Peripheral leucocytes from 10 patients with chronic myelogenous leukaemia (CML) were cultured in liquid medium and observed morphologically for 6 to 10 weeks. All cultures showed an increased proportion of mature basophils at about 3 weeks and most cultures consisted predominantly of mature basophils at about 5 weeks. The results indicate preferential basophilic differentiation of CML leucocytes in vitro and are consistent with basophilia frequently observed in CML patients.

Serum ferritin and erythrocyte 2,3‐DPG levels were followed in 3 healthy males who were phlebotomized to iron depletion and a moderate anaemia. In 2 of the subjects, the expected rise in DPG levels was seen but not in the third, in spite of a Hb concentration of 95 g/1. Serum ferritin levels were found to reflect changes in iron stores, and subnormal serum ferritin indicated depleted iron stores. However, there was no correlation between pre‐phlebotomy ferritin levels and calculated iron stores. The conclusion is that no fixed ratio can be established between serum ferritin and iron stores. 10 patients with anorexia nervosa were compared with controls with normal weight, regarding their peripheral blood polymorphonuclear (PMN) granulocyte reactions. The anorexia patients showed a statistically significant decrease in PMN bactericidal capacity and PMN adherence. The mean chemotaxis did not differ, but in two of the anorexia patients chemotaxis was almost absent. The intracellular activity of alkaline phosphatase was below the reference values in 5 of the 6 patients in whom it was investigated. ‐ It is concluded that changes in granulocyte function may be noted in anorexia nervosa, but their clinical significance is uncertain, as no patients had recurrent or severe infectious disease

Starch gel electrophoresis was used to screen for intravascular haemolysis in 15 patients with clinical disorders known to be characterized by intravascular haemolysis. Decreased serum haptoglobins, increased free plasma haemoglobin and the presence of methaemalbumin was documented in all 15 patients. It is suggested that starch gel electrophoresis and staining with haem specific stain is a simple and reliable method for confirming intravascular haemolysis.

In 34 patients with aregenerative anaemia and hypercellular bone marrow, a condition earlier suggested to be of a preleukaemic nature, erythroblast thymidine labelling indices (LI) were significantly decreased, as would be expected in preleukaemia. The59Fe erythrocyte incorporation after 2 weeks was generally low. The59Fe plasma clearance was fast in 22 of 31 patients, normal in 5 and moderately prolonged in 4.These 4 patients had a low relative number of erythroblasts in the marrow. A significant relationship between the marrow erythroblast number and the59Fe plasma clearance could be demonstrated.Cases with an increased number of myeloblasts in the marrow did not differ significantly from the rest in LI and iron incorporation.

The number of Fc and C3 receptors on normal B lymphocytes, isolated from blood, was assessed semiquantitatively by a new method in which the marker cells (pre‐treated sheep erythrocytes) for Fc and C3 receptors are labelled by99Tc and51Cr respectively. The mean number of bound marker cells per rosette was assessed simultaneously by measurements of radioactivity.Control studies showed that a measuring system using 1/4 agglutinating titre of antibody on the marker cells is particularly advantageous: (1) Fc and C3 receptors are not demonstrable on T lymphocytes; thus, B lymphocyte fractions with Fc and/or C3 receptors are well‐defined. (2) The binding affinity of the marker cells for normal lymphocytes is so weak that the number of bound marker cells per rosette rarely reaches the maximum which was calculated and measured as about 30 marker cells per rosette; thus, the measuring system is sensitive to changes in the number of receptors. (3) By binding of marker cells to lymphocytes bearing Fc as well as C3 receptors it has been demonstrated that interference phenomena between the two marker cells are not operative.Studies of blood lymphocytes from normal human subjects showed B lymphocytes carrying either Fc or C3 receptors as well as a fraction of B lymphocytes having both Fc and C3 receptors. The latter make up about 6 % of the lymphocyte mass. The serum of a patient with a non‐aggressive plasma cell proliferative disorder contained two monoclonal immunoglobulins: IgG3λ in moderate concentration and having cryoglobulin features, and IgAκ in low concentration and without cryo‐precipitability.The patient's serum had low complement concentration and C3 was partly converted into split products in vivo.
