AbstractA newly‐synthesized, monofunctional psoralen derivative, 7‐methyl pyrido (3,4‐C) psoralen (MPP) was compared with 8‐methoxypsoralen (8MOP) with respect to their therapeutic efficacy in photochemotherapy of psoriasis. Psoriatic lesions of six patients were treated with topical application of MPP plus UVA (MPP PUVA) or with 8MOP plus UVA (8MOP PUVA). The UVA doses used in each treatment were 7.5 or 10 J/cm2with MPP and from 1.2 to 3.6 J/cm2with 8MOP. In every patient, marked improvement was observed after 2 to 6 treatments with MPP PUVA or 8MOP PUVA. Three patients showed clearance of each psoriatic lesion treated 9 to 17 times with MPP or 8MOP PUVA. Althought MPP required much higher UVA doses than 8MOP, MPP PUVA was as effective as 8MOP PUVA in treating psoriasis. When irradiating with identical doses of UVA, MPP PUVA appeared to be less active than 8MOP PUVA. None of the patients developed any severe dermatitis reactions during 20 exposures to MPP PUVA, indicating that the probability of inducing allergic contact and photocontact dermatitis may be extremely low. Erythemogenic and pigmentogenic activities of MPP and 8MOP were also compared. The data demonstrated that 8MOP is more than 8 times as effective as MPP for both activities. With the UV doses used in this study, however, every patient produced marked pigmentation after MPP PUVA therapy. Finally, the UVA dose‐dependency of MPP PUVA was studied with an additional patient. AbstractA polyclonal antibody elicited by a newly purified haematoxylin stainable protein (HSP) from newborn rat epidermis by Superose 12 column chromatography and Mono Q anion‐exchange column chromatography showed localization in the cell membrane region of the stratum corneum cells of the flank skin and the foot pad skin of newborn rats, but not of adult rats in indirect immunofluorescent studies. In addition, although the stratum corneum of neither the fetal flank nor the foot pad skin (20th day of gestation) reacted with the antibody, both of them became positive shortly after birth (5 hours). However, the positive areas in the stratum corneum of the foot pad skin gradually moved to the upper part. Only the upper one third was positive at the 15th day after birth and the entire stratum corneum finally became negative in the adult. By immunoblotting technique, antigenic materials were detected in the fetal flank and foot pad skins, and in the adult foot pad skin, but not in the flank skin of adults. In immunofluorescent studies, neither the cornified nor non‐cornified epithelia of the oral membrane, tongue, esophagus, trachea or bladder of the newborn rats (3‐days old) reacted with this antibody. In studies of the reactivity of the stratum corneum of the other species, the outer one third of the stratum corneum of the flank skin of newborn mice positively reacted. However, the fluorescence was not membrane‐located, but rather diffuse. Newborn guinea pig epidermis showed no positive reaction. AbstractThe distribution of transferrin receptor in normal human skin and its expression in psoriatic skin and various skin tumors have been investigated. Immuno‐peroxidase staining was performed on biopsy specimens using monoclonal OKT 9 antibody, which reacts with transferrin receptor.Normal human skin showed positive staining with OKT 9 in eccrine glands and outer root sheaths of the hair. Sebaceous glands were also strongly positive. The basal layer stained very weakly.Psoriatic skin expressed OKT 9 strongly in the epidermis, especially in the area of the rete ridge.In squamous cell carcinoma, Bowen's disease, and malignant melanoma, a widespread and strong labelling reaction was found. Basal cell epithelioma and genital Paget's disease were partially and moderately positive in their staining pattern. No such positive staining pattern could be found in either nevus cell nevi or seborrhoic keratosis.These findings indicate that, in normal skin, transferrin receptor exists in eccrine glands, sebaceous glands, and outer root sheaths of the hair in greater amounts. High amounts of expression of this receptor in psoriatic epidermis and malignant tumors suggests that immunohistochemical demonstration of transferrin receptor parallels the proliferating activity of the tissue or tumor of the skin and may provide an useful aid for detecting such condition

