AbstractThe cell electrophoretic mobility of polymorphonuclear leukocyte responses to scale extracts from psoriasis vulgaris and other dermatoses (callus and pompholyx) was determined by analytical free‐flow cell electrophoresis technique. Incubation with scale extracts from psoriasis significantly reduced the electrophoretic mobility of polymorphonuclear leukocytes compared with those from callus and pompholyx; ultrafiltrated scale extracts separated 10,000–30,000 d showed the highest reduction. Pretreatment of polymorphonuclear leukocytes with etretinatein vitroblocked reduction of the cell electrophoretic mobility. It is concluded that psoriatic scale extracts contain factor(s) which reduce the charge density of polymorphonuclear leukocytes and that this reduction is inhibited by etretinate pretreatm

AbstractThe retinoic acid binding protein (CRABP) characterized in human epidermal cytosol exhibits a three‐fold increased binding capacity in lesional psoriatic epidermis as compared to normal or uninvolved skin. Treatment of epidermal homogenate from normal subjects by phorbol ester + ATP decreases the specific binding capacity of CRABP without affecting its dissociation constant (Kd=10 nM). The same effect was not observed in involved and uninvolved psoriatic epidermis. These results could be related to the previously reported decreased protein kinase C activity in psoriatic skin. AbstractIntercellular junctions of keratinocytes are essential for coordination of the epithelial tissue function. Neoplastic transformation is often accompanied by changes in the glycolipid and glycoprotein composition of the cell membranes, although no evident structural modifications can be observed. We have investigated one aspect of the keratinocyte cell‐cell contact in squamous cell carcinoma using KM48 monoclonal antibody as a differentiation‐related marker for desmosomes. Quantitative analysis of the immunogold labelling revealed that the tumor cells lose the KM48 antigen during their neoplastic transformation. The process could also be observed, although to a lesser degree, in clinically and ultrastructurally normal marginal epidermis. The desmosome frequency in the tumor tissue also decreased, but not as radically as did the KM48 expression. AbstractIn vitroandin vivoeffects of various vitamin A analogues on the transglutaminase activity of mouse epidermal cells were investigated.In vitroaddition of all‐trans‐retionic acid (retinoic acid), retinyl acetate, or Ro 10‐9359 (etretinate) to a culture medium induced transglutaminase activity in a transformed mouse epidermal cell line (PAM 212 cells). Induction by a new synthetic vitamin‐A‐like compound (E‐5166, polyprenoic acid derivative) was significant but less marked than that by other vitamin A analogues. Simultaneous addition of retinoic acid and the phorbol ester 12‐O‐tetradecanoylphorbol‐13‐acetate (TPA), which also induces transglutaminase, did not cause any increase in transglutaminase activity. These retinoic acid derivatives inhibited the proliferation of PAM 212 cells at concentrations of 10–5–10–6M in the culture medium.In vivoanalysis consisted of oral administration of retinoic acid and Ro 10‐9359 to adult mice. These mice were found to have increased liver transglutaminase activity in the soluble fraction but not in the particulate fraction. Orally administered retinoic acid did not induce transglutaminase activity in the epidermis in the whole skin. These results suggest that retinoids modify transglutaminase activity and epidermal proliferation and differentiationin vitro, but the induction of the enzyme seems not to be related to the clinical effects of orally administered retinoi

AbstractA 45‐year‐old woman had a triple cancer, multiple Bowen's disease, breast cancer, and carcinomain situof the cervical portion of her uterus. Development of all three carcinomas was discovered within a period of three years. Southern blotting analysis of the epidermal growth factor receptor (EGFRc) gene of DNA extracted from the individual lesions of multiple Bowen's disease revealed no amplification or variant other than one extra band observed in one lesion. DNA extracted from other patients with solitary Bowen's disease showed essentially the same result. In contrast to squamous cell carcinoma DNA which sometimes has amplification of this gene, our present case suggests that there is only a small possibility that the EGFRc gene plays a role in the carcinogenesis of Bowen's dise

AbstractTwo types of human malignant melanoma were treated either with peplomycin alone, hyperthermia alone, or a combination of peplomycin and hyperthermia. The antitumor effect of combination therapy was evaluated by tumor growth curves, maximal suppression rate, and histopathology. A marked antitumor effect of combination therapy with peplomycin (5 mg/kg) and hyperthermia (43°C, 30 min) was obtained for both types of melanoma. Histopathologic findings revealed advanced central necrosis and pyknotic nuclei in the tumor cells. Although peplomycin alone (20 mg/kg, 6 times) had no antitumor effect on C24 melanoma, the peplomycin (5 mg/kg, 6 times) + heat group showed complete inhibition of the tumor growth. AbstractThe clinical manifestations of types I (lepra) and II (ENL) reactions are now well‐appreciated. The correlation of clinical expression to bacterial index, lepromin response and histological features has hardly been attempted. In a study of 35 reaction patients, 18 type I (lepra) and 17 type II (ENL), the preceding parameters were studied in detail. Their incidence among leprosy patients was 20%. Males in the age group 20–30 years were frequently affected, the ratio of males to females being 6:1. The mean age at onset in type II (ENL) was younger than in type I (lepra) reactions. However, the duration of the reactions was variable. They were frequently precipitated by antileprosy drugs. The clinical features were cardinal. The bacteriological features reflected wide variations, but were considered imperative. Early Fernandez and Late Mitsuda reactions were useful adjuvants for monitoring progress.
