The specific activity of glycine‐2‐14C into haem and globin has been studied in normal marrow, in cases of thalassaemia and refractory normoblastic anaemia, in patients with iron deficiency anaemia and porphyria of erythropoietic origin. In controls the ratio of haem to globin was less than 1.0. This ratio was reduced in the 2 cases of thalassaemia probably due to the associated reduced activity in ALA synthetase and haem synthetase enzymes. Haem synthesis by iron deficient erythroid precursors can be stimulated by iron with corresponding increase in globin synthesis. There is apparently a mechanism controlling the relationship between haem and globin. Some cases of sideroblastic anaemia resemble patients with thalassaemia in the reduction of the ratio of haem to globin and in the marked reduction of the synthesis of haemoglobin when related to cell counts.When the ratio of the specific activity of haem to globin is reduced the in vitro addition of pyridoxine to the duplicate may allow one to determine whether it will be effective in therapy.Reduced globin synthesis in pre‐leukaemic sideroblastic anaemia may carry a poor prognosis but conversely adequate globin production in relationship to cell counts may be associated with a rapidly fatal outcome.Only in the patient with erythropoietic porphyria was the specific activity of glycine‐2‐14C into haem synthesis greater than into globin. Electron microscopic studies of circulating neutrophil granulocytes of a boy suffering from Fatal Granulomatous Disease (FGD) showed two abnormal kinds of cytoplasmic granules, both larger than normal granules. One was characterized by a homogeneous matrix of very low electron density, the other contained one or more crystalloid inclusions of larger size than found in normal neutrophils.In neutrophil granulocytes from adults with long‐standing bacterial infection, by light microscopy showing toxic granulation, the granules displayed the same ultrastructural abnormalities as in FGD.It is concluded that the ultrastructure of the granules of neutrophils in FGD reflects non‐specific toxic granulation. The radial segmentation (RS) of nuclei of human lymphocytes and monocytes was inhibited by vinblastine. Sulfhydryl reagents did not inhibit the formation of RS nuclei. The yield of RS nuclei after incubation in heparinized blood is probably due to the temperature during incubation and not to heparin, as evidenced by comparison with blood samples held fluid by defibrination with streptokinase. In the concentration range that potentiates muscle cell contraction caffeine had no significant influence on the formation of RS nuclei. These findings are discussed in relation to the hypothesis that microtubule shrinkage due to their slow decomposition is instrumental in the formation of RS nuclei.

The technique of double‐label, double‐emulsion autoradiography was used to study the proliferative activity of bone marrow eosinophils in male rats of four age groups (weanlings, young adults, adults and aged adults). No alterations in the duration of the cell generative cycle (Tc), DNA synthetic period (Ts), or mitosis (TM) were observed to accompany aging. Estimates of Ts were 10 1/2–11 hours; of TM, 0.33 hours; of generation time, 22–25 hours. Hourly production rates varied with age only when related to body weight, with estimates ranging from 10.2 ± 2.1 times 103eosinophils/hour/mg. marrow/100 g. in the youngest rats to 1.6 ± 0.3 times 103eosinophils/hour/mg./100 g. in the oldest rats. The ability of subcellular fractions of rabbit spleen and liver to induce haemolysis of rabbit erythrocytes was investigated. It was shown that:1Subcellular fractions of normal spleen and liver induce haemolysis of rabbit erythrocytes modifiedin vivoby injections of small doses of phenylhydrazine.2The haemolytic activity of both tissue preparations was located mainly in the mitochondria and was much higher in the spleen than in the liver.3Fresh preparations of the spleen or the liver do not induce haemolysis of intact erythrocytes.4When the tissue fractions and also the erythrocytes were obtained from phenyl‐hydrazine treated rabbits, the haemolytic activity of the spleen preparations was lower and that of the liver was slightly higher than the respective activities of preparations of normal tissues.5The haemolytic activity of the subcellular fractions kept frozen for 7 days at pH = 7.0 was significantly increased.Phospholipase A is indicated as a possible haemolytic agent present in the spleen and the liver mitochondri

A case of thrombasthenia in a 63 years old man is presented. He had cancer of the colon and bled continuously from the rectum. After transfusion of platelets from ten donors, platelet count and bleeding time were unchanged. Nevertheless, a resection of the colon was successfully performed.

Five young, healthy women with regular menstruations and who were not taking oral contraceptives were followed with repeated blood samples for 5 weeks. On each individual, about 15 determinations were made of the number of platelets, the fibrinogen and plasminogen concentration, the P&P‐test, the Factor VIII activity and the spontaneous fibrinolytic activity of the euglobulin fraction of plasma on fibrin plates. It was shown that the variation in these parameters in the individual was smaller than the variation between individuals and that distinct individual ranges could be found for some women. This was most pronounced with respect to the number of platelets, the fibrinogen concentration and the P&P‐test. The phagocytic activity of peripheral blood granulocytes was assayed in 12 normals, 7 cases of rheumatoid arthritis and 7 cases of systemic lupus erythematosus. Phagocytosis was indistinguishable in the normal group and the R. A. group. The phagocytic capacity of the granulocytes from the S. L. E. group was significantly lower than in the normal group and the R. A. group. Phagocytosis was not reduced when normal granulocytes were suspended in S. L. E. plasma. Nor did the activity of S. L. E. granulocytes increase in normal plasma. The lowest phagocytic indices found in the S.L.E. group tended to be associated with neutrocytopenia.

The turbidimetrically obtained curves of ADP‐induced platelet aggregation in plateletrich plasma at 37° C. can be mathematically characterized by a logarithmically normal frequency distribution suggesting that the phase of aggregation and the subsequent disaggregation are governed by the same mechanism. Every curve can be described with reference to three parameters, viz.: the maximum change in optical density (ODmax), the time at which this occurs (tmaxand the standard deviation of the logarithms of the time variable (s̀).Aggregation curves of 21 normal subjects, 17 uraemic patients and 11 patients with Von Willebrand̂s disease were studied.Parameter ODmaxalthough dependent on the platelet concentration was significantly diminished in a proportion of the uraemic patients.Neither group of patients differed significantly from the normals in parameter tmax. A significant number of uraemic patients and Von Willebrand patients showed lower s̀ values than those found in normal subjects. The abnormality thus indicated entails abnormally rapid disaggregation.The new parameter (s̀) proved to be a better reproducible and more sensitive measure of the aggregation process than the hitherto usual parameters ODmaxan
