A calcium binding IgG was isolated and purified by column chromatography from serum of a myeloma patient with asymptomatic hypercalcaemia. The myeloma IgG, characterized as an IgG kappa, revealed a normal sized heavy chain (56000 dalton), and a light chain of 31000 dalton. Another population of IgG separated and purified from the same patient's serum did not bind calcium and had a normal 26000 dalton light chain.Calcium binding activity in vitro is optimal at pH 8.0, and reaches its maximum after 3 h of45Ca myeloma IgG incubation. Cleavage of the purified IgG by trypsin yielded peptides which were further isolated by column chromatography and characterized as Fab and Fc fragments. Light and heavy chains were obtained by reacting the immunoglobulin with dithiothreitol and iodoacetamide followed by Sephadex G‐100 chromatography. Diffusion Chambers (DC) were used to culture human bone marrow of ALL patients at the beginning and after the cessation of therapy.No growth was observed in patients before treatment, while growth patterns similar to the normal were obtained in patients after 3 years of maintenance therapy.We suggested the possibility of intensifying the multiple drug chemotherapy in ALL without permanent damage of the stem cell proliferative compartment.

Leukaemic human bone marrow and peripheral blood cells were cultured for 25 d in diffusion chambers implanted into cyclophosphamide treated mice. Normal bone marrow cells were cultured simultaneously. These cells were studied both morphologically and functionally (CFU‐C).The leukaemic cells behaved heterogeneously, 2 groups being distinguishable in accordance with their initial in vitro growth pattern (1: no growth or microcluster growth, 2: macrocluster growth). Group I showed progressive cellular death with a diminution of granulocytic progenitors and the appearance of a predominantly macrophagic population. This behaviour resembled that of the control group. The initial microcluster growth pattern remained identical throughout the entire culture period. Activation of Factor IX and Factor X was studied by adding125I‐Factor IX or125I‐Factor X to reaction mixtures and quantitating cleavage products by reduced sodium dodecylsulfate gel electrophoresis. Thrombin failed to activate Factors IX or X; Factor Xaproduced insignificant amounts of cleavage products of both factors. In contrast, the reaction product of tissue factor and Factor VII cleaved large amounts of both Factor IX and Factor X in purified systems and in plasma. In incubation mixtures of plasma containing added125I‐Factor IX or125I‐Factor X, tissue factor and Ca2+ions, the percentage of total radioactivity in the heavy chain peak of125I‐IXaand the heavy chain peak of125I‐Xaincreased at a similar rate. When the tissue factor was diluted, similar curves were obtained for percent cleavage of125I‐Factor IX and percent cleavage of125I‐Factor X plotted against tissue factor concentration. Incubation of the non‐adherent low density cells from normal human bone marrow with an antiserum to DR antigens markedly inhibited their capacity to form granulocytic colonies in fibrin clot diffusion chambers implanted intraperitoneally into irradiated mice. The inhibitory effect of antiserum was abrogated after absorption of antiserum with B cells. Inhibition was obtained both with and without complement treatment. Neutrophilic colony formation exhibited a higher sensitivity to diluted antiserum to DR antigens as compared with eosinophilic colony formation. We measured the concentration of granulocyte‐committed progenitor cells (CFU‐c) and the alkaline phosphatase activity of neutrophils harvested from colonies cultured from the peripheral blood of 30 patients with high‐count chronic granulocytic leukaemia (CGL) in the chronic phase. Neutrophils in colonies cultured from marrow of normal donors or patients with acute myeloid leukaemia in remission served as controls. CFU‐c numbers in CGL peripheral blood were on average 3 times higher than in normal marrow (130.1 + 126.2 (SD) versus 43.0 + 25.2 per 1 × 105cells plated, respectively). The mean NAP scores in cultured CGL neutrophils were substantially higher than control values (89.9 + 48.0 versus 55.9 + 33.0 units, respectively). There was a tendency for peak values of NAP in culture to be reached at earlier points in CGL cultures with high CFU‐c numbers (i.e. high proliferative rates) than in those with lower CFU‐c numbers (lower proliferative rates). In response to specific treatment of vitamin B12deficient, anaemic patients there is an influx of folate into the young, circulating red cells.To separate new and old cells, capillary tubes filled with whole blood were centrifuged and the packed red cell column divided into top (T), middle (M) and bottom (B) layer. The newest cells are found in the T layer.The increase in red cell folate (RCF) concentration starts before, during or after the reticulocyte response, and is therefore not directly related to folate metabolism in the red precursor cells in the marrow. The low RCF concentration at the peak of the reticulocyte response in some of the cases demonstrates that the folate material, which may have been accumulated in the red precursor cells in the marrow, may be lost by the time the red cells enter the peripheral blood.The influx of folate into the young, circulating red cells is rapidly followed by an efflux of folate, suggesting that much of the folate material is still in the monoglutamate form. A new influx of folate is noted after a time lapse of from 5 to 10 d. Iron deficiency seems to prevent the uptake of folate by the circulating red cells.

A simple and highly reproducible method is described for separation and density distribution analysis of human bone marrow cells in continuous density gradients of polyvinylpyrrolidone coated silica gel (Percoll). Colony and cluster forming cells in agar separated from the bulk of cells and peaked at densities of 1.063‐1.064 g/ml. The enrichment of clonogenic cells was approximately 10 times and recovery varied between 41–321%. The overall recovery of cells was 80% (60–94%). Density distribution analysis of morphologically identifiable cells demonstrated the progressive increase in density with maturation of cells within the granulocytic series: myeloblasts peaked at 1.0624 g/ml, promyelocytes at 1.0734 g/ml, myelocytes at 1.0776 g/ml, metamyelocytes at 1.0799 g/ml and mature neutrophils at 1.0864 g/ml. The eosinophil had the highest density, 1.0904 g/ml, of all cells analyzed. Monocytes and lymphocytes peaked at 1.0661 and 1.0681 g/ml respectively. Using stereological techniques at the ultrastructural level, 7 quantitative parameters for 7 patients with hairy cell leukaemia, 27 patients with chronic lymphocytic leukaemia, and 8 normal individuals. These quantitative data show clear differences between normal lymphocytes, CLL cells and the abnormal cells of HCL. They also show that it may be possible to subdivide CLL into distinct groups on quantitative criteria.
