ABSTRACTThe effects of interleukin 2 (IL2) on drug‐specific lymphocyte proliferative responses and their suppression by elicitation of sulbenicillin (SBPC) rash and by induction of SBPC tolerance were studied. GR‐LC and TO‐LC, the draining lymph node cells taken from animals with SBPC generalized rash (GR) and SBPC tolerance (TO), respectively, were incubated with IL2 rich medium (IL2RM) overnight, then washed, and tested for antigen‐specific response in a standard 3‐day proliferation assay. The GR‐LC and TO‐LC showed low responsiveness or unresponsiveness to specific drug before overnight incubation with IL2LM, but recovered a distinct responsiveness after overnight incubation with IL2LM. This result is encouraging for the establishment of the drug‐specific lymphocyte proliferation test for determination of the causative drug in patients with delayed type hypersensitivity

ABSTRACTIn this study, the author observed the adhesion ofStaphylococcus (St.) epidermidisto three kinds of cultured cells—human epidermal cells (JTC‐17), human synovial cells (McCoy) and mouse connective tissue cells (L). In the same manner,St. aureus, Micrococcus (M.) luteusandPseudomonus (Ps.) aeruginosaadhered to the cells, butEscherichia (E.) colidid not. Heat killingM. luteusdecreased its adhesiveness, and heat killingPs. aeruginosaeliminated its ability, but the adhesion ofStaphylococciwas not altered by the heat treatment. ABSTRACTThe specific activities of histamine‐N‐methyltransferase, the only enzyme to degrade histamine in human skin, were examined in 7 specimens from normal skin, 42 from erythema multiforme, erythema nodosum, erythema annulare or drug‐induced erythema and 12 from psoriasis or eczema. The enzyme activity was 3.45 ± 0.17 pmol/min/mg protein (mean ± S.E., n=7) in normal skin. The activities were significantly lower in 24 out of 30 freshly active lesional sites of erythematous dermatoses (lower than 2 S.D. from the mean in normal skin), while the activities in the central clearing areas were rather higher than those in the freshly active lesions. No significant changes were seen in the lesional skin of psoriasis or eczema. Decreased histamine‐degrading enzyme activity in the freshly active lesions of erythematous dermatoses may suggest the long lasting effect of the amine that was released in the lesions. In spite of the different clinical figures and multiplicity of etiologies, decreased histamine‐N‐methyltransferase activity was quite commonly observed, suggesting a similarity in the pathomechanisms of the

ABSTRACTBALB/c mice were immunized with human hair fibrous proteins. Then their spleen cells were fused with Sp2/0‐Agl4 mouse myeloma cells. Antibody‐producing hybridomas were selected by ELISA method and cloned by limiting dilution. A monoclonal antibody was chosen and designated as HKN‐2. Immunohistochemically, on frozen sections of normal human skin, HKN‐2 was found to recognize the suprabasal cells of epidermis and hair follicle, the cells in the keratogenous zone of hair shaft, the sebaceous cells and the ductal and myoepithelial cells of sweat glands. The basal cells of the epidermis and lower hair follicle, hair matrix cells and secretory cells of sweat glands did not react with HKN‐2. An immunoelectron microscopic method showed that the positive reaction to HKN‐2 was located on the tonofilaments in the cytoplasm. ABSTRACTThe skin elasticity of psoriasis plaques was measured in 10 patients suffering from psoriasis vulgaris with a recently introducedin vivomethod with high reproducibility. The method is based on repeated applications of suction to a 0.79cm2circular skin area, and measurement of tensile distensibility (TD), hysteresis (H) and resilient distension (RD) by the principle of electrical capacitance as calibrated with distance. Measurements of 23 plaques showed a decrease in TD and increases in H and RD when compared to regional control measurements. Experiments in 19 control regions with histamine skin‐pricks to evaluate the effects of cutaneous oedema on skin rheology showed that TD as well as H and RD became increased. The skin thickness of the plaques was increased by 68% as a mean, probably resulting from oedema of the outer dermis. The decreased TD, representing a component of stiffness, is probably related to a fixed zone of interdigitation between elongated and club‐shaped rete ridges and oedematous papillae while the increase in H and RD, representing a component of softness, is probably explained by the oedema of the outer dermis. ABSTRACTWe studied the conversion ofT. rubrumfrom hyphae to spherical cells using the agar‐implantation method. Thirteen cultures ofT. rubrumwere isolated from tinea superficialis and granuloma trichophyticum and they were implanted into the peritoneal cavities of ddY male mice. The cultures were divided into three groups according to their parasitic forms. The first group consisting of two cultures were isolated from tinea unguium and tinea pedis (hyperkeratotic type); they were destroyed in the peritoneal cavities of mice by the sixth week. The eight cultures in the second group were isolated from tinea cruris, tinea circinata and tinea pedis (vesicular type); they converted to spherical cells and survived for 16 weeks. The three cultures of the third group were isolated from granuloma trichophyticum; they survived and grew as hyphal forms in the peritoneal cavities of mice for over 26 weeks without spherical cell conversion. ABSTRACTFour children with porphyria cutanea tarda symptomatica were reported. Two of them had a previous history of exposure to pesticides. All four received a daily treatment of 250 mg of chloroquine for seven days. Liver function test results were altered immediately after conclusion of the therapy and porphyrin levels were very high.A light and electron microscopic examination of eight control liver biopsies revealed fatty changes before treatment, extensive liver damage during treatment, and regeneration of the liver parenchyma after the treatment.In view of the transient, but severe, liver damage, this therapeutic modality is not recommended for the treatment of children with porphyria cutanea tarda.

摘要:
ABSTRACTA case of giant irritated seborrheic keratosis is reported. The tumor showed an unusually gross appearance for seborrheic keratosis; hence, before surgery, it was suspected of being a verrucous carcinoma. However, examination of the whole histologic constitution of the excised tumor warranted the diagnosis of irritated seborrheic keratosis.

ABSTRACTTwo cases of basal cell epithelioma with giant tumor cells are reported. Differentiation into squamoid cells and phagocytosis of melanosomes and remnants of the degenerated tumor cells may be factors in initiating giant cell formation. The localization of these giant tumor cells into specific nests suggests the presence of unknown factors contributing to their formation.

ABSTRACTAcquired periungual fibrokeratoma is essentially a topographical variant of acquired fibrokeratoma and shows a full variety of clinical features according to the specific location of each neoplasm in its relation to the nail. The protean clinical manifestations of this condition are difficult to fit together into an entity. A new classification into two groups with five subtypes, based on the anatomical and histological architecture of the nail is proposed. According to this classification, the obscure relationships among previously described cases are discussed.
