AbstractTyrosine phosphorylation is widely recognized as playing important roles in cell differentiation, proliferation, and carcinogenesis. We have used the polymerase chain reaction (PCR) method to identify protein tyrosine kinases that are expressed in the skin. Mixed oligonucleotide probes were used to amplify and screen a neonatal murine skin cDNA pool for clones encoding amino acid contiguities whose conservation is characteristic of the protein tyrosine kinase family. When the PCR products were sequenced, 13 distinct clones were found, of which one is novel to date and has provisionally been namedtks(for tyrosine kinase identified from skin). Sequence homology comparison showed that thetksgene is homologous to the src and fes/fps families. Northern blotting using PCR products oftksas a probe revealed that the mRNA oftksis detected ubiquitously and weakly in other tissues such as brain, lung, liver, thymus and kidney. This fact suggests that thetksgene is expressed in widely distributed cell types.

AbstractWound healing substance (WHS) from culturedClostridium perfringenshas been reported to be effective in the treatment of wounds. The effects of WHS, now named SNK‐863, on proliferation and differentiation of human keratinocytes were examined.The characteristics of WHS are as follows: 1) WHS stimulates human keratinocyte growth and DNA synthesis; 2) WHS and EGF show some additive effects on human keratinocyte growth; 3) WHS does not interfere with the binding of EGF to its receptor; 4) WHS does not counteract the growth inhibitory effects of TGF‐β or vitamin D3on human keratinocytes; 5) WHS has no significant effect on human keratinocyte differentiation. AbstractIncreases in the number and activity of peripheral polymorphonuclear neutrophils (PMNs) is often found in Behçet's disease (BD), indicating that PMN may play an important role in the pathogenesis of this disorder. It has recently been reported that G‐CSF and GM‐CSF, a family of hematopoietic growth factors, enhance PMN activity. To explore the role of these two CSFs in BD, we first examined the chemotactic response of PMNs to these CSFs by performing a polarization assay. PMN response to G‐CSF in BD patients was lower than that in controls, while PMN response to GM‐CSF was similar in patients and controls. However, PMNs from BD patients showed an enhanced chemotactic response to N‐formyl‐L‐methionyl‐leucyl‐phenylalanine. Thus, it is speculated that the PMNs of the patients might have already been activatedin vivoby G‐CSF and thus could not respond further to this agentin vitro. We examined G‐CSF and GM‐CSF mRNA expressions in peripheral mononuclear cells stimulated with LPS, PMA, and Con A by Northern hybridization. G‐CSF mRNA expression levels in BD patients were higher than in the controls, while GM‐CSF mRNA expression levels were lower than in the controls. We also examined the serum levels of the two CSFs by ELISA and EIA. However, all levels of the two CSFs in both patients and controls were not detectable, except in the case of one BD patient in the active stage of the disease, who showed high levels of G‐CSF, but not of GM‐CSF. AbstractFormalin‐fixed and paraffin‐embedded tissue specimens of six cases of extraocular sebaceous carcinoma were studied immunohistochemically with eight anti‐keratin monoclonal antibodies, 34βB4, 35βH11, Ks13.1, Ks19.1, PKK1, LP34, KL1 and AE1. The staining patterns of sebaceous carcinoma were compared with those of normal sebaceous glands and other skin cancers which should be distinguished from sebaceous carcinoma histopathologically. The other skin cancers compared were eccrine porocarcinoma, malignant clear cell hidradenoma, extramammary Paget's disease with underlying adenocarcinoma, malignant trichilemmoma, and squamous cell carcinoma. Most cases of sebaceous carcinoma were stained with 35βH11, Ks19.1, LP34, KL1 and AE1, while normal sebaceous glands were positive only with 35βH11, LP34, KL1 and AE1. By immunostaining, sebaceous carcinoma was distinguishable from extramammary Paget's disease with underlying adenocarcinoma, squamous cell carcinoma, malignant trichilemmoma, and eccrine porocarcinoma, but was not clearly distinguishable from malignant clear cell hidradenoma. These findings demonstrate that sebaceous carcinoma shows positive reactions with antibodies to simple epithelial keratin, probably as a result of neoplastic transformation, and that immunohistochemical examination using anti‐keratin monoclonal antibodies is useful in distinguishing sebaceous carcinoma from several other s

AbstractThe epidermal skin lesions of herpes zoster were studied by scanning (SEM) and transmission electron microscopy (TEM). When erythematous lesions were observed by TEM, many of the infected keratinocytes showed evidence of cell degeneration, being characterized by swollen nuclei, disappearance of desmosomes, and widening of intercellular spaces. Macrophages and/or lymphocytes migrated through the intercellular spaces between degenerated keratinocytes. In the vesicular lesions, SEM and TEM showed some infiltrating neutrophils, directly adhering to the virus‐infected keratinocytes, with swollen nuclei and irregularly clumped chromatin. In some specimens, balloon‐degenerated keratinocytes were observed in the cavity. In the pustular stage, ruptured keratinocytes and numerous neutrophils were observed in the reticular‐degenerated epidermal tissue. 摘要:
AbstractWe present three patients with severe primary hyperhidrosis, refractory to conservative medical treatment, who were successfully managed with sympathetic ganglion blockade with ethanol. We also summarize 10 patients with hyperhidrosis who underwent sympathetic ganglion blockade in the past 2 years. This closed percutaneous method offers the patients considerably less discomfort and less stress with minimal morbidity and has a efficacy similar to that of surgical sympathectomy, which has previously been the only effective and permanent therapy for severe primary hyperhidrosis. It is concluded that chemical sympathectomy is an effective and useful method for treating severe hyperhidrosis which has advantages over surgical sympathectomy.

AbstractPaget's bone disease developed in a patient with vitiligo. Scrupulous physical examination excluded further systemic or cutaneous involvement. The immunological workup revealed a reversed CD4/CD8ratio due to a very low CD4cell percentage and almost negligible responses to PHA as well as Con A, T cell mitogens. The pathogenic significance of these results, which point to phenotypic and functional T cell defects, is discussed.
