The effect of Vibrio cholerae neuraminidase (VCN) treatment of tumor cells on lymphoblastic response was studied using the assay of mixed lymphocyte-tumor cell reaction in 24 patients with gastric carcinoma. Lymphoblastic responses as indicated by stimulation index were significantly augmented in cultures with autologous tumor cells pretreated with mitomycin (MMC) and VCN as compared to those pretreated with MMC alone when cultured in the presence of normal plasma (p < 0.05). In autologous plasma cultures, however, the mean stimulation index in MMC-VCN-treated cell cultures was only slightly higher than that in MMC-treated cell cultures. Accentuation of the response was not observed in any type of cultures using MMC-VCN-treated normal tissue cells. In 20 of these patients, the blastic response to nonviable frozen stored tumor cells was measured simultaneously and significant increase in blastic transformation by pre treatment of the cells with VCN was also observed in normal plasma cultures (p < 0.05). There was significant correlation between the response to nonviable frozen VCN-treated cells and to these, not frozen, viable cells (p < 0.001). These results indicate that lymphoblastic response to autologous tumor cells could be enhanced by pretreating the cells with VCN in patients with gastric carcinoma and the freezing could render the cells nonviable without interfering with the augmented immunogenicity of VCN-treated tumor cells.

13 patients with advanced measurable colorectal cancer, having failed chemotherapy including flu-orouracil, were treated on a weekly schedule with ICRF-159, 1.0 g/m2, and triazinate, 250 mg/m2. Only 1 patient had an objective partial remission, lasting 6 months. Toxicity was moderate, but acceptable. This is a case report of hyperpyrexia, shock and death following initial injection of low-dose bleomycin (7.5 units) for a poorly differentiated metastatic carcinoma. Premedication with high-dose dexamethasone and acetamiophen did not abort the reaction. A review of the literature uncovered one nearly fatal reaction to a test dose of 1 unit of bleomycin. All previously reported fatal cases were in patients with lymphoma who had received an initial dose of at least 25 units/m2. 68 cases of gastric carcinomas and 31 tissue samples adjoining the carcinomas were investigated by impulse cytophotometry, cytomorphology and histology, respectively. Differentiated adenocarcinoma tended to aneuploidy, while undifferentiated carcinoma often showed euploidy. The adjoining mucosa was characterized by polymorphism of the mucous superficial epithelial cells and the histograms showed slight abnormalities.

During an 11-year period, 76 patients with pathologic stage (PS) and 69 with clinical stage (CS) I and II Hodgkin’s disease were irradiated. The PS and CS patients were comparable by sex, age, stage, and histopathologic types. Treatment group 1 received a mantle for I and II A lymphocyte predominant and nodular sclerosis plus paraaortic for I–II A mixed cellular and lymphocyte-depleted or B symptoms; group 2 received mantle plus paraaortic for I–II A lymphocyte predominant and nodular sclerosis, and total nodal irradiation for the unfavorable group. The disease-free survival is 47% for the PS and 64% for CS patients in group 1. For group 2, 85% of PS and 83% of CS patients are disease free. A placebo-controlled treatment with the immunorestorative agent levamisole as an adjuvant to surgery and/or radiation was evaluated in a pilot study of 24 patients with squamous cell cancer of the larynx or hypopharynx. During the randomized treatment, started as close as possible to the beginning of the operation or to the irradiation sessions, 12 patients received a placebo and the 12 others levamisole in a dose of 50 mg t.i.d. for 3 consecutive days every 2 weeks. The recurrence rate in the control group was 50% after about 20 months and in the levamisole group only 20% after 2 years which is a significant difference. Also regarding cancer deaths there was a trend to the advantage of levamisole. The most promising effects were seen in patients with stages III or IV. Some evidence was found that an early start might be decisive for an optimal effect of levamisole. The beneficial effect of levamisole was only seen in the group of patients with a low pretreatment level of circulating lymphocytes.

Some properties and kinetics of synthesis of the carcinoembryonic antigen (CEA-LoVo) produced by an established human colon carcinoma cell line were analyzed. CEA-LoVo was assayed by the method of Chu and Reynoso 70% loss). The elution profile of CEA-LoVo in Concanavalin A-Sepharose B coincided with that of the First British Standard for CEA. The effects of theophylline treatment on mouse B-16 melanoma cell growth, metabolism, and membrane antigen expression in vitro were studied. Theophylline treatment inhibited DNA synthesis and the cell growth rate, and caused an elevation of intracellular cAMP levels. Cells treated with theophylline became elongated and assumed a normal fibroblast-like morphology. Theophylline treatment of B-16 cells also reduced the levels of tumor specific antigen and H-2 antigen detectable on the cell membrane.

Ingestion of 5-fluorouracil at 17 mg/kg/day by female retired breeders of the Wistar-Furth strain of rats beginning at 11½ and extending to 30 months of age prevented the appearance of a significant number of palpable mammary tumors and a highly significant number of spontaneous pituitary tumors in animals which reached the age of 30 months. Testosterone administration to castrated rats leads to increased concentrations of radioactivity associated with estracyt (a conjugate of carbamate with E2) in the prostate gland, particularly the ventral prostate. This contracts with the decreased concentrations of labeled E2. The findings appear to shed further light on the possible mechanism of action of estracyt in cancer of the prostate.
