AbstractThe dermis and sera of 11 patients with pseudoxanthoma elasticum (PXE) were analyzed by several techniques which test for the presence and quantity of trace elements (Fe, Ni, Co, Cu, Mn, Cr, Zn, Pb, Cd, Al, S, K, Cl, Ca and P). The investigative methods included 1) electron microscopic point X‐ray microanalysis, 2) inductively coupled plasma atomic emission spectrophotometric analysis (ICP‐AES), 3) X‐ray diffraction analysis and/or 4) atomic absorption spectro‐photometric analysis. 1) Among 7 specimens of dermis showing histological degeneration of the elastic fibers, 2 showed prominent levels of A1. In early degeneration of the elastic fibers, Ca was present in only small amounts; however, in more fully developed lesions, large quantities of both Ca and P were found, while Fe, Co, Ma and Zn were detected in trace amounts. 2) ICP‐AES analysis confirmed large deposits of Al, Ca, and P in diseased dermis (Al: 32.1 ppm wet tissue); other elements were found in normal quantities. Al was not detected in the control dermis. 3) X‐ray diffraction analysis suggested the pattern of hydroxyapatite [Ca10(PO4)6(OH)2], which indicates that most of the Ca exists in a crystalline form. 4) Atomic absorption spectrophotometric analysis confirmed a large concentration of Al (11–21μg/dl, mean=17.2; normal<10) in all test patient's sera. The combined analysis revealed an increased amount of Al in patient's sera as well as in the elastic fibers of the dermis at an early stage of this disease (PXE). AbstractHuman Langerhans cell free epithelia can be culturedin vitro, and then can be used as epidermal allografts (EAG) without evidence of rejection. We studied the cellular basis of this phenomenon with mixed epidermal cell lymphocyte reactions (MELR). The capacity of donor‐derived epidermal cells to stimulate allogeneic control or recipient cells was abolished when stimulatory cells were Langerhans cell‐free cultured keratinocytes and respondors were obtained prior to grafting. Donor‐type cultured keratinocytes were able to induce a low response by host‐derived cells in assays conducted 2, 4 and 6 weeks after grafting, but not thereafter. They were unable to stimulate allogeneic cells unrelated to the recipient. The ability of crude epidermal suspensions with 2–4% Langerhans cells to stimulate host‐derived cells did not increase with time after grafting. No secondary type MELR could be evidenced, suggesting that grafting of EAG did not induce anin vivoimmunization to class I antigens expressed by cultured epidermal cells. Lastly, host‐derived Langerhans cells were not able to restore the allostimulatory ability of Langerhans cell free epidermal cells from EAG donors when tested against host‐derived cells. This suggests that host‐derived Langerhans cells which colonize the grafts during the first few weeks following grafting cannot act in the presentation of foreign keratinocyte‐bound antigens, which may account for the absence

AbstractIn order to elucidate the mechanism of UV‐induced skin inflammation, the effects of antimalarials, inhibitors of phospholipase A2and membrane phospholipid methylation, on UV‐induced ear swelling response (ESR) was examined. Antimalarials inhibited ESR induced by 8‐methoxypsoralen (0.5%) plus UVA irradiation (PUVA treatment) or UVB irradiation. AbstractThe polarization assay was used to assess the effects of chemoattractants on neutrophil motility by means of cellular configuration. The addition of N‐formyl‐L‐methionyl‐phenylalanine (f‐Met‐Phe) to a neutrophil suspension from human peripheral blood induced a polarized change in cellular configuration, and the ratio of such transfiguration reached a maximum at 10–8to 10–6M of f‐Met‐Phe. The polarized configuration appeared 1 min after addition of f‐Met‐Phe and showed its maximum response at 10 min. The number of polarized cells were decreased after exposure to cytochalasin B or incubating with corticosteroid. The increased response of random polarization was induced in a dose‐dependent fashion by ascorbic acid. Tetracycline HCl did not cause any changes in cell shape at any concentrations examined. AbstractIn order to study the pathogenic role of autoantibodies in the serum of EBA patients, we tried to induce EBA lesions in mice by administration of the IgG fraction of EBA serumviathe intraperitoneal (i.p.) or subcutaneous (s.c.) routes into neonatal or adult mice. We also tried to induce EBA lesions in fetuses by intravenous (i.v.) injection of EBA serum into pregnant mice.This study demonstrates that IgG fraction of EBA serum injected into neonatal or adult mice, but not into fetal mice, deposits at the epidermo‐dermal junction in a linear fashion but fails to induce bullae. AbstractUsing immunoblotting techniques, sera from patients with systemic lupus erythematosus (SLE) and progressive systemic sclerosis (PSS) and from normal individuals were tested for the presence of antibodies to the intermediate filament protein (vimentin) of the cytoskeleton of human fibroblasts. Antibodies to vimentin were found in sera from both patients and normal controls. By the enzyme‐linked immunosorbent assay (ELISA) using vimentin purified from cultured human skin fibroblasts, we determined the titer of the antibodies in PSS (50 cases), and SLE (8 cases), and normal sera (48 cases). Significantly higher titers of anti‐vimentin auto‐antibodies were detected in the patient sera than in normal controls. According to the titers, the cases of PSS were divided into two groups: one with titers higher than the range of normal controls, and another with titers within the range of normal controls. The relationships of the titers to the clinical features were examined. AbstractThirty four cases of PSS were treated once or twice a week by a low frequency electrical acupuncture method for periods of 3 months to 9 years and 2 months. On clinically worse side of arm, two meredian points,HatijaandShokai, were used for the electrical stimulation. As the result, various degrees of improvement were obtained in ulcerations on the fingertips or toetips, arthralgia, cutaneous sclerosis, hypo‐ and hyperpigmentation and sensory disturbance etc. and in respiratory functions, plethysmography, capillary microscopic features of the nail bed and histopathologic features biopsied from the forearms. AbstractPropionibacterium acnesstrains were isolated from the comedones of 46 acne patients, and their susceptibilities to penicillin G, ampicillin, erythromycin, clindamycin, tetracycline, doxycycline, minocycline, cephalexin and gentamycin were studied by examining their minimum inhibitory concentrations. Two additional standard strains were included in the study. In the results, differing antibiotic susceptibilities were found, andP. acnesstrains were the most sensitive to erythromycin and clindamycin, followed by ampicillin and minocycline. The highest levels of resistance were observed against clindamycin, erythromycin and tetracycline. Non‐resistant strains were detected only against minocycline. A comparison of these results with others obtained by similar studies in our department during the last 15 years showed a progressive tendency toward antibiotic resistance forPropionibacterium acnes, probably related to the systemic administration of antibiotics for the treatment of bacterial infections other than acn
