The effect of VK 744 (an analogue of dipyridamole) on the function of human blood platelets was studied. VK 744 inhibits collagen aggregation at a concentration of 25 μM. Preincubation at 37° C enhances the effect. ADP aggregation is inhibited at 50 μM. Adrenaline aggregation is likewise inhibited, and becomes reversible at 50 μM. VK 744 causes active disaggregation when added during ADP aggregation. The glass‐bead column retention and clot retraction are not inhibited until a concentration of 1 mM is attained. The glass adhesiveness according to Breddin is inhibited by 10 μM.The availability of platelet factor 3 as well as serotonin uptake are inhibited at the ED 50 of 6 μM and 0.2 mM, respectively. Comparison with RA 233 disclosed that two separate pharmacological receptors are involved. A study of serotonin release in collagen aggregation revealed that VK 744 is a primary inhibitor of aggregation. A set of conditions is defined for freeze‐storage of lymphocytes circulating in chronic lymphocytic leukaemia (CLL‐lymphocytes) and in healthy individuals (normal lymphocytes). Isolated cells are frozen in culture medium containing 15 % serum and 8.3 % (v/v) dimethyl sulphoxide (DMSO) at a post‐freezing cooling rate in the range 1.8‐–3.4° per min to −40° or below followed by immersion in liquid nitrogen, and thawed rapidly (ca.400° per min) at 37°. This procedure induced consistently low mortalities (2–7%) in normal and CLL populations, uninfluenced by freeze‐storage lasting up to 57 weeks, and was accompanied by large‐scale destruction (67–100%, average 95 %) of residual isolated red cells. The lymphocytes classified as surviving cells by cytologic analysis could display partial loss of initial capacity for respiration but capacity for survival in short‐term culture was retained. Brief reference is made to unchanged ultrastructural appearance of surviving cells. No pronounced differences were observed in the capacity of normal populations to transform under the influence of phytohaemagglutinin (PHA) as a result of freezing and thawing, but tests for altered proliferative capacity of transformed cells were hampered by culture conditions proving sub‐optimal. A patient with widespread IgD‐myelomatosis is described. The serum electrophoretic pattern exhibited at discovery of the disease two M‐components: one of IgD‐lambda type migrating in the γ1‐region and one of Bence Jones protein lambda type in the β1‐region. A Bence Jones protein of lambda type was also detected in the urine. During treatment with cyclophosphamide the IgD‐lambda‐ and serum Bence Jones lambda‐proteins disappeared and the urine Bence Jones lambda protein decreased. After 8 months' treatment another M‐component was recognized in the slowest γ‐region. This M‐component was IgG, subclass IgG1 of kappa type. No signs of the IgGl component were seen when the first serum samples were retested with a specific antiserum raised against the IgG‐kappa component. In the following samples also the IgG‐kappa component decreased but traces were still demonstrable in all late samples. The patient improved during treatment. In 6 patients with diffuse intravascular coagulation (DIC) (3 of them on long term anticoagulant treatment with warfarin sodium), low Thrombotest values and a discrepancy between Thrombotest and Normotest was found. During heparin treatment, the Thrombotest values rose and the Thrombotest‐Normotest discrepancy decreased. These observations indicate that Thrombotest may be a sensitive indicator of consumption coagulopathy. A rise in Thrombotest during heparin treatment probably substantiates this concep

3 patients with myelosclerosis have subsequently developed paroxysmal nocturnal haemoglobinuria, with positive in‐vitro lysis tests (acidified‐serum lysis, cold‐antibody lysis and sucrose lysis), characteristic of PNH. These cases provide further evidence of a relationship between PNH and myeloproliferative disorders; they support the proposition that PNH is an acquired defect of erythropoiesis due to an abnormal clone. In two of the patients PNH has been severe with marked intravascular haemolysis, so that in these cases the clinical course and management of the original disease have been influenced by the PNH. Digestion of stabilized and non‐stabilized fibrin in clots made from isolated fibrinogen, contaminated by fibrin stabilizing factor (FSF), and plasma is evaluated qualitatively by Immunoelectrophoresis and quantitatively by haemagglutination inhibition immuno assays (HIIA). Non‐stabilized systems are obtained by means of SH‐ and transamidase inhibitors and, in plasma systems, also by genuine FSF‐decient plasma. Stabilized and non‐stabilized fibrin in fibrin clots is easily degradated by plasmin (porcine) to the final D and E antigenic fragments, which are non‐reactive in HIIA. Stabilized fibrin in plasma clots is rather resistant to activator‐induced lysis, as antigenic intermediates, reactive in HIIA, remain even after sustained activator (+ plas‐minogen)‐induced lysis. In testicular biopsies two types of human erythrocytes were discovered as a result of different dye uptake after staining with iron alun, haematoxylin, picric acid, acid Fuchsin, Ponceau de Xylidine and Fast Green FCF. The two types were distinctly different, one type stained deep purple, the other pale bluishgrey or brownish. The distribution of the purple erythrocytes in venules, arterioles and capillaries was random. Two types of erythrocytes were also demonstrated in clots. In these it was not possible to correlate the dye uptake either with the O2or the CO content of the erythrocytes or with pH of the blood or with the age of erythrocytes. The differentiation of the erythrocytes in two types was caused by a combined effect of acid Fuchsin, Ponceau de Xylidine and Fast Green FCF which together presumably form a new complex dye in the tissue.

Injection into rabbits of heparin or one of the heparin‐neutralizing substances protamine and polybrene induced a rapid, transient fall in the number of platelets, without affecting the fibrinogen levels. The lowest platelet counts in the different animals ranged from 4 % to 52 % of the preinjection values. A few seconds after the injection, platelet aggregates were observed in the blood samples. Later the aggregates disappeared, but smaller aggregates reappeared when the platelet count began to rise. Marked pulmonary symptoms in many animals indicated trapping of platelet aggregates in the pulmonary vascular bed. Spleens from healthy adult cats and dogs were perfused with oxygenated Ringer solution. The majority of the erythrocytes were quickly washed out but approximately 10 % were released very slowly with a half‐life of 1 hr (slow compartment). After perfusion the morphological characteristics of the cells in this slow compartment can be studied using histological sections prepared with blood stains. Under the light microscope a variety of immature and abnormal erythrocytes was seen. The immature cells were identified as rubricytes and reticulocytes, but no proerythroblasts were found. The pictures show rubricytes in various stages of maturation and denucleation evidently occurred by extrusion of either an intact or a fragmented nucleus.
