Patients treated with phenytoin for periods of several months or years have slightly, but significantly, lower whole blood and red cell folate values than comparable controls. This holds true irrespective of whether folate is determined microbiologically with Lactobacillus casei or by a radioligand method using the bovine milk binder. Haematological parameters including Hb, MCH, MCV and red blood cell counts did not show any alterations and no patient developed any signs of anaemia. Antithrombin III (At‐III) concentration and amidolytic assays reflecting functions of the At‐III molecule were studied in members of a family with a strong tendency to thrombosis. Antifactor Xa and heparin cofactor activity closely paralleled the At‐III concentration. In all individuals with a history of thrombosis At‐III concentration was about 50% of normal. 摘要:
Neuraminidase treated RBC were used to detect incomplete unbound anti RBC antibodies in the plasma of patients with immune diseases. 80% (48 out of 60) of the examined patients gave positive results with this method while all the 50 normal normal controls were negative.The indirect antiglobulin test was negative in all the patients. In the direct antiglobulin test only two RBC samples, whether treated with neuraminidase or not, gave positive results.The present study indicates that the assay of plasma on neuraminidase treated RBC may serve as a sensitive method for the detection of the presence of anti RBC antibodies in various immunological diseases.

The effect of polymorphonuclear granulocytes (PMN) on colony stimulating activity (CSA) was studied in double layer cultures of human Ficoll Isopaque*** separated white blood cells (mononuclear cells = MNC). Previously published data have been confirmed that granulocytes are able to enhance or inhibit MNC derived CSA. Further analysis of the mode of action of PMN in vitro indicates that the enhancing activity ascribed to granulocytes coincides with low CSA in MNC basal layers. In contrast, in cultures with high levels of CSA as provided by lysed red blood cell enhancement rather low concentrations of PMN are sufficient to induce inhibition of colony growth. A very similar effect to that achieved with basal layer derived CSA could be obtained with conditioned media of PMN and MNC short term liquid cultures. The data indicate, that enhancement and inhibition of colony growth reflect a specific reactivity of granulocytes (PMN) to a given CSA level in the cultures. These findings are discussed in terms of a speculative role of PMN in a negative feed back control mechanism regulating granulopoiesis in vivo.

Myeloperoxidase, restricted to primary granules, and lactoferrin, restricted to secondary granules, were determined in plasma and neutrophils of peripheral blood in chronic granulocytic leukaemia (CGL). Plasma myeloperoxidase was increased 2–3 times while plasma lactoferrin increased 2–8 times. This discrepancy indicates different modes of release or elimination. A correlation was found between the leucocyte count and plasma myeloperoxidase or lactoferrin. A correlation was also found between cellular and plasma levels of lactoferrin but not for myeloperoxidase indicating the source for plasma lactoferrin to be circulating leucocytes, which may not be the case for plasma myeloperoxidase. Decreased neutrophil lactoferrin was found in 71% of the CGL cases while myeloperoxidase was decreased in 18 %. Serial studies on individual CGL subjects showed low cellular lactoferrin during phases with rapidly expanding leucocytosis indicating defective maturation of neutrophils or abnormal release because of prolonged intravascular life

Changes in numbers of peripheral blood lymphocytes from healthy individuals were calculated from samples collected before and after parenteral administration of adrenaline, insulin and hydrocortisone, respectively. A marked increase in circulating lymphocytes was noted in response to adrenaline and insulin. However, subpopulation analysis showed a decrease in the proportion of T‐lymphocytes, estimated as cells forming rosettes with sheep red blood cells after incubation in the cold and a corresponding increase in proportion of lymphocytes having receptors for C3 (non‐T lymphocytes). In contrast, lymphocyte numbers were unaffected by hydrocortisone. A double diffusion chamber technique (DDC) has been established. The bone marrow cells (BMC) were cultured in the peritoneal cavity of mice in DDC consisting of 2 compartments separated from one another by a Millipore membrane. One chamber half contained the mouse bone marrow target cells, and the other half (regulator compartment) either medium (control), spleen cells or BMC. In the controls the BMC proliferated rapidly from day 2, and the cell yield on day 7 was reduced by only 20% when compared with single diffusion chambers. Diffusible factors from spleen cells stimulated the growth of CFU‐S and CFU‐C in the bone marrow, and increased the number of granulocytes and macrophages harvested in 7 day cultures. Peripheral blood lymphocytes and their in vitro reactivity have been recorded prior to treatment in 18 patients with Hodgkin's disease, 11 with lymphosarcoma, 13 with reticulosarcoma, 20 with various solid tumors and 37 normal control persons. The mean total numbers of lymphocytes, those of T lymphocytes, and the mean reactivities to PHA and ConA were reduced in all groups except lymphosarcoma, although with varying degrees of statistical significance. The percentages of T and B lymphocytes appeared to be normal in all groups, but the ranges of values were somewhat greater than among the normal controls. The mean total numbers of B lymphocytes were normal in all patient groups. All reductions seemed to be more pronounced in patients with disseminated than in those with localized disease, but none of these differences was statistically significant. All patient groups appeared to have reduced reactivity in MLC, while the ability to stimulate control lymphocytes was nearly normal.The results fail to indicate any in vitro immunological abberation specific to Hodgkin's disease. It seems that human malignant, neoplastic diseases are associated with a relatively selective reduction of the total numbers and reactivity of blood T lymphocytes. Various explanations of the reactivity impairment are proposed. The pathogenesis of the reduction of the total number of blood T lymphocytes remains obscure.

Blood lymphocyte DNA synthesis, as expressed by 24‐h uptake of3H‐thymidine in culture of mononuclear peripheral blood leucocytes has been recorded prior to treatment in 18 patients with Hodgkin's disease, 11 with non‐leukaemic lymphosarcoma, 13 with reticulosarcoma and 20 with various solid tumors, and compared to 37 normal control subjects. The3H‐thymidine uptake was significantly increased in all the patient groups, but no significant difference existed between them. Increased uptake of3H‐thymidine did not appear to correlate with the degree of dissemination in any of the patient groups or with the presence or absence of general symptoms in malignant lymphoma. It is concluded that increased DNA synthesis in peripheral blood lymphocytes is a feature not limited to Hodgkin's disease, but found in non‐Hodgkin malignant lymphomas and other malignancies to about the
