摘要:
ABSTRACTFor the purpose of demonstrating ultrastructurally the distribution of anionic sites in the rat skin, we applied two polyethyleneimines of different molecular weights and ruthenium red by the perfusion or immersion method. Using each cationic probe, sites were observed as electron dense particles along both the epidermal and interstitial sides of the lamina densa in the dermo‐epidermal junction and the interstitial side of the periendothelial lamina densa in the dermal capillary. The ordered distribution of the anionic sites may function to transport fluid and macromolecular substances through these particular compartment

ABSTRACTA sliced tumor of dermatofibrosarcoma protuberans was labelled with3H‐glucosamine in Ham's F12 medium with 10% fetal calf serum at 37°C in 5% CO2with 95% air for 24 hours. Electrophoresis of extracted glycosaminoglycan was undertaken on cellulose acetate strips. The radioactive content of the separated glycosaminoglycan in the tumor was counted and compared with the glycosaminoglycan of normal skin. A larger amount of hyaluronic acid and chondroitin sulfate was synthesized in dermotofibrosarcoma protuberans than in normal skin.The qualitative and quantitative changes in glycosaminoglycan in the stroma suggest that it is unlikely to metastasize to the regional lymph nodes and internal orga

摘要:
ABSTRACTA model of contact sensitivity (CS) based upon ear swelling has been developed in rats, which are regarded as low responders to common contact sensitizers. For sensitization, a 1% solution of tolylene diisocyanate (TDI) in ethyl acetate was applied to male F344 rats in three different ways; the solution was applied to the shaved backs (1st group), to the unshaved tails (2nd group), and to the tails and then, seven days later, to the backs (3rd group). All the rats were ear‐challenged seven days after the above treatments. The most remarkable ear swelling was observed in the 3rd group, the rats sensitized by the double application method. Delayed onset of the ear response together with histology of the swollen ear showed that the response was characteristic of delayed‐type hypersensitivity. This was confirmed by the successful achievement of CS transfer with lymphocytes from sensitized rats. When picryl chloride was used as a sensitizer instead of TDI, no comparable ear swelling was induced irrespective of the sensitization method. It was also found that topical application of a corticosteroid cream dramatically reduced the ear swelling induced by TDI. These results suggest not only that an experimental model for CS can be developed in rats using TDI, but also that CS induced by TDI can be used as a model for evaluation of anti‐CS

ABSTRACTFibroblasts from guinea pig skin were incubated with [3H] glucosamine. The cells secreted [3H] galactosamine‐labeled compounds predominantly as dermatan sulfate, which accounted for 65–75% of the total glycosaminoglycans. The majority production of dermatan sulfate was observed regardless of the composition of the culture medium, cell growing phase or age of animal. The labeled glycosaminoglycans were identified by enzymatic (chondroitinase ABC, chondroitinase AC and hyaluronidase) or nitrous acid treatment, followed by fluorography of two‐dimensional electrophoresis on cellulose acetate membranes. The molecular weight of the newly synthesized dermatan sulfate from medium of a confluent stage was calculated as 15,000 to 17,500 from molecular sieve chromatography. The radioactive dermatan sulfate was treated with chondroitinase ABC, yielding 68.4%ΔDi‐4S, 16.2%ΔDi‐6S, 13.3%ΔDi‐OS and 2% di‐sulfated hexuronosyl‐N‐acetylhexosamine units. Chondroitin sulfate was detected on a cellulose acetate membrane as a minor component in the culture medium from the cell growing stage. It was not detected in medium from a confluent stage. 摘要:
ABSTRACTA survey of nylon cloths friction dermatosis (NCFD) was taken by gathering information by a questionnaire sent to university hospitals in Japan. The replies were obtained from 44 hospitals (55% recovery) reporting 158 patients. Pigmentation was noted mainly on the skin over the trunk bones and extremities. The pigmentation was distributed over more than two regions in most of the patients. Along with the clinical and histological features, a history of repeated friction, usually with nylon towels or scrub brushes, was required for diagnosis. A few months after discontinuing the use of nylon towels or scrub brushes, NCFD disappeared or improved in 15% of the patients.

ABSTRACTThe enzyme activity and mRNA level of M‐type lactate dehydrogenase (LDH) were studied in dermatofibrosarcoma protuberans (DFSP), a low grade sarcoma of the skin with a long clinical course and pronounced tendency toward progression and local recurrence. The isoenzyme pattern of LDH in DFSP was quite similar to that of other malignant skin tumors. The total LDH activity, ratio of M‐type activity, and mRNA level of M‐type LDH in DFSP were higher than those in control skin. It is suggested that this higher enzyme activity of M‐type LDH in DFSP was mainly due to the higher activity of mRNA coding for M

ABSTRACTHistogenetic identification of poorly differentiated spindle cell tumours can be very difficult if only routine histology is used. In the case of a 64‐year‐old man suffering from a recurrent and spreading malignant tumour on his neck, the precise diagnosis of malignant schwannoma was not established until a skin node was examined by electron microscopy. The neurogenic nature of the neoplasm was additionally confirmed by demonstration of S 100 protein within most of the tumour cells. Ultrastructurally, the tumour was composed predominantly of atypical Schwann cells and, in varying number, of perineurial cells, endoneurial cells and some myofibroblasts exhibiting features enabling their differentiation.
