AbstractThe emergence of anionic sites during basement membrane zone formation was studied using migrating epidermis in organ culture as a model system. Ultrastructural investigations using a strongly cationized probe revealed that the anionic sites were found to be formed synchronously with the newly built basal lamina after 7 days in culture.Additionally, we examined the effect of chemically generated hydroxyl radicals on basal lamina‐associated anionic sites. Cytochemical studies showed that hydroxyl radicals produced a loss of cationic tracer positive anionic particles; this effect was inhibited by a specific scavenger, thiourea. These data might suggest that anionic sites were degraded by leukocyte‐derived hydroxyl radicals during acute inflammation near the dermal epidermal junction, resulting in disturbance of the charge‐selective permeability ba

AbstractInteractions between the histidine‐rich protein of keratohyalin granules and keratin fibers were investigated. Keratin fibers of the stratum corneum from sole skins of both normal persons (NP) and those with hereditary palmoplantar keratoderma (HPPK) were examined. HPPK samples showed trichilemma‐oid keratinization except at the sweat duct. Epidermis around the sweat duct showed stratum granulosum formation. The keratin pattern was lost in the non‐sweat duct region; any changes in the size and distribution‐pattern of keratin fibers were, however, observed by electron microscopy. In sodium dodecylsufate polyacrylamide gel electrophoresis (SDS‐PAGE), keratin fibers in the HPPK stratum corneum, from both non‐sweat duct and sweat duct regions, and in the stratum corneum of the sole lesion of the HPPK's mother, lacked 57kd subunits, but the electrophoretic profiles of other keratin subunits were identical to those from NP samples. AbstractCapillaries of the rat skin were sectioned in planes parallel, perpendicular, and oblique to the vascular long axis for the purpose of studying the perivascular fibrous stroma. Fine structural observation revealed that the vessels had organized stromal coats. Delicate collagen fibrils were regularly arranged in alternate, largely orthogonal layers pursuing a spiral or longitudinal course at the innermost layer. However, the fibrillar density and layering differed between the continuous and fenestrated segments of the capillary course. Along segments with a narrow lumen and a thick endothelium, the fibers were dense and both the longitudinal and circular layers exhibited almost complete turns around the vascular perimeter. The fenestrated segments, in contrast, showed a loosely arranged fibrillar coat and sometimes lacked the circular layer.

AbstractThe localization of ornithine decarboxylase (ODC) in human skin and cultured keratinocytes was studied with an immuno‐histochemical method. ODC was found in the epidermis, hair follicles, sweat glands and errector muscles. Irritation induced by stripping or UV‐B irradiation did not change the staining pattern in the epidermis. In psoriasis, the staining was most marked at the tip of the epidermal rete ridges. In basal cell carcinoma, there was a homogeneous labelling of the tumor cells and, in squamous cell carcinoma, the labelling was strong but less homogeneous. Melanoma and dermal naevus also positively stained for ODC. Cultured human keratinocytes also showed ODC positive immunofluorescence. AbstractUsing an immunofluorescent technique, the localization of angiotensin converting enzyme (ACE) was investigated in granuloma lesions from the skin of three sarcoidosis cases. Specific fluorescence was observed in epithelioid cells in the sarcoidosis granulomas examined. However, it was not found in any other granulomatous diseases observed as controls, which included granuloma annulare, foreign body granuloma and lupus miliaris disseminatus faciei. These results indicate that ACE is specifically localized in the epithelioid cells in cutaneous granuloma lesions of sarcoidosis; in one case, it was also observed in Langhans giant cells. Therefore, it is suggested that examination for ACE in cutaneous lesions using an immunofluorescent technique is very useful for the diagnosis of sarcoidosis in the skin. We suggest that epithelioid cells of granulomas in sarcoidosis cutaneous lesions may play an important role in the increase of serum ACE activity.

AbstractSeventeen rabbits were inoculated with 0.1 ml of herpes simplex virus (HSV), type 1, Miyama strain (105,75TCID50/0.1 ml),viathe auricular artery; twelve of them developed zosteriform eruptions (ZE) on the skin. Seven rabbits treated with control medium showed no eruptions. This phenomenon was significant (P<0.01).ZE was observed about 7 days (mean time: 7.4 days) after inoculation. Although HSV antigen was found in dorsal root ganglia, spleen and liver by an immunofluorescence method, it was not detectable in the skin lesion. IgG, M and C3were also undetectable in the skin lesion. On the other hand, complement requiring neutralizing antibody (CRN‐Ab) against HSV was detected on the 6th day after inoculation. AbstractWe investigated the effect of 8‐methoxypsoralen (8‐MOP) plus long wave ultraviolet irradiation (PUVA) on the pig skin epidermal cyclic AMP (cAMP) system. Following PUVA treatmentin vitro, pig skin squares were incubated in RPMI 1640 medium, and the cAMP accumulation by various adenylate cyclase stimulators was determined. A significant increase of the beta‐adrenergic adenylate cyclase response was observed as early as 6‐h following PUVA treatment; the maximal effect was reached at 24‐h and lasted at least for 72‐h. This augmentation effect of PUVA treatment was potentiated by increasing the 8‐MOP concentration (0.5–160 μg/ml) and UVA irradiation dose (0.05–1.4 J/cm2). At higher irradiation doses (2.1–2.8 J/cm2), the beta‐adrenergic augmentation effect was less marked. There were no significant differences in the adenosine‐and histamine‐adenylate cyclase response up to 1.4 J/cm2irradiation; however, the latter was decreased at a higher irradiation dose (2.8 J/cm2). Although UVA irradiation alone had no effect on the beta‐adrenergic response, 8‐MOP treatment alone increased this receptor response at higher concentrations; this effect was much weaker than that induced by PUVA treatment. AbstractBlood histamine levels during topical corticosteroid therapy have not been reported, although decreased blood histamine levels are known to occur after the systemic administration of corticosteroid. Blood histamine levels and serum 11‐hydroxycorticosteroids (11‐OHCS) after systemic corticosteroid were compared with those after topical corticosteroid. Both blood histamine and serum 11‐OHCS levels decreased in a parallel and dose‐dependent manner during systemic administration of 0.5‐1.0 mg/day of betamethasone. Blood histamine levels increased and serum 11‐OHCS levels decreased during systemic administration of 0.25 mg/day to the betamethasone and topical corticosteroid group. Changes in blood histamine levels in the topical corticosteroid group appeared between 0.25 and 0.5 mg/day of systemically administered betamethasone.Fifteen cases of dermatological inpatients suffering from wide‐spread eczema or exfoliative dermatitis were treated with the topical corticosteroids betamethasone 17‐valerate, budesonide and diflorasone diacetate. In each case, blood histamine levels were examined and an equipotent dose of oral administeration of betamethasone was calculated. Thirteen cases in whom blood histamine levels were measured fell into the dosage range from 0.1 to 0.5 mg/day.In each case, serum 11‐OHCS levels were also examined and an equipotent dose of oral administration of betamethasone was calculated by the same method. Fourteen cases in whom serum 11‐OHCS levels were measured compared to a dose of less than 1.0 mg/day of betamethasone.
