Twelve patients with a history of thrombo‐embolic disease for more than 4 days were given a four‐fold titrated initial dose of streptokinase during 2 hours. The fibrinolytic response was investigated and the split products of fibrinogen and/or fibrin studied with immunological technique. 摘要:
The influence of an increased haemolysis on the urinary iron excretion after the administration of desferrioxamine (DF) was studied in a series of experiments. An increased haemolysis was induced by transfusing stored red cells, by injecting nicotinic acid or by infusing antibody‐coated red cells. The results indicate that under normal conditions, the urinary iron excreted with DF does not originate from destroyed red cells.Iron absorbed from the intestines is no probable source of the iron excreted in the urine with DF, as judged from a study in which the iron excretion was compared during periods with iron‐rich and iron‐free diets.The results of our present and previous studies indicate that stored iron is the probable main source of the iron excreted with DF in the

Factors influencing the rate of glucose conversion in red blood cells have been investigated using haemolysates and a purified hexokinase system.The KM‐values for glucose and ATP indicate that the glucose concentrations usually occurring in red blood cells are optimal, whereas variations around the physiological ATP‐concentration will strongly influence the conversion rate.Glucose 6‐phosphate is a potent competitive inhibitor of hexokinase with respect to ATP. Other phosphocompounds tested, fructose 6‐phosphate, fructose 1,6‐diphosphate and 2,3‐diphosphoglycerate, have no inhibitory effect. Inorganic phosphate decreases the inhibition caused by glucose 6‐phosphate. The mechanism for this action is not clear; an interpretation of the data obtained is suggested, however.In intact red blood cells, inosine blocks the phosphorylation of glucosee. By density gradient ultracentrifugation and preparative zone electrophoresis, the normal serum factor causing reversible agglutination of trypsinised red cells behaved as a γM‐globulin. After splitting of disulphide bonds by 2‐mercapto‐aethanol, the agglutinating activity was lost. A survey is presented of 4168 sera containing irregular blood‐group antibodies. The antibodies were found during 309,335 antibody screenings and 135,402 cross matchings of blood samples from about 200,000 individuals.About 50 per cent of the antibodies were anti‐D, sometimes together with other antibodies in Rh negatives (D neg), 8 per cent were Rh antibodies in Rh positives (D pos) and many of these antibodies were detectable only by a two‐stage enzyme technique and not by AHG‐technique.Almost 15 per cent of the antibodies belonged to the Lewis‐system. There was a significantly increased number in females of the fertile age, and there was a significant decrease in the number of group O individuals among the Lewis‐immunized persons. The low number of group O individuals indicate a hitherto unrecognized relationship between the ABO‐ and Lewis‐systems.Altogether 2.1 per cent of the individuals described possessed one or more irregular antibodies. The importance is discussed of avoiding complications from these antibodies during blood transfusions. It is concluded that a reliable cross‐match should include a two‐stage enzyme technique as well as the usual saline and AHG tests. The effectiveness of galactose as a substrate for methaemoglobin reduction in human red cells has been investigated. The rate of methaemoglobin reduction has been found to depend on the concentration of galactose up to levels of 0.5 M. Methylene blue, which markedly accelerated methaemoglobin reduction when glucose was a substrate, only minimally accelerated methaemoglobin reduction with a galactose substrate. Pre‐incubation with high concentrations of galactose did not inhibit methaemoglobin reduction in the presence of glucose and methylene blue. No inhibition of G‐6‐PD activity was observed with very high concentrations of Gal‐1‐P, glucose‐1‐P, or galactose, even at low concentrations of G‐6‐P. Several possibilities are presented to explain the lack of stimulation of methaemoglobin reduction by methylene blue when galactose is the substrate: 1) inhibition of methylene blue‐stimulated methaemoglobin reduction by galactose or its metabolites; 2) inability of the red cell to metabolize very low concentrations of glucose‐6‐phosphate by way of the hexose monophosphate pathway; and 3) that β‐glucose‐6‐phosphate is not an important product of galactose metabolism. In the latter case, which appears the most likely, a previously undescribed pathway of carbohydrate metabolism must exist in the human erythrocyte.
