A microspectrophotometric method was used for determination of the amount of periodic acid‐Schiff reactive material (PASMa), most likely a measure of the glycogen content, in individual blood and bone marrow neutrophil leucocytes from seven cases of chronic myelocytic leukaemia (CML) and five non‐leukaemic subjects. Neutrophils from untreated CML contained low PASMa amounts as compared to neutrophils from normal subjects, while normal amounts of PASMa were found in neutrophils from CML patients in busulfan‐induced remission. Blood and bone marrow neutrophils in the same individual subject contained similar PASMa amounts whether sampled from a normal person, untreated CML (low PASMa level), polycythaemia vera (high PASMa level), or CML in complete remission (normal PASMa level). Treatment of CML with busulfan initiated earlier normalization of PASMa amounts in bone marrow neutrophils than in blood neutrophils, although similar normal PASMa amounts were found in both blood and bone marrow neutrophils in complete remission. The pattern of normalization showed that the busulfan‐induced normalization of PASMa content in CML‐neutrophils was due to action on precursor‐myeloid cells and not to direct action on mature neutrophils. The main abnormalities in two sisters with congenital macrothrombocytic thrombopathia who were previously described (Kanska et al. 1963) consisted of enormous platelets, considerable decrease of platelet factor 3 in intact and homogenized platelets, decrease of thromboplastic activity of erythrocytes and periodic thrombocytopenia. Furthermore platelet factor 3 did not become available following platelet aggregation by ADP and thrombin. Ultrastructural and enzymologic examinations revealed decrease of glycogen of some enzymes of citric acid cycle and of ATP in patient platelets. The abnormality of microtubules was observed. It has been postulated that the nature of macrothrombocytic thrombopathia may be connected with the abnormality of lipoproteins which form structural part of platelet and erythrocyte membranes.

The specific indication of intravascular fibrinolysis is the demonstration of fibrinogenfibrin degradation products.To evaluate the effect on fibrinolysis 48 patients were treated with clofibrate, metformin, tolazamide or testosterone for periods of one to fourteen weeks. Blood fibrinolytic activity was assessed before, during and after treatment by measurements of euglobulin clot lysis time, plasminogen and fibrinogen, and immunoassays of fibrinogen‐fibrin degradation products.17 normal persons, 23 patients with atherosclerotic vascular disease, and 11 patients with other diseases were exposed to physical exertion or injections of nicotinic acid. Fibrinolytic activity was assessed as above, before and after stimulation.Acceleration of euglobulin clot lysis was found in only 2 of 11 patients treated with metformin, but in 47 persons exposed to short‐term stimulation. Twelve patients and their families were studied to obtain information of a possible relationship between certain HL‐A genotypes and acute leukaemia in children. The frequency of the individual HL‐A antigens among the patients did not deviate significantly from the frequency of the antigens among healthy persons. The HL‐A genotypes and haplotypes of the patients were established through family analysis, but in the present material no certain genotype or haplotype was found that seems to be related to acute leukaemia. Eighteen patients were studied, selected only on the basis of a persistently elevated cold haemagglutinin titre.The concentration of γM‐globulin in the initial serum sample varied from 0.7–24.5 mg./ml. In all of 15 patients studied in detail, the cold haemagglutinin behaved as a monoclonal γM‐globulin.In two patients, a homogeneous γ‐globulin remained in the serum after absorption with red cell stromata in the cold. During routine examination of the erythrocyte sedimentation rate in blood donors an unusual sedimentation pattern was found in two healthy males. The sedimentation curve showed a very pronounced S‐shaped course, the lag phase varying between 15 and 60 minutes followed by a period of very rapid sedimentation. The erythropoietic effect of testosterone cyclopentylpropionate (TCP) was studied in normal, plethoric, nephreetomized and ureter‐ligated rats. The results of this study support the contention that TCP exerts its erythropoietic effect though the kidney by increasing REF production; thereby potentiating this organ to generate greater amounts of erythropoietin (EP) in response to normal and cobalt‐stimulated erythropoietic challenges. From the data, it is further concluded that TCP neither exerts a direct EP‐like effect on haemopoietic tissues nor potentiates the effect of preexisting endogenous EP.
