Peripheral blood mononuclear cells (PBMC) from normal human donors were cultured in Marbrook flasks in the presence of purified IgG or IgA myeloma proteins. The culture supernatants were tested for their ability to suppress pokeweed mitogen (PWM)‐ or Epstein‐Barr virus (EBV)‐driven Ig synthesis by normal PBMC, Two supernatants from PBMC cultured with IgG and one from PBMC cultured with IgA were tested and suppressed PWM‐driven Ig synthesis as measured by a reverse haemolytic plaque assay and by quantitation of the Ig secreted into the culture medium of the PWM‐driven cells. IgA and IgM antibodies were detected in rat milk after immunization wish ferritin in Peyer's patches (Pp) 1 day after parturition but not after intramammary gland or intravenous immunization. The antibody levels decreased from day 9 to day 17 of the nursing period and were undetectable during a second lactation period. Despite the absence of milk IgM antibodies after intramammary gland or intravenous immunization, the serum levels of the IgM antibodies were similar after all three immunization methods. IgA antibodies were not found in serum after any of the immunization methods. IgG antibodies appeared in serum and milk after Pp. intramammary gland, and intravenous immunization. Milk and serum IgG antibodies from all the Pp‐immunized animals decreased from day 9 to day 17 of the lactation period. After intramammary gland immunization, however, the IgG antibody levels increased in all the milk samples, but only in four of seven sera. The milk and serum IgG antibody levels were lower but still detectable during a second lactation period. BL13, a mouse monoclonal IgGl antibody raised against human B cells, blocked the function of the C3d receptor (CR2) and bound with high affinity (5x 108L M−1) to CR2 on B lymphoma cells. Following capping with the second antibody BL13 inhibited C3d‐dependent rosette formation of Daudi and Raji cells and C3b‐dependent CR2‐mediated rosette formation with B lymphoma cells, but did not inhibit CRI‐mediated rosettes between C3b‐bearing cells and peripheral blood lymphocytes. Competitive binding experiments between biotinylated BL 13 or anti‐CR2 antibody HB‐5 and unlabelled antibodies demonstrated that BL13 bound to an epitope that is distinct from that recognized In HB‐5, and closely associated with that recognized by monoclonal antibody ann‐B2, BL13 only reacted with some B cells and follicular dendritic cells in germinal centres in human lymph nodes, whereas HB‐5 strongly reacted with circulating B cells and bound to most cells in the follicles. We evaluated the molecular and physicochemical properties of surfaces formed by defined layers of block copolymers which were especially effective as adjuvants or in the induction of granulomas. The copolymers which were adjuvants formed hydrophilic surfaces with a large area. They bound protein in a way which left it particularly accessible to antibody and induced the activation of complement. Copolymers which induced granulomas, in contrast, formed hydrophobic crystalline surfaces. They bound less protein and did not activate complement, but were toxic for macrophages. Their surfaces were found to be similar to those of the mycobacterial glycolipid trehalose‐6.6‐dimycolate or quartz, in that they consisted of regular geometric arrays of hydrophilic and hydrophobic adsorptive domains. Native agarose beads (diameter 5–10 μ), activators of the alternative complement pathway, are slowly phagocytosed when incubated with human monocytes cultured under serum‐free conditions. Agarose beads preincubated with monocyte cultures and then transferred to new cultures are more easily phagocytosed than native heads. These results indicate that the phagocytosis of agarose beads depends on opsonization of the beads by one or several substances of monocyte origin. By using antihuman C3 antibodies. trypsin treatment, and sodium dodecyl sulphale washing, we were able to demonstrate C3b and iC'3b son the agarose beads. The molecules were covalently bound to the surface of the beads. We conclude that in vitro human monocytes produce and secrete the essential factors for activation and propagation of the alternative complement pathway (factors C3. B, D. H and I), which becomes evident with an external activator like agarose beads in the cultures. The study shows that serum from pneumococcal polysaccharide type III (SSSIII)‐immunized and nonimmunized mice that has been absorbed until devoid of anti‐SSSIII antibodies inhibits anti‐SSSIII plaque‐forming cells (PFC) but not antisheep red blood PFC in antibody plaque assay. A large fraction of the inhibitions activity can be absorbed by and eluted from anti‐SSSIII‐Sepharose, but not normal mouse serum‐Sepharose. It has low or no affinity for protein A and concanavalin A The titre of the inhibitor increases, with repeated immunizations and inhibits the anti‐SSSIII PFC of rats or mice of different allotypes. Twenty‐eight children infected withSchistosoma haematobiumandS. mansoniwere tested for natural killer (NK) cell activity in vitro using the myeloid/erythroid cell line K562 as target. In addition, the frequency of large granular lymphocytes (LGL) and the number of HNK‐1−lymphocytes were examined in peripheral blood. NK cell activity was found to be markedly reduced in most patients when compared with a group of healthy Caucasian individuals (P<0.005). Moreover, the impairment of NK activity clearly correlated with the intensity of infection, which was quantified by parasite ova excretion in stool and urine. Within the lymphocyte compartment the percentages of cells with the NK phenotype (HNK‐1+) were found to be normal, although the majority of patients exhibited decreased numbers of LGL (P<0.005). The absolute and relative frequencies of LGL and HNK‐1+lymphocytes by no means correlated with the parasite load. Polyclonal syngeneic, allogeneic. and xenogeneic and monoclonal syngeneic anti‐anii‐idiotypic antibodies have been produced against previously described monoclonal anti‐idiolypic anti‐bodies with specificity for monoclonal RTI alloantigen‐specific antibodies. The anti‐anti‐idiolypes could again be shown to be highly specific for the monoclonal anti‐idiotype used for the induction of the anti‐anti‐idiotypie antibodies and lo carry the same, or a very similar, idiotype as the original monoclonal idiotypic antibody used lo induce the monoclonal anti‐idiotypic. We have analysed the cytological structure of inflammation in the different parenchymal target organs during acute graft‐versus‐host disease after bone marrow transplantation in the rat. Inflammation (recovery of increased numbers of white cells) was recorded in the liver, skin, and lungs of the allograft recipient compared with the syngeneic graft recipient from day 5 onwards, accompanied by reduced recovery of inflammatory cells from the gut. The major cytological manifestation of the disorder was an increase in the total number of blast cells, large granular lymphocytes (LGL), and lymphocytes in the liver and skin, and of the number of blast cells and LGL in the lung.
