Carboxyl groups in five proteinases from microorganisms, the bacterial mesentericopeptidase, subtilisin DY and subtilisin Carlsberg as well as the fungal proteinase K and thermitase were modified with glycinamide in the presence of water soluble carbodiimide. Computer graphic studies using crystallographic models of the investigated proteinases showed a reasonably good correlation between the chemical reactivity of the carboxyl groups on one side and the degree of their exposure to the solvent or participation in ionic interactions, on the other. Differences in the binding of a large protein substrate by proteinases from microorganisms belonging to two subgroups are discussed.

摘要:
The time course of growth and changes of phenolics formation were examined in Lavandula vera-MM suspension culture. After 9 days of cultivation, 11 g.dm−3of dry biomass was accumulated, which contained a sufficient amount of phenolic compounds (230 mg. dm−3). The HPLC analysis identified rosmarinic and caffeic acids as main components. To our knowledge this is the first report on cell culture from Lavandula vera producing rosmarinic acid.

Male and female pigs—a cross-breed of Big white and Landrasa, were treated with 3 mg pig somatotropin (pST) daily for 56 days during the period of fattening (from 30 kg up to 100 kg body weight). The animals were given a fodder mixture (15% raw protein) with 0.6% and 0.8% lysine content. In the case of 0.6% lysine, pST caused a 3.3% increase of daily growth, at the same time it cut down the forage consumption by 246 g/kg growth and decreased the thickness of bacon with 4.1 mm. When the fodder contained 0.8% lysine, pST decreased the thickness of the bacon only. pST did not change the concentrarion of free fatty acids in the blood serum. It has been concluded that it is possible to increase the production of high-quality pork by applying a suitable combination of dose and scheme of pST treatment of pigs.

The control of mixing conditions is one of the prerequisites for the successful fermentation performance of shear sensitive fungal cultures. The Fusicoccin (a biosynthetic plant growth regulator) producing fungus Fusicoccum amygdali Del. grows in the form of large mycelial agglomerates with long peripheral hyphae and partially as filamentous mycelia. The broth is characterized by high viscosity and the non-Newtonian, pseudoplastic flow behaviour, which causes the problems of mixing and mass transfer during fermentation. In this work, an attempt has been made to improve the fusicoccin (FC) production in FU-8 bioreactors using different mixing systems: TMS (The turbine mixing system), CMS1 (The counterflow mixing system with one drive), and CMS2 (The counterflow mixing system with two drives—the upper and lower ones). Higher agitator speed rates were possible in both CMS, since they caused less damage to the hyphae in comparison with TMS. Hydrodynamic conditions in fermenters were controlled using a Stirring Intensity Measuring Device, SIMD-F1. The distribution of the local flow kinetic energy e was analysed in the planes of the maximum mixing intensity. The energy e values in the local intensive planes were higher for TMS in comparison with those for CMS even at lower agitator speed rates of TMS, which could be indicative of an increased shear stress in TMS and obviously caused undesirable morphological changes of F. amygdali and a lowered FC yield. Principal fermentation parameters were analysed. The use of counterflow impellers improved the oxygen transfer efficiency, provided the formation of a more productive morphology, lowered the viscosity of the broth, increased the concentration of the biomass and FC. The improvement of FC production using the optimum mixing system was characterized by higher values of the oxygen uptake rate and lower values of the respiratory quotient during the fermentation. The control of the local mixing intensity using SIMD-F1 gives the possibility to understand the mixing behaviour in bioreactors and, for example, predict the influence of mixing on the mycelial fermentation performance.

The unstructured dynamics models of fed—batch fermentation process for microbiological L -lysine production have been discussed. The linear regression models show the correlation between the basic kinetic and physical—chemical variables of the process. The appropriate experimental data transformation has been done. The aim of this transformation is emphasised in order to reach the empirical probability data distribution to the normal probability distribution. It has been a base for adequacy of the proposed linear regression models. By means of linear and nonlinear regression the structure and parameters of the specific rates are selected. The unstructured dynamic models are set. The adequacy of the proposed models is proved through simulation research.

摘要:
A comparative analysis of conventional and neural model for forecasting the wearing out of bearing meeting of stirrer bioreactor is made in the paper. It shows that the preciseness of the neural model is comparable with this of the conventional one and can be used for forecasting. An axial sliding with contact elements “sphere-plane” is investigated. The resource and parameters of pressure of the bearing are determined and the construction is optimized on this basis, thus increasing the reliability of the bioreactor.
