In order to develop cell substrates suitable for the isolation of human oncornaviruses, a large number of human tumors, mostly sarcomas, were cultured in vitro. All explants yielded primary outgrowth of cells. These were of various nature and morphology and exhibited different growth potentials. About 40% of the tumors yielded cell strains which were undistinguishable from diploid human embryonic cell strains. About 20% yielded ‘difficult’ diploid cultures; 35% yielded short-term cultures mostly composed of undefinable cells, rarely of clearly recognizable tumor cells. About 5% yielded cell strains or lines which were aberrant in morphology or karyotype, or both. The malignant nature of the cell strains obtained in our study is conjecturable. 53 patients with intractable malignant diseases that were treated with a new alkylating agent, 4-N, N-bis (2-chloroethyl) amino phenyl N (P-carboxyphenyl) carbamate, also known as ‘IC-140’ were evaluated. The records of an additional 41 patients entered on this study could not be assessed from the standpoint of toxicity. At the dose level of 150 mg/m2/week, severe leukopenia ( < 2,000) and thrombocytopenia ( < 75,000) were encountered in 23 of 34 patients. On the other hand, at the 100 mg/m2 /week dose level, the severe toxicity was reduced to 8 out of 19 patients. Tumor response was evaluated in 43 patients. The overall response was 23% (29% at the 150 mg/m2, 13% at the 100 mg/m2) and the duration of the response varied from 3 to 32 weeks with a mean duration of 13 weeks. Two patients, each with three primary malignant neoplasms are presented. In an effort to identify a possible common denominator of induction of these tumors, it was found that both patients were alcoholics with liver disease, and that both had impaired delayed hypersensitivity reactions. The possibility of an increased susceptibility to cancer among alcoholics with liver disease is noted and the need for careful follow-up of these patients is suggested.

A total of 103 patients with lung cancer was treated with CCNU 130 mg/m2 orally every 6 weeks; 65 patients survived at least 6 weeks. Partial responses occurred in 7 patients. Leukopenia, thrombocytopenia, and nausea were frequent toxic effects. CCNU has slight efficacy in advanced lung cancer. No increase in survival was attributed to therapy.

A phase III study was designed comparing the effectiveness of Hexamethylmelamine (NSC 13875) to Dibromodulcitol (NSC 104800) in lung carcinoma. 250 of the 316 patients entered on the study were stratified into groups according to stage of disease and cell type. The results showed Hexamethylmelamine to be more effective in patients with squamous cell carcinoma and slightly superior to Dibromodulcitol in patients with anaplastic/undifferentiated cell carcinoma, whereas Dibromodulcitol proved to be more effective in patients with adenocarcinoma.

Beard’s theory can be restated in a modified form in modern terms in the following way. Cancer represents primarily trophoblastic tissue derived either from an aberrant germ cell or from a somatic cell whose normally repressed ‘asexual generation’ genes are abnormally reactivated (‘derepressed’). After topical treatment of cheek pouches of 10 hamsters with 9,10-dimethyl-1,2-benzanthracene and 2,4-dinitrophenol for 10 weeks no carcinomas developed in any of the animals. After treatment of 10 animals with the carcinogen only, there were 49 carcinomas in the 8 survivors. Inhibition of chemical carcinogenesis by dinitrophenol was tentatively attributed to suppression of ATP synthesis and to activation of ATPase by dinitrophenol.

The three major nuclear DNA-dependent RNA polymerases (enzymes I, II and III) were present in nuclear extracts from transplantable R-35 rat mammary tumors. Except for somewhat less enzyme III, their relative distribution resembled that of nuclear extracts from late-pregnant rats. When enzyme II from normal tissue extracts was incubated for RNA synthesis with cyclic AMP, inhibition was frequently observed, but this occurred less often with nuclear extracts from the R-35 tumor. In some experiments with both normal and tumor tissue, cyclic AMP and cyclic GMP increased the apparent activity of nucleolar enzyme lb and nucleoplasmic enzyme II, respectively. Nuclear extracts from both normal and tumor tissue contain proteins which bind radioactive cyclic AMP and cyclic GMP. Their patterns of binding were not identical. These results are consistent with the following hypothesis: altered binding by the tumor of cyclic nucleotides to putative nuclear ‘regulatory’ proteins (e.g. protein kinase subunits, or possibly other high affinity cyclic nucleotide-binding proteins unrelated to protein kinases) contributes to and may be responsible for some of the differences in response to cyclic nucleotides that were observed.
