A 79‐year‐old man with acute myelo‐monocytic leukaemia was found to have an IgG paraprotein on serum electrophoresis and a moderate increase in plasma cells (15 %) in the initial marrow examination. Treatment of the acute leukaemia with cytosine arabinoside and daunorubicin was followed by an increase in plasma cells in the marrow to 40 %. Detachment of parts of cytoplasm (clasmatocytosis) from lymphocytes was observed in the bone marrow of normal and sublethally irradiated rats. Though the function of the detached parts remains to be established, it is postulated that clasmatocytosis may represent a secretory function of the lymphocyte. The phenomenon is compared to clasmatocytosis in plasma cells thought to represent one way of secretion of immunoglobulins.

NBT tests performed on simultaneously drawn venous and capillary blood from 56 patients showed a considerably lower proportion of NBT positive neutrophils in capillary blood than in venous blood. The presence of serum was necessary for the NBT test to function and fresh serum was more efficient than inactivated serum. Haemolysed blood reduced the proportion of NBT positive neutrophils and this may explain, at least in part, the observed difference between venous and capillary blood.

The effects of adrenergicα‐and β‐receptor stimulating and blocking drugs on the splenic release of white cells into the blood were studied in guinea pigs. Furthermore, the effect of chemical sympathectomy was investigated. The release of white cells from the spleen was measured by the comparison between the cellular content in splenic afferent and efferent blood. The determination of the splenic blood flow with the xenon 133 clearance method enabled calculation of the number of white cells released from the spleen per min.Within 5 min after administration, noradrenaline (NA) and isoproterenol resulted in an increased splenic veno‐arterial difference in number of lymphocytes and granulocytes. As the splenic blood flow was not significantly altered, this indicates that the two drugs markedly increase the release of white cells from the spleen. NA also increased the number of lymphocytes per μl of arterial blood and the arterial haematocrit.The effects of NA were blocked by phentolamine and the effects of isoproterenol were blocked by propranolol. Propranolol, phentolamine, or chemical sympathectomy did not abolish the splenic veno‐arterial difference in cellular content found in control animals.The results indicate that the basic release of splenic lymphocytes occurring in normal guinea pigs is independent of adrenergic innervation and stimulation, and that specific adrenergic stimulation ofα‐or β‐receptors causes a considerable mobilization of lymphocytes and granulocytes

The effects of ristocetin on human platelets were investigated by aggregometry and electron microscopy. We confirmed the finding that ristocetin induces platelet aggregation. Biphasic platelet aggregation, such as is observed with ADP, was also seen with ristocetin. The gross appearance of ristocetin aggregated specimens differed markedly from those induced by ADP or collagen. On electron microscopic examination the boundaries of the aggregated platelets were noted to be extremely tortuous with electron dense lines between membranes, and there were many swollen α‐granules. At the periphery of the aggregates, the platelets were markedly degranulated. The plasma membranes of degranulated platelets were occasionally lysed. The postirradiation recovery of the myeloid system of normal and hypoxia pretreated mice was studied by quantitative means. In contrast to normal mice, the myeloid system of which recovered with a rise‐fall‐rise pattern, the hypoxia pretreated irradiated mice showed a sustained recovery with an earlier return of cell numbers to the normal level. Quantitative measurement of the ‘available pool’ of mature neutrophils revealed an enhanced production of neutrophilic granulocytes in mice pretreated with hypoxia. The effect of erythropoietic suppression, which occurs in these mice after cessation of hypoxia, on the myeloid recovery was also studied in mice made polycythaemic by hypertransfusion. The suppression of erythropoiesis per se seems not to be entirely responsible for the improved recovery of the myeloid system. An immunological study has been made of 12 patients with myelofibrosis. The study included determinations of serum protein and immunoglobulin concentrations, serial tuberculin skin tests, and in vitro stimulation of peripheral lymphocytes with phyto‐haemagglutinin (PHA). In two patients, very low gammaglobulin values were found (low IgG and IgA concentrations; low IgM in one of them). Four other patients had subnormal IgA values. Depressed delayed‐type hypersensitivity reactions were observable in four of eight patients tested. The immuno‐deficiency noted was probably secondary to the bone marrow damage. An erythropoiesis inhibiting factor (EIF) with a low molecular weight has been demonstrated in urine from normal healthy individuals, indicating a dual, humoral regulatory mechanism of the erythropoietic homeostasis. Since oestrogens are known to inhibit erythropoiesis and are excreted in the urine, the question was raised whether the EIF preparations could contain eostrogens in such amounts that this could explain the inhibitory effect. Fractionated eostrogen determinations in EIF preparations revealed only micro‐quantities of the different oestrogen‐metabolites. An inhibitory effect on erythropoiesis was obtained with commercial oestrogens only in doses 104‐105times higher than the oestrogen amounts found in the EIF preparations. Isoenzymic patterns of 12 enzymes were investigated in extracts of eosinophils isolated from the peripheral blood of a patient with eosinophilic leukaemia using the poly‐acrylamide gel electrophoresis technique.
