A method for quantitative evaluation of blood platelet pseudopod formation is presented. Pseudopod formation was studied in whole blood incubated for 0, 5, 10, 20 and 40 minutes at + 4°, + 15° or + 37° C. Samples were anticoagulated with EDTA or sodium citrate, or incubated without anticoagulant. In instantly fixed blood, platelets showed a pseudopod index of about 0.5. In samples incubated at + 4° C., the pseudopod index rose to about 3 after 20 minutes. After 40 minutes there was no further increase. At + 37° C. the pseudopod index did not increase during the first 20 minutes of incubation, but after 40 minutes it had risen to levels almost similar to those of the + 4° samples.EDTA and sodium citrate exerted no influence upon pseudopod formation at any temperature level tested. Erythropoietin levels in both serum and urine have been measured in seven patients with various haematologic disorders and three normal male subjects before and after the short term administration of androgen. Increase in erythropoietin was seen in the urine of the normal subjects. Increase was also seen either in the urine or serum of all 7 patients, 5 of whom had already elevated erythropoietin titers.

Leukocytes from human blood were separated from erythrocytes by accelerated sedimentation of the red cells. Lymphocytes were isolated by selective absorption of the polymorphonuclear leukocytes on acryl fiber columns. Generally more than 95 per cent, and often 100 per cent, of the white cells recovered were lymphocytes mixed with, as a mean, double the amount of red cells.

The influence of heparin on the reaction between purified human thrombin and fibrinogen was studied. The appearance of N‐terminal glycine, which reflects conversion of fibrinogen to fibrin, was measured. The inhibition of the thrombin‐fibrinogen reaction was studied by recording the appearance of N‐terminal glycine. The reaction was progressively inhibited by heparin and purified human cofactor. No immediate effect could be demonstrated Thrombin activity was not restored following neutralization of heparin with protamine. The inhibition was therefore apparently due to ‘progressive’ inactivation of thrombin. The thrombin half life was approximately inversely proportional to the cofactor concentration. As compared to inactivation by cofactor alone, heparin (0.1 I.U./ml.) increased the inactivation rate about 20 times. Consequently, an inhibition could be demonstrated at much higher thrombin concentrations than with cofactor alone. α1‐glucoprotein counteracted the inhibition, but calcium had no effect. The relationship between the type and quantity of dietary fat and the incidence of adenosine diphosphate induced platelet thrombosis was studied in rats fed a 10 per cent and 40 per cent saturated fat diet, a 40 per cent fat diet including 8 per cent cottonseed oil and a 40 per cent fat diet including 8 per cent linseed oil. The diets were given for 24 hours or 5 to 6 weeks. Platelet thrombosis was induced in pulmonary capillaries, arterioles, and small arteries by the intravenous injection of ADP. No significant difference in the incidence of thrombosis was observed following a 24 hour feeding period. After a 5 to 6 weeks feeding period, animals given a 40 per cent saturated fat diet had a higher incidence of thrombosis than animals given a 10 per cent saturated fat diet. When the 40 per cent fat diet included cottonseed oil, the incidence of thrombosis due to a low dose of ADP was reduced. When linseed oil was included in the diet, the incidence of thrombosis was reduced when both a low and a high dose of ADP was given. Significant changes occurred in the fatty acid pattern of the plasma and platelet phospholipids in animals fed the diets for 5 to 6 weeks. The diets induced small changes in the quantity of the main plasma lipid fractions.

Aggregation of human platelets by addition of purified bovine platelet fibrinogen is described. Bovine plasma fibrinogen showed the same but much weaker effect. Human fibrinogen produced no aggregation. No absolute requirement for divalent cations or plasma proteins could be demonstrated. The aggregation of washed platelets appeared monophasic whereas in platelet‐rich plasma it was usually biphasic. The use of inhibitors of ADP‐induced platelet aggregation, inhibition of intracellular ATP‐production, enzyme‐catalyzed removal of ADP, and direct determinations of ADP in the medium showed the second phase to be mediated by ADP released from the platelets whereas the first phase was nearly independant of ADP. While the ability of platelet fibrinogen to aggregate platelets and to clot with thrombin were otherwise intimately interconnected, some aggregation activity remained after heat‐denaturation of the platelet f

Two fatal cases of severe anaemia in infancy with ellpitocytosis in a Danish family are described. The severe progression of the disease is due to homozygosity for the gene for elliptocytosis.
