AbstractOmission of L‐glutamine (Gln) from the culture medium decreases tyrosinase activity and increasesγ‐GTP activity in cultured melanotic B‐16 melanoma cells; inclusion of this amino acid in the medium shows the reverse effects. Its absence, however, decreases theγ‐GTP activity in non‐pigmented cells like HeLa cells, fibroblasts and amelanotic melanoma cells and its presence induces a dose dependent increase in this enzyme activity in these cells. The decrease ofγ‐GTP activity in the melanotic melanoma cells by Gln is attributed to increased production of dopa and its derivatives by these cells, as evidenced by our results. Abstractα‐Aminoisobutyric acid transport has been demonstrated in cultured human epidermal keratinocytes as well as in the transformed state of these cells. The concentrative uptake is sodium‐dependent and may be ascribed to one Michaelis‐Menten component whose maximal velocity is 6.9 nmol/min × 106cells, with an apparent affinity of 3.8 mM. These parameters may be modified, depending upon the nature of malignant transformation. In SVK14 SV40‐virus‐transformed cells, there is no change of affinity but the maximal velocity is 1.5 fold less than in normal cells. In the spontaneous epidermoid carcinoma‐derived A431 line, this phenomenon is inverted; the maximal velocity is unmodified but the system affinity is 2.2‐fold higher than in normal cells. AbstractCounterflow centrifugal elutriation (CCE) is one of the most powerful and rapid methods for separating free cells by sedimentation velocity based on cell size, density, or shape. To obtain a sufficient number of cells, the fractionation of guinea pig epidermal keratinocytes was developed with the use of a newly‐developed elutriation system. An EDTA‐trypsin method was used to isolate keratinocytes; they were then separated into three groups according to flow rate by CCE. Morphologically, the keratinocytes in each region were identified as basal cells, spinous cells, and granular cells. The DNA distribution pattern agreed well with that obtained by Percoll's density gradient. AbstractA haematoxylin stainable protein (HSP) extracted from the EDTA‐separated epidermis of 3‐day‐old rats was purified through preparative isoelectric focusing followed by fast protein liquid chromatography (FPLC) using a Superose 12 column and a Mono‐Q anion exchange column. The purified protein was a single protein in SDS‐PAGE; it had a molecular weight of 14,500 daltons and an isoelectric point of pH 4.7. AbstractThe distribution of 2,4‐dinitrophenyl (DNP) groups on Thy‐1 positive epidermal cellsin vivoafter 2,4‐dinitrochlorobenzene (DNCB) was painted on the skin surface of C3H/He mice was examined. Epidermal cell suspensions (EC) derived from trypsin‐separated mouse ear skin painted with DNCB were double‐stained for DNP groups and Thy‐1 protein using FITC‐anti‐DNP antibody, biotinylated anti‐mouse Thy‐1,2 monoclonal antibody, and phyco‐erythrin conjugated avidin. There were on the average 1.7% Thy‐1+ECs; DNP groups were detectable on 77.8% of them when the specimen was treated with anti‐DNP followed by anti‐Thy‐1,2 incubation. AbstractSera of a 30‐year‐old Italian female and a 32‐year‐old Japanese female with prolidase deficiency were analysed by two‐dimensional immunoelectrophoresis, rocket immunoelectrophoresis, and single radial immunodiffusion (SRID). AbstractIn order to improve the oxidative stability of hydroquinone (HQ), the only depigmenting agent available clinically for skin, 14 hydroquinone fatty acid esters (HQ‐Rs) were synthesized and examined for their stability, efficacy, and intradermal metabolism. HQ‐Rs containing more than 3 carbon atoms in the fatty acid side chain showed excellent stability in an aqueous base (pH 6). HQ‐Rs containing fewer than 8 carbon atoms in the fatty acid side chain exhibited skin depigmenting potencies better than that of HQ. HQ‐R was quantitatively hydrolyzed by a skin homogenate to liberate HQ. The hydrolysis velocity of the HQ‐Rs was dependent on the length of the fatty acid side chain; the longer the length, the slower the hydrolysis velocity. From these results, we concluded that HQ‐R is absorbed into the skin after topical application, hydrolyzed by esterase, and it depigments the skin by liberating HQ. AbstractSerum gastrin levels did not change during topical corticosteroid therapy with an experimental dose of 20 to 60 g/day for seven consecutive days, and serum 11‐hydroxycorticosteroid levels were significantly suppressed during the treatment. On the other hand, after daily oral administration of 1.5 mg/day betamethasone for seven consecutive days in volunteers who had fasted overnight, serum gastrin levels were higher and 11‐hydroxycorticosteroid levels were significantly lower.Fasting serum gastrin levels were significantly increased in rats after intraperitoneal administration of prednisolone at dosages of 2, 10, 20, or 50 mg/kg/day, but they were not significantly altered after the administration of prednisolone at 0.5 mg/kg/day. Serum gastrin levels were not significantly altered in man or rats after the administration of a histamine H1receptor antagonist. AbstractFixed drug eruption incidence was 0.3% of outpatients. It affected individuals of both sexes in their 3rd and 4th decades, with a preponderance of males.
