and is not due to anti‐isotype specificities contaminating the test system. The reactivity of the Gm(a‐) IgG is greatly increased by heat aggregation, and this increase is clearly related to the amount of aggregates formed. Rheumatoid anti‐Gm(a) reacts with aggregated IgG1. IgG2. and IgG3 Gm(g+) proteins but not with IgG3 Gm(b+) or IgG4 proteins. Thus there is a clear specificity in the reactions with aggregated IgG of different subclasses. Quantitative hemagglutination inhibition studies in the AutoAnalyzer suggested that the antigenic determinants of native and aggregated Gm(a+) or Gm(a‐) IgG involved in the reaction with rheumatoid anti‐Gm(a) were very similar. The effect of mitomycin C on DNA synthesis and plaque formation in mouse spleen cells was studied in vitro, with special reference to the synergistic effect observed between some antigens and mitogens on lymphocyte stimulation. Mitomycin C inhibited DNA synthesis in doses that did not cause an extensive cell death It blocked almost completely a primary plaque‐forming cell response to sheep erythrocytes (SRC) and, to a great extent, PPD‐induced antibody synthesis detected against SRC. The synergistic effect earlier observed in vitro between SRC and PPD was not abolished in cultures where replication of DNA was depressed. A new purification method is based on a combination of conventional fractionation procedures (ammonium sulphate precipitation, gel filtration, and anionic‐exchange chromatography) and immunological removal of contaminating proteins. An in vitro method that permits the primary antibody response of chicken spleen cells to sheep erythrocytes to occur is described, and the characteristic of the antibody response are given. Significant but variable plaque‐forming cell (PFC) responses were obtained—that is, some animals showed no or a very weak response In these the addition of small numbers of allogeneic spleen cells much improved the PFC responses. The characteristics of this allogeneic stimulation are described. Germ‐free and conventional NMRI nine were injected unit3H‐thymidine daily for 30 days to label a high percentage of short‐lived (fast‐proliferating) small lymphocytes and cells with a long interphase–that is. long‐lived small lymphocytes Scintillation counting and autoradiographical procedures, including registration of labeling indexes and grain counts, were carried out on smears and sections of lymph nodes, spleen, thymus, bone marrow, thoracic duct lymph, and blood, and the number of blood and bone marrow lymphocytes was quantitated The distribution of labeled small lymphocytes in the various lymphoid smears and organs was similar in germ‐free and conventional mice, as were the numbers of blood and bone marrow lymphocytes The findings show that the percentage of short‐and long‐lived lymphocytes as well as the average turnover time of the latter cell category are identical in the two groups of mice studied This suggests that mechanisms other than antigen‐triggering are operating in the generation of shortlived and long‐l

ASalmonella typhiglycoprotein preparation in concentrations of 5 to 10μg/ml strongly inhibited the migration of peritoneal exudate cells (PEC), obtained from guinea pigs immunized withS. typhivaccine in Freund's complete adjuvant (FCA) PEC from animals injected with saline in FCA were also inhibited but to a lesser extent, whereas cells from guinea pigs that had received onlyS. typhivaccine or normal guinea pigs showed variable and weaker inhibition. PEC from all guinea pigs receiving FCA were strongly inhibited by 5 to 15μg PPD/ml. These PPD concentrations had no inhibitory effect on exudate cells from guinea pigs receivingS. typhivaccine only. TheS.typhiglycoprotein concentration (5 and 10μg/ml) primarily used in the migration inhibition studies had negligible inhibitory effect on the phytohemagglutinin (PHA)‐induced DNA, RNA, and protein syntheses in guinea pig lymph node cells.