Abstract1)The prolidase (Pd) and prolinase (Pn) activities of cultured skin fibroblasts derived from two prolidase‐deficient sisters, the elder with typical clinical manifestations [symptom (+)] and the younger with only slight clinical manifestations [symptom (–)]were examined biochemically. Pd activity against several substrates other than Gly‐Pro were present to some degree in both sisters. There were no detectable differences in Pd activity between the symptom (+) patient and the symptom (–) sister. Pn activity seemed to be increased in both. The lower Pn activity found against Pro‐Gly as compared with those against other substrates indicates that Pro‐Gly, which has been used for Pn assays in most previous reports, may not be the best substrate for this test. Pd derived from control fibroblasts was activated by Mn2+against all substrates tested in this experiment. Cu2+, Hg2+, Cd2+and Zn2+remarkably inhibited enzyme activity, Co2+slightly inhibited it, and neither Mg2+nor Fe2+had any remarkable effect. The Pd derived from the prolidase‐deficient patients was also activated by Mn2+. This Pd seemed to be more inhibited by Co2+than was the control. AbstractWe cultured non‐epithelial cells derived from dermatofibrosarcoma protuberans (DFSP). The cells proliferated well, to the same degree as fibroblasts. AbstractA case of genital Paget's disease examined by a transmission‐, scanning‐, and immuno‐electron microscopy was reported. Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) observations indicated that Paget cells had the characteristic features of glandular cells. In immunoelectron microscopy, carcinoembryonic antigen was found on the entire plasma membrane of Paget cells. AbstractTwo strains ofT. rubrumand one strain ofT. mentagrophyteswere inoculated into human skin grafted onto BALB/c nude mice by the needle puncture method. Infection was established in 1 of the 10 animals inoculated with fluffy colony typeT. rubrum, 2 of the 10 animals inoculated with powdery colony typeT. rubrum, and 7 of the 10 animals inoculated with granular colony typeT. mentagrophytes, suggesting that the skin grafts are infectible by anthropophilic and zoophilic strains of dermatophytes.T. rubruminfection continued for a maximum of 9 weeks andT. mentagrophytesinfection for more than 11 weeks. In the animals inoculated withT. mentagrophytes, fungal elements were localized in the stratum corneum of the human skin grafts. In the acute stage, microabscesses consisting of neutrophils were observed under the stratum corneum in contact with fungal elements; in the chronic stage, epidermal thickening and infiltration, mainly consisting of histiocytes and a smaller number of lymphocytes, was noted in the upper and middle dermis. Ultrastructural findings from the parasites were similar to those of dermatophytosis in man. This experimental system should be useful as a model of chronic dermatophyte infection in the human skin.

AbstractNatural killer (NK) cell activity and NK‐related surface markers in peripheral blood mononuclear cells (PBMC) in thirty‐three patients with adult type atopic dermatitis (AD) were investigated. NK cell activity, detectable by cytolysis of erythroleukemic cell line K562 cells, was reduced in patient PBMC as compared with healthy controls. Imbalances in the lymphocyte populations reactable with the monoclonal antibodies, anti‐HNK‐1, and anti‐CD16 were found in the patient PBMC. Some positive correlations between these cell populations and the NK cell activities were recognized. In most cases, high levels of serum IgE were inversely correlated with NK cell activity. Depletion of the macrophages from patient PBMC had no effect on NK cell activities. These activities were enhanced by culture withβ‐interferon for 18 hours, as were the NK cell activities of healthy controls. These results suggest that reduced numbers or imbalances in the lymphocyte population with NK cell activity, are responsible for the pathogenesis and the prognosis of adult type atopi

AbstractStudies were performed to determine whether the sera of the patients with granuloma annulare had monocyte modulating activities. As indicators of monocyte modulation, spreading and aggregation were used. As a control, similar experiments were performed on pooled normal human sera. Normal human monocytes were incubated with either test sera or control sera. The percentage of spread cells was estimated at 6 hr of incubation and that of aggregated cells was estimated at 24 hr of incubation. The percentage of spread cells and aggregated cells in the monocytes incubated with granuloma annulare sera was higher than that of the cells incubated with pooled normal human sera. Thus, granuloma annulare sera may have a greater ability to induce monocytes to spread and aggregate than do normal human sera.

AbstractAn improved and simplified hamster ear model (whole mount technique; WMT) is described. Excised ear skin specimens were incubated in 2N NaBr at 37°C for 2 hours, yielding a sheet of dermis with the sebaceous glands attached. After Sudan III staining, the areas of the sebaceous glands in the sheet were measured with an image analyzing system. The ratios of a mean area of androgen‐stimulated sebaceous glands/a mean area of non‐treated sebaceous glands (AS/NT) were 2.56 in females and 1.69 in males; these results closely resemble AS/NT ratios found in histological sagittal sections. Over 1.0% concentrations of topically applied cyproterone acetate were highly effective in suppressing the proliferation of sebaceous glands in androgen‐stimulated hamsters, as measured by WMT. Since no routinely processed histological sections are necessary and the exact size of a whole sebaceous gland can be obtained by WMT, this method seems to be useful for routine work to assess the efficacy of topically applied anti‐comedogenic or anti‐lipog
