Thin layer chromatography was used to identify hexoses, pentoses and methyl‐pentoses, disaccharides, hexuronic acids and hexosamines in acid hydorlysates of human, bovine, pig and horse erythrocyte stroma. The presence of galactose, glucose, mannose, fucose, glucosamine, and galactosamine was found in all of these stroma samples, arabinose was present in human and bovine stroma, and glucuronic acid in bovine and pig stroma. In the acid hydrolysate of human stroma there was one fraction that had about the same Rf‐value as lactose or isomalt

The double‐labelling, two‐emulsion autoradiographic technique was used to study DNA synthesis time and related nuclear sub‐cycle durations of erythroid cell precursors in three week‐old rats. As employed in this short‐term investigation, the double‐labelling technique permitted direct estimates of DNA synthesis time to be made independently of labelling indices from the flow of two populations of labelled cells through the S‐phase of the nuclear cycle. The application of a second autoradiographic emulsion over single‐emulsion preparations made possible a more objective distinction between the two labels than can be done with single emulsion preparations. The following estimates were obtained from counts made on nine rats and a total of 45,000 cells: DNA synthesis time, 7.0 ± 0.5 hours; mitotic time (not including early prophase), 20 ± 3 minutes; average turnover time,

Autoradiographs prepared from histological sections of thymus were studied at different intervals after a single intraperitoneal injection of tritiated thymidine into male CBA mice. The initial uptake was located mainly in the cortex, especially in the periphery, to a lesser extent in the medulla. Immediate incorporation was seen in reticular cells and in large and medium‐sized lymphocytes, not in small lymphocytes. As time elapsed more cells became labelled, and a migration of labelled cells from the cortex to the medulla took place. Simultaneously an increasing number of small thymocytes acquired the label. The pattern of initial uptake and spread of label with time was similar for the thymus of mice aged 3 weeks, 7 weeks and one year.Autoradiographs of smears of whole thymus revealed that 13 per cent of the cells were labelled one hour after the administration of3H‐thymidine. The labelling percentage increased with time and reached a maximum of 37 per cent, 3 days after the injection. After that time a fall in labelling percentage was observed with a final disappearance of labelled cells from the thymus about the 14th day.Calculations on the basis of mitotic counts with and without colchicine pre‐treatment gave an average mitotic rate in thymic cortex of 3.7 per cent per hour, which renders an average renewal time of 27 hours by cell division for thymic cortex ‐ indepent of age.It is concluded that a large cell production takes place in the thymus, especially in the outer cortex, the end result being the production of thymocytes, mainly small ones, which migrate to the medulla from where they di

Among 3,000 Norwegian individuals two haemoglobin variants were found. One was a Haemoglobin D Punjab. The other was a new variant, Haemoglobin Sogn, in which leucine is replaced by arginine at the internally sited residue 14 of the β chain. LDH isoenzymes in thrombocytes from 41 healthy subjects and 85 patients suffering from various diseases combined with changes in platelet function were studied. The isoenzymes were separated by disc acrylamide electrophoresis. In normal thrombocytes LDH 3 constituted around 41 per cent, LDH 2 around 32 per cent, LDH 1 around 12 per cent and LDH 4 around 14 per cent of the total amount, whereas only very little LDH 5 was found.The results have been evaluated statistically. No differences were found between younger and older healthy persons.In patients with hepatic cirrhosis and polycythaemia vera an increase in ratio between H and M units (increase in T‐values) was demonstrated. The relationship between the dense bodies found in the exterior coat of negatively stained platelets, and the dense bodies apparent in the hyaloplasm of sectioned cells has been examined. Dense bodies (S.D.B.) similar to those reported by Bull were readily found in whole mount preparations. Some extended beyond the platelet surface membrane and possessed long, whip‐like tails. Examination of thin sections of glutaraldehyde‐osmium fixed, plastic embedded platelets revealed dense bodies (H.D.B.) in the hyaloplasm, but not exterior to the cell surfaces. The H.D.B. occasionally had long, whip‐like tails similar to S.D.B. in the negatively stained preparations. H.D.B. with narrow extensions were encountered frequently in platelets aggregated with ADP or thrombin. While most granules were centrally oriented in altered cells, H.D.B. with long extensions were found in the peripheral hyaloplasm close to the cell surface. Examples were observed in which membranes enclosing H.D.B. appeared continuous with the platelet surface. These results suggest that S.D.B. found in the coat exterior to the plasma membrane of negatively stained platelets, and H.D.B. present inside plastic embedded cells are identical organelles. The S.D.B. may be accumulations of phospholipid destined for release to the plasma as was previously suggested, but they do not appear to be derived from the cell mem

摘要:
Two cases of jejunal diverticulosis associated with intrinsic factor deficiency are reported. One case had megaloblastic anaemia and full correction of58Co vitamin B12absorption was achieved by broad spectrum antibiotic therapy alone. The second case had intrinsic factor deficiency and full correction of58Co vitamin B12absorption was achieved by the addition of oral intrinsic factor alone. The variable contribution made to the development of vitamin B12deficiency in jejunal diverticulosis by intrinsic factor deficiency and bacterial colonisation of the small intestine is discussed.

On the assumption that, under certain conditions, acid phosphatases may serve as a biochemical marker for the physico‐chemical state of lipoprotein of platelet lysosymes, the changes in the thromboplastic activity of platelets were studied simultaneously with those in acid phosphatases.The quantitative relationship between the platelet‐factor‐3‐like activity of the serum and the acid phosphatases released from the platelets into the serum during coagulation of native platelet‐rich plasma is pointed out.A study of the changes of acid phosphatase activity during kaolin clotting time showed deep ultrastructural changes in platelets.The results obtained indicate that it is product I and not thrombin alone which increases the activity of acid phosphatases in suspension of isolated platelets.The relationship of platelet lysosomes to the platelet‐factor‐3‐activi
