Reproductive involution produced by surgical removal of the olfactory bulbs and of the eyes from adult female rats was prevented by pinealectomy, superior cervical ganglionectomy or by bilateral transection of the medial forebrain bundle within the lateral hypothalamus. The activity of the melatonin-forming enzyme, hydroxyindole-O-methyltransferase, was diminished after sympathetic denervation of the pineal gland; this decreased activity of the pineal probably explains its failure to induce gonadal regression in blinded, anosmic rats. Interruption of the medial forebrain bundle may have prevented involution of the ovaries and associated sexual structures by at least two different means. The transections may have interrupted the inferior accessory optic tract, which transmits information about photic stimuli to the pineal gland or, alternately, they may have negated the pineal’s effect by interrupting afferents to the hypothalamus from cell bodies within the mesencephalon, which are responsive to pineal secretory product

Cytochrome oxidase, succinic dehydrogenase, and succinate oxidation were studied in the anterior, middle, and posterior hypothalamus during the sexual cycle in normal female rats as well as in rats rendered persistent-estrous by androgen treatment soon after birth. Similar values for succinate oxidation and succinic-dehydrogenase activity were found in the anterior hypothalamus of diestrous and androgenizedrats, and these values were significantly lower than those found in estrous rats. On the other hand, both biochemical parameters in the posterior hypothalamic area of estrous rats were similar to those observed in androgenized rats; such values were higher than those obtained in diestrous animals. No differences between the three groups of rats were apparent in the succinate oxidation and succinic-dehydrogenase activity of the middle hypothalamus. No significant changes were found in the cytochrome oxidase activity in the different hypothalamic areas of any of the groups of rats studied.

In intact gentled female rats, plasma GH levels decreased significantly 30 min after ether stress. Injection of a large dose of dexamethasone (400 µg/100 g) 30 min before, also decreased plasma GH levels. However, the suppressive effect of dexamethasone on plasma GH levels injected 3 h before seemed to be greater than that observed at 30 min. In bilaterally adrenalectomized rats, as in the intact rats, plasma GH levels decreased significantly either in response to ether stimulus or dexamethasone injection 3 h before. Resting plasma GH levels in adrenalectomized rats were lower than those in intact rats. Pentobarbital injection 15 min before in intact rats, not only increased plasma GH levels, but also restored the lowered plasma GH levels resulting from dexamethasone pre-treatment. It was concluded that increased output of adrenocorticoids following ether stress did not play an essential role in the reduction of plasma GH levels

The effect of chemical and enzymatic treatments on the biological activity of porcine follicle-stimulating hormone-releasing hormone (FSH-RH) was studied. FSH-RH activity was not affected by trypsin, pepsin, neuraminidase, carboxypeptidase A, aminopeptidase M and leucine aminopeptidase. However, incubation with chymotrypsin, subtilisin, papain and pyrrolidone carboxylyl peptidase abolished the FSH-RH activity as did treatment with hydrochloric acid (0.9 m, 1 h, 100 °C), diazotized sulfanilic acid, and N-bromosuccinimide. Nitrous acid, sodium metaperiodate, and ninhydrin did not affect the FSH-RH activity appreciably. Amino acid analyses of the purest FSH-RH preparation showed the following composition: His 1, Arg 1, Ser 1, Glu 1, Pro 1, Gly 2, Leu 1, Trp 1, and Tyr 1. These results and information obtained during purification procedures indicate that a basic polypeptide composed of ten amino acids possesses both LH-RH and FSH-RH activity

In male rats pinealectomized at 4–9 weeks of age and decapitated after survival periods of 2–6 weeks, the neurosecretory activity of the hypothalamic magnocellular supraoptic nucleus (SON) was studied as compared with the same activity in the nucleus of sham-operated controls. The distribution of thiamine diphosphate-phosphohydrolase (TPP-ase), an enzyme specific to the Golgi apparatus, was used as the parameter of this activity, employing a Hennig ocular for the purpose. Plasma corticosterone values were obtained using cortico-sterone-binding-globulin florisil assay for normal, sham-lesioned and septal-lesioned rats at 2 time periods, 12 h apart, corresponding to 2 h after the switch to light or dark under a 12:12 h light:dark regime. At a time 2 h after the switch to light, when normal and sham-lesioned animals evidenced low resting corticosterone responses, septal rats exhibited a fivefold increase in resting corticosterone levels compared to non-lesioned animals. At a time 2 h after the switch to dark, when basal corticosterone values are higher, normal, sham-lesioned and lesioned rats evidence no differences in their plasma corticosterone levels. Results suggest that the 24 h plasma-corticosterone rhythm is altered in rats following septal damage.
