摘要:
AbstractA novel protein, haematoxylin stain‐positive, non‐nucleoprotein has been extracted from the epidermis of 3‐day‐old rats. This protein was purified through preparative isoelectric focusing, followed by sodium dodecylsulfate polyacrylamide gel electrophoresis. The purified protein had a molecular weight of 14,500 daltons and an isoelectric point of 4.7. The amino acid composition included 15% glycine, 11% serine, 13% glutamic acid and 10% aspartic acid, but only 1.9% histidine.Using the immunofluorescent and immunoelectron microscopic techniques, observation showed that the antibody formed as a result of purified antigen was located on keratohyalin granules and in the cell membrane of the stratum corneum of the skin.This haematoxylin stain‐positive protein could be synthesized in the stratum granulosum as one component of keratohyalin granules, coat the stratum corneum cell membrane and perhaps, we think, may act as one important component of the epiderma

摘要:
AbstractTo induce chronic skin changes simulating human porphyria, mice which had been injected intraperitoneally with hematoporphyrin dihydrochloride were exposed to metal halide lamps. Clinical and histopathological findings in the mice were observed. Clinically, erythema, edema, erosion, necrosis and crust formation were initially seen in all of the mice treated. Hypertrophy of the skin, scarring, and deformities of the ear lobes followed gradually. Histopathological findings in these skin changes were mainly seen in the dermis and included infiltration of lymphocytes, proliferation of mast cells, dilatation and hyperemia of blood vessels in the upper dermis, and swelling and proliferation of connective tissues. PAS positive materials were deposited in the dermo‐epidermal junction and around the small blood vessels in the upper dermis. Using the direct immunofluorescent technique, the deposition of immunoglobulin G (IgG) was confirmed in the same areas, and some deposition of IgM, IgA and C3 was also observed. AbstractIdentification and enumeration of both Ia + dendritic epidermal cells (Ia + DECs) and Thy‐1 + dendritic epidermal cells (Thy‐1 + DECs) from various parts of the body and non‐irradiated and ultraviolet B (UVB) irradiated back skin were examined using epidermal sheets of C3H/He inbred mice of different age groups and indirect immunofluorescent technique. The following results were obtained: [1] There was a significant decline in both Ia + DEC and Thy‐1 + DEC density in the mice in the oldest group (48–50 weeks); [2]The densities of Ia + DECs were significantly higher than those of Thy‐1 + DECs in comparisons of various parts of the body; [3] At 24 h after 60–120 mJ/cm2UVB irradiation, the Ia + DECs and Thy‐1 + DECs decreased significantly in a dose‐dependent fashion. The Ia + DECs decreased drastically (p<0.01) while the Thy‐1 + DECs decreased mildly (p<0.05). [4] The degree or resistance to UVB differed between Ia + DECs and Thy‐1 + DECs in older mice (40–48 weeks). These findings may imply that the decline of the Ia + DECs and Thy‐1 + DECs reflects alterations in immune response during aging; As do Ia + DECs, Thy‐1 + DECs might also play a role in UVB induced specific unresponsiveness in contact hypersensitivity; each type of Ia + DECs and Thy‐1 + DECs follows a distinct biological kinetic

AbstractThe skin lesions of MRL/l mice were examined by electron microscopy and the following changes were disclosed in the dermis. Fibroblasts and mast cells were increased in number and activated. A major part of the infiltrating mononuclear cells was morphologically T‐lymphocytes. The capillaries, degenerated to varying degrees with indistinct basal laminae, were remarkably increased, and exudative amorphous material was deposited around them. Neither electron‐optically dense deposits around the vessels suggesting immune deposits nor tubulo‐reticular structures in the endothelial cells were detected.The basic lesions occurring in the dermis of MRL/l mice seem to be primary injuries of the capillary endothelial cells and consequential exudative changes of perivascular areas, similar to those of erythematous lesions in human systemic lupus erythema

AbstractMouse epidermal transglutaminase was partially purified from the epidermis of newborn BALB/c mice. The epidermal homogenate was subjected to CM‐cellulose chromatography and then to gel filtration on Sephadex G‐150. Two separate peaks with enzyme activity were reproducibly observed in CM‐cellulose chromatography. Gel filtration on Sephadex G‐150 of the peak fractions in CM‐cellulose chromatography gave a single enzyme activity peak (molecular weight about 55,000). Native polyacrylamide gel electrophoresis at pH 4.5 showed two major bands with enzyme activity. The final enzyme preparation (Sephadex G‐150 fraction) had a pH optimum of 10.0 in glycine‐NaOH buffer. Calcium ion and SH‐protecting agent (dithiothreitol) were essential for enzyme activity. The enzyme was not inactivated by heating at 56°C for 45 min in the presence of calcium. Trypsin treatment of the enzyme enhanced the activity about threefold; this enhancement was blocked by trypsin inhibitor. The enzyme bound to lysine‐Sepharose 4B in the presence of casein in Tris‐acetate buffer, pH 6.0, at 37°C; 40% of the enzyme activity was eluted from this enzyme‐lysine‐Sephar

摘要:
AbstractConcentric multilamination of the basal lamina around the subepidermal capillaries was observed after reaction to dinitrochlorobenzene. It consisted of luminal obstruction, necrosis of endothelial cells and pericytes, regeneration of these cells, and new deposits of basal lamina. Beneath the two basal laminae, delicate collagenous fibrils were present. These findings help explain the similar but more pronounced lamination of the vascular basal lamina in patients with diabetes, protoporphyria,etc.and the endothelial production of fibrillar collagen under certain condition.
