The anticoagulant, EDTA, is known to produce changes in the external contour of platelets. In the present investigation the fine structural alterations which develop in EDTA platelets were examined in the electron microscope. Platelets exposed to EDTA at 37° C. lose their disc shape and become irregularly swollen with multiple pseudopods. This external transformation is identical to the change which occurs in platelets exposed to ADP, thrombin, or low temperature. Internal changes in EDTA platelets, however, are markedly different from those which occur under the influence of aggregating agents or cold. The differences indicate that the effects of EDTA on platelet shape are not due to dispersal of marginal band microtubules or to the trigging of an internal wave of contraction. Chelation of membrane calcium by EDTA appears to cause marked irregularities in the platelet wall and massive swelling of the canalicular system. Swelling of internal channals is the dominant alteration in EDTA platelets, and this change appears ultimately to be related to the breakdown of platelet granules

The influence of temperature on thein vitroformation of radial‐segmented (RS) nuclei in lymphocytes and monocytes from peripheral blood was studied quantitatively. The RS nuclei were found to be most abundant after incubation at room temperature, independent of the anticoagulant used. The smallest yield of RS nuclei was found after incubation at refrigerator temperature, a somewhat higher yield after incubation at 37° C. These findings are discussed in relation to the hypothesis that the formation of RS nuclei is due to a contraction of fibrous structures radiating from the centrosomal region, presumably interphasic remnants of the mitotic spind

Report of a case of immune haemolytic anaemia and thrombocytopenia antedated by 19 years with biologic false positive serological reactions for syphilis and initially associated with an aplastic crisis. The erythrocytes were heavily sensitized with complement and weakly with γG globulin. In spite of elevated cold agglutinins, the patient had not experienced Raynaud's phenomenon and there was no abnormal cold haemolysis. Ultracentrifugation studies revealed a quality change in the Wassermann antibodies that roughly parallelled the development of the haemolytic disease

A woman developed severe thrombocytopenia a few days after the 8th injection of sodium aurothiomalate. She was treated with BAL, prednisone and a fresh blood transfusion, and the platelet count came back to normal after about 2 weeks. The micro‐incubation chamber technique of Cunningham (1965) has been used for testing transformed cells in sheep red cell stimulated lymphocyte cultures for the ability to give pericellular lysis (lytic plaques) in sheep red cell monolayers. The variations around the means for total cell numbers and plaque‐forming cell numbers in series of chambers plated in parallel have been estimated.The plaque‐forming ability was found to be dependent on complement and temperature indicating an immunological lytic reaction. Sheep red cell‐stimulated cultures containing lymphocytes from sheep red cell immunized rabbits were harvested after different culture periods. Blastoid cells appeared and increased in numbers in the first 4–6 days of incubation, with subsequent reduction. Transformed cells with the ability to form pericellular lytic plaques in sheep red cell monolayers were regularly found after 6–8 days' incubation. A material consisting of 13 normal controls and 32 patients suffering from various types of secondary thrombocytopathy and some special cases has been tested by a modified thrombin generation test and Stypven clotting time in order to evaluate platelet factor 3 activity.Intact platelet suspensions, platelets damaged by freezing and thawing or ultrasonic vibrations and phospholipid extracts have been added to platelet‐free, non‐activated citrated plasma prepared from the same normal person in all experiments. The effect on velocity and amount of thrombinformation and on Stypven clotting time have been estimated in the total group. The results have been evaluated statistically.The modified thrombin generation test did not show any significant differences in activity in any cases tested.The Stypven‐test was technically much more satisfactory and significantly less activity was found using platelets from patients with hepatic cirrhosis. The optimal conditions for the hydrolysis of hexose and hexosamine components in the mucosaccharide fraction of red cell stroma were found to be 2 N HCl at 92° C. for 3 hours. The hydrolysate of stroma was converted by concentration, neutralization and ion exchange technique to a form in which the hexose and hexosamine components could be assertained. By determining the recovery and standard deviation the methods were found reproduceable. The normal values of total aldohexose, galactose, glucose and hexosamine concentrations were determined in stroma in about 20 cases of each of the following species: human, bovine, pig, and horse. The stroma was divided into two parts with a chloro‐form‐methanol mixture (2:1) and the hexose and hexosamine concentrations in each part were determined. Pig stroma, contrary to that of the other species, had markedly more hexoses in the lipid fraction than in the stroma residue fraction. The content of glucose in both fractions was the same in human stroma, but there were more hexoses and hexosamines in the stroma residue fraction.
