Human peripheral blood lymphocytes were treated at first with phytohaemagglutinin (PHA) and then with 9,10-dimethyl-1,2-benz-anthracene (DMBA) in different concentrations. Lymphocytes treated with PHA and then with MBA in concentrations of 0.5 μg/ml continued their division during the whole observation period (14 days). The DMBA action on intact lymphocytes not only did not induce their division but also made them lose the sensitivity to the following action of PHA at least for several days. The results received are discussed in the light of the general theory of oncogenesis having been advanced earlier

The occurrence of breast cancer in twin sisters is a rare phenomenon. Twins developing breast cancer showing a mirror image are reported. Genetic and environmental factor play an important role in the development of the disease. It is recommended that, when one twin develops a malignant neoplasia, the other one should be examined and watched carefully.

Total hydroxyproline excretion in urine, and mucoprotein, alkaline phosphatase activity and calcium contents of plasma were determined in 66 female patients with cancer. Significant increases in the hydroxyproline content of urine and the mucoprotein content of plasma have been found to be associated with the patients bearing advanced breast carcinomas with skeletal metastases. The urinary hydroxyproline excretions, unlike the plasma mucoprotein concentrations, were persistently increased even before the osseous deposits were radiologically demonstrable in the patients bearing early breast carcinoma. The present results, however, do not demonstrate any correlation between either the plasma alkaline phosphatase activity or the calcium levels and the extent of skeletal metastases.

An attempt to evaluate the role of drug therapy in the development of primary liver cancer was made by investigating 37 consecutive patients diagnosed during a period of 10 years. 14 patients had previously been apparently healthy, 21 had been treated with drugs, and 2 were chronic alcoholics. The most notable finding was that 9 of the patients had used compounds known to alter the activity of drug-hydroxylating enzymes in the liver. In microscopic examination, the differentiation pattern of the cancer in these 9 patients was of the hepatocellular type, whereas in others a mixture of differentiation types was found. The findings evoke the question of whether long-term therapy with drugs changing the activity of metabolizing enzymes in the liver has some causality for the development of hepatic cancer in man.

The ‘G’ kindred is remarkable for the high incidence of a specific cancer aggregate in its members, particularly adenocarcinoma of the colon and endometrium, with relatively early ages of onset and a remarkably high incidence of multiple primaries. This specific familial association has previously been detailed in the literature as the ‘cancer family syndrome’. The parent-child concordance, observed between cancer-affected parents in the kindred and their 219 offspring, is 36.5%. The sibling-pair concordance for cancer, between first-affected siblings and their brothers and sisters, is 36.6% of 153 pairs. Manifestation of the familial malignancy in this particular kindred does not show sex influence, sex limitation, or disparity between matriarchal and patriarchal association for the cancer diathesis. The cell-mediated immunological response to a rat gliosarcoma was studied. A cytotoxicity assay, in which tumor cells prelabeled with 3H-thymidine were challenged in vitro with immune spleen lymphocytes and PEC, was used. Incubation for 72 h, released significantly higher levels of tritium label from tumor cell-immune spleen cell co-cultures than that released from tumor cells alone or with control lymphocytes. PEC were similarly cytotoxic. A cytostatic assay, where tumor cells were incubated with immune spleen cells and 3H-thymidine, showed significant reduction of incorporation of the labeled precursors into the DNA of the tumor cells compared to that of the controls. There was no significant difference in the inhibitory activity of PEC between the controls and immune animals, except when rats were further immunized by intraperitoneal injection of tumor cells. Incubation of the gliosarcoma cells with the cell-free supernatants from lymphocyte-tumor cell cultures showed that spleen lymphocytes and, to a lesser extent PEC, release in the presence of tumor cells a soluble inhibitory factor, or lymphotoxin. Supernatants of spleen cell-tumor cell co-cultures had an inhibitors effect comparable in terms of cell numbers to that of the spleen lymphocytes when these were directly incubated with tumor cells. This was not the case for PEC, whose supernatants were not effective. Dilution experiments with spleen lymphocytes and PEC, in which the volume of the incubation mixture was increased to 2 and 3 times the original value, keeping the concentration of the labeled thymidine constant, indicate that a major portion of the inhibition by spleen lymphocytes is attributable to a soluble inhibitory factor. On the other hand, the PEC inhibitory activity appears to depend more on a direct interaction with the target cells.

The immunologic reactivity of C3H/HEJ mice was evaluated with age increase. Tested was the response of the animals to a coupled tumor protein antigen (Lewis, CTPA) and the implantation of tumor tissue. The results of these studies indicate that although treatment with CTPA had supressed tumor proliferation in 7-week-old-mice, the tumors proliferated in 30-week-old mice. It is suggested that in C3H/HEJ mice, aging brings on some type of immune dysfunction.

The experience of Mitomycin-C given as a once weekly intraveneous injection for carcinoma of gastroenteric and breast origin with review of recent literature is given. Limited activity was seen in gastric and colorectal carcinoma. Similar to the experience of others, hematological toxicity (thrombocytopenia and/or leukopenia) was the dose-limiting toxicity encountered. Although the once weekly schedule was not directly compared with other reported schedules, it seems clear there was no reduction of significant and dose-limiting hematological toxicity nor enhancement of therapeutic efficacy.

The gastric pseudolymphoma (GP) is regarded as a benign non-tumorous hyperplastic alteration starting out from the lymph elements of the gastric wall. The gastric ulcer can play the role of an eliciting factor. In the reported GP angioproliferative foci were observed. GP is related to or identical with lymphoid hyperplasia in other localisations. The presence of angioproliferative foci is an important differential diagnostic sign.
