Zirconyl phosphate gel at pH 6.25 binds selectively to transcobalamin II and not to transcobalamin I. The in vivo effect of daunomycin on the cell kinetics of leukaemic blast cells has been investigated in 4 patients with acute myeloblasts leukaemia and in 2 patients with acute lymphoblastic leukaemia by a method of combined autoradiography and microspectrophotometry. After a dose of about 1.5 mg daunomycin/kg bw a decrease in the mitotic index was observed, and in 4 cases also a depression of the in vitro3H‐thymidine labelling index was noticed. Microspectrophotometric analysis showed an accumulation of cells in G2phase and a depression of DNA synthesis. In 3 of the patients studied a rapid decline in the peripheral blast cell count was noticed. The data indicate that the effect of daunomycin on cell proliferation is a randomized cytocidal effect, superimposed on which there is a specific inhibitory action on the progression of cells through G2phase and a depression of DNA synthesi

Aspirates obtained simultaneously from the bone marrow and the spleen in 19 patients with chronic myeloid leukaemia (CML) were incubated with tritiated thymidine (3HTDR). The labelling indices (LI) of the myelocytes were determined autoradiographically. With increasing leucocyte counts the LI of the bone marrow myelocytes did not change significantly. In the spleen aspirates there was a significant increase in the LI of the myelocytes with increasing WBC counts from 10, 000 to 300, 000. The results indicate that the granulopoietic tissue in the spleen is not functionally identical with that of the bone marrow in CML.

The hereditary syndrome selective malabsorption of vitamin B12is analyzed from a genetical point of view. 32 families with 55 affected children were collected from the literature. The proportion of affected children in these sibships is consistent with a recessive autosomal transmission. Furthermore, the ancestors of 15 Finnish propositi were traced several generations back. The accumulation of families at local and national levels is discussed.

The present study distinguishes the morphological changes associated specifically with the thrombin‐induced platelet release reaction as isolated from platelet aggregation. At a time when maximal amounts of calcium and nucleotides are discharged from the cell, there also occurs an almost complete loss of alpha granules from the cytoplasm. Partial inhibition of the thrombin‐induced release reaction by incubation of the platelet suspensions at different temperatures is correlated with the discharge of fewer alpha granules from the cells.A complex lamellar‐vacuolar network gradually develops in the cells as release becomes maximal. Portions of this system may be derived from the limiting membranes of alpha granules that have fused with the lamellae and deposited their contents. Because this membrane system is continuous with the surface of the cell during release, it may provide a conduit whereby the alpha granule contents are transported to the surrounding medium.At 37°, when maximal thrombin‐induced release occurs, a heretofore undescribed electron‐dense fibrous material appears within the lamellar spaces. The present data indicate that disseminated intravascular coagulation experimentally induced leads both to consumption as well as synthesis of clotting factors. In massive DIC all parameters are markedly affected, and the diagnosis easily settled. In low graded DIC normal or even high levels of clotting factors may be met with. In this situation the ethanol gelation test appears to be a sensitive indicator.

Inbred albino rats, aged 5–6 months, were given a single injection of Cortisol or injections of Cortisol every 6 h for up to 3 d. At varying times in relation to the Cortisol administration a single injection of3H‐thymidine was given. The eosinophils in the peripheral blood and in suspensions of splenic tissue were studied by counting the absolute number of eosinophils and by autoradiography. These investigations showed that Cortisol induces a sharp decrease of eosinophils in the splenic tissue due primarily to a reduction in the number of mature eosinophils. At the same time, there was a relative increase in the number of proliferating eosinophils. Long‐term administration of Cortisol results in an inhibiton of mitotic activity of the eosinophil precursors. There was no indication that the peripheral eosinopenia following Cortisol administration could be due to migration of eosinophils to the spleen and/or splenic destruction of eosino

The effect of surgery on the number of circulating megakaryocytes in the inferior vena cava was studied in 58 female rats 1–20 d after intervention. The effect of busulphan on haematopoiesis was investigated in mice. Bone marrow and blood cells were sampled 2 h to 55 d after a single treatment with busulphan. The effect on progenitor cells was examined with the diffusion chamber (DC) technique and the spleen colony assay.Busulphan had a severe depressive effect on progenitor cells in bone marrow, and their number was reduced to 0.01–0.1 % of the control value on day 5–6. During the first 2 d the CFUs were reduced more than granulocyte progenitors as determined in the DC system. Histological examination of spleen colonies showed that in the same interval erythroid colony number was more depressed than granuloid colony number. This situation reversed after 4–6 d.Our interpretation is that busulphan affects all types of progenitor cells irrespective of their proliferative state. However, multipotent CFUs in Go‐phase are more depressed than granulocyte progenitor cells (DC), which have a higher proliferative rate. The selective effect on erythroid colonies during the initial period might indicate that busulphan impairs the ability of multipotent CFUs to erythroid differentiation. This damage was repaired within a few days.An abortive regeneration of bone marrow cells was observed on day 7–10.
