Whole blood glutathione peroxidase was found significantly increased relative to controls in a group of 21 black patients with sickle cell anaemia. One control group consisted of 15 normal black subjects. A second control group, consisting of 21 black patients with various abnormal haemoglobins including a‐thalassaemia, also showed a tendency to enhanced glutathione peroxidase activity, confirming previous reports that elevated glutathione peroxidase levels are secondary to a variety of haemolytic conditions, rather than typical of sickle cell anaemi

The bone marrow was morphologically analysed in 25 patients receiving long‐term azathioprine therapy for chronic renal diseases or after a renal transplantation. There were no significant changes within the granulopoietic or lymphopoietic part of bone marrow even after 10 years of treatment. However, there was a significant ‘shift to the left’ within erythropoiesis as well as greater frequencies of megaloblasts and sideroblasts. A significant increase of ‘erythroblastic islands’ may indicate an increased death rate of defective erythro‐blasts. Rosettes formed between human lymphocytes and unsensitized mouse erythrocytes not only detect a subset of normal B‐lymphocytes, but furthermore seem to be of considerable importance in the differential diagnosis of various B cell malignancies. From a series of experiments aimed at optimizing this rosette test, we conclude: (1) Erythrocytes from different mouse strains vary in their affinity for lymphocytes; (2) Experimental conditions are less strict when testing chronic lymphatic leukaemia lymphocytes than normal lymphocytes; (3) Treatment with the sulphydryl compound aminoethyl‐isothiouronium bromide, which enhances sheep erythrocyte rosette formation, has an opposite effect on mouse rosettes, and (4) Neuraminidase treatment of the lymphocytes is not necessary when optimal experimental conditions are u

30 children with acquired aplastic anaemia are presented. The mortality rate was 60%. The period of observation exceeded 8 years for 10 patients. The importance of long control and the timely conclusion of therapy are emphasized. Prognostic criteria and alternatives to steroid therapy are discussed.

Bone marrow granulocyte colony forming cells (CFU‐C) were measured in 10 HL'A compatible sibling donor‐recipient pairs. The regeneration of granulocytes and CFU‐C compartments were also studied in order to evaluate haemopoietic recovery. The number of nucleated bone marrow cells in the donation was 23 ±4 times 109cells, which recipients received (3.5 ±0.4) × 108nucleated cells/kg and (1.19 ±0.32) × 105CFU‐C/kg. This produced a bone marrow reconstitution of (67 ±26) × 105CFU‐C/kg by day 30. 40 patients with acute leukaemia and severe granulocytopenia were treated with granulocyte transfusions for bacteriologically documented or clinically suspected septicaemia. The patients received an average of 5 transfusions each with 2.1 times 1010granulocytes per m2body surface area per day. 26 patients showed clinical benefit from transfusion therapy as documented by the course of blood cultures, local lesions and fever. Only patients with regeneration of granulopoiesis had definite benefit from granulocyte transfusions. All other patients died ultimately from septicaemia.

Spontaneous (3H)‐TdR incorporation was studied in cell suspensions from 64 patients with monoclonal B‐cell neoplasias. Among various incubation periods a long‐term (20 h) assay with (3H)‐TdR had the greatest discriminatory power versus non‐neoplastic lymph node cell suspensions. The (3H)‐TdR uptake correlated positively with cell volume, nuclear volume, and cells in S + G2+ M phase of the cell cycle. A high degree of heterogeneity with regard to (3H)‐TdR incorporation was found within several histological groups of the Kiel classification, especially in low‐grade malignant lymphomas of centroblastic/centrocytic origin and in the lymphoplasmacytoid groups. In highly malignant lymphomas (3H)‐TdR uptake was statistically, significantly higher than in lymphomas of low‐grade malignancy. Patients with localized disease (stages I and II) and those with ‘B’‐symptoms showed increased incorporation of radioactive thymidine as compared to patients with disseminated disease (stage IV). Blood was obtained from 63 consecutive patients within a 24 h period after the admission to a coronary care unit for the determination of plasma platelet factor 4 (PF‐4) concentration. 28 of the subjects proved to have an acute myocardial infarction (MI), 24 had evidence of ischaemic heart disease (IHD) but no MI, and the remaining 11 patients had no signs of IHD. 40 healthy subjects served as controls. The mean PF‐4 value in the MI group was 10.5 ±0.8 ng/ml. The corresponding values for patients with and without IHD were 8.7 ±0.6 and 8.3 ±0.6 ng/ml, respectively. Chromosome banding studies of the bone marrow were performed in 35 adult (>14 years) patients with acute lymphoblastic leukaemia (ALL). Surface marker analysis was done in 24 of these and revealed 4 B‐ALL, 5 T‐ALL and 15 non‐T non‐B ALL. Most patients were studied at diagnosis before any treatment. A clonal karyotypic abnormality was found in 16 patients (46 %) initially. A Philadelphia chromosome was found in 3 patients, all belonging to the non‐T non‐B group. 5 patients, who all had blast cells with morphologic characteristics of Burkitt type L3, were found to have bone marrow cells with a 14q+ marker chromosome.
