Lipid composition of the mitochondria in Ehrlich Lettré Hyperdiploid (ELD) ascites tumor cells of Swiss mice was determined. The total lipid contained 19% neutral lipid and 81% phospholipid. The major neutral lipids were sterols (64% of neutral lipids); free cholesterol (44.5%), esterified cholesterol (15.7%) and other plant sterols (3.8%), such as campesterol (2.3%) and β-sitosterol (1.5%). Nonsterol neutral lipids were triglycerides (22.0%), diglycerides (2.9%), free fatty acids (8.8%), and CoQ (2.3%). Phospholipids were comprised of choline glycerophosphatides (48.9%) of lipid p), ethanolamine glycerophosphatides (27.3%), cardiolipin (11.3%), inositol glycerophosphatides (4.2%), sphingomyelin (6.4%), and lysolecithin (1.9%). Fatty acids in the neutral lipids were highly unsaturated; cholesterol esters (83%), triglycerides (64%), and free fatty acids (51%). However, fatty acids unsaturation varied among the different phospholipids; cardiolipin (72%), ethanolamine glycerophosphatides (59%), choline glycerophosphatides (55%), phosphatidyl inositol (52%), and sphingomyelin (19%). Carbon 18 fatty acids predominated (50 to 77%) in neutral lipids and phospholipids except for sphingomyelin (6%) and phosphatidyl inositol (37%). The acidic phospholipids, such as cardiolipin and phosphatidyl inositol of tumor mitochondria, was similar to that of comparable rodent liver mitochondria but the amount of cholesterol was strikingly greater than had been reported previously for normal mitochondria of various tissue

Treatment with cyclophosphamide alone or in combination with methotrexate can cause considerable depression of the in vitro P.H.A.-lymphocyte transformation. This means that the longliving lymphocytes are affected. As a rule this depression is reflected by a fall of the number of circulating lymphocytes. One the other hand no influence on the content of the immunoglobulins has been found. Therefore it seems that the short living lymphocytes populating the germinal centres of the lymph glands remain functionally intact. More drastic forms of cytostatic therapy may have more serious consequences. Impaired lymphocyte transformation is the rule in the cachectic stage of malignant disease. It was also observed in several untreated cases of bronchus carcinoma.

Present evidence supports the original view of the medulloblast as a bipo-tential cell with the capacity for differentiation along either spongioblastic or neuro-blastic lines. Medulloblastomas behave much like other tumors of presumed embryonal origin, and in this respect the period describing risk of recurrence after treatment follows Collins’ rule of age at the time of treatment plus nine months. Always regarded as highly radiosensitive, medulloblastomas now emerge as radio-curable with almost one-third of postoperative patients surviving ten years beyond radiation therapy. Clinical trials have shown the sensitivity of medulloblastomas to two oncolytic drugs, methotrexate and vincristine. The rationale behind intermittent intrathecal administration of methotrexate is presented in the light of present knowledge. In the future, chemotherapy will assume an increasingly important role in the treatment of medulloblastomas resisting cure by irradiation.

L-tryptophan was found to consistently inhibit in vivo growth of the Walker 256 intramuscular carcinoma in rats. Doses of 8, 4 and 2 mM/kg body weight of L-tryptophan inhibited the tumor approximately 55%, 50% and 35%, respectively, the inhibitions being statistically significant. Other L-amino acids such as histidine, proline, leucine and glutamic acid, given in identical fashion and dosage as L-tryptophan, showed no inhibitory effect; L-phenylalanine showed only a very weak inhibitory effect at borderline levels of statistical significance. It was concluded that the action of L-tryptophan was specific and not related to an ‘amino acid imbalance’ brought about the administration of simply any amino acid in large dosages. The primary mechanism of action of L-tryptophan was thought to be due to its inhibition of the enzyme PEP carboxykinase, resulting in a block to gluconeogenesis and retardation of cancer cachexia. The toxicity of L-tryptophan was found to be nil in dosages below 6 mM/kg animal body weight/day and only slight at double that dosage. Morbidity, characterized by anorexia, was present at a dosage range of 8–12 mM/kg/day but disappeared within 24–36 h after the drug was stopped. In some experiments L-tryptophan administration resulted in a lower net animal weight gain than in control animals, whereas histidine, proline, leucine and/or glutamic acid administration stimulated the net animal weight gain; the latter effect was thought to represent at least in part a correction of cachexia, rather than simply a compensation for lack of nutrient intake by tumor bearing

An American patient with lymphosarcoma of the Burkitt type was treated with autologous remission plasma following relapse of his disease. No therapeutic effect was noted within a two-day period. Assay of the remission plasma for anti-HLV antibodies was negative. This case is compared with the successful results of similar treatment reported in the literature.

A precise, sequential staining procedure which demonstrates sulfated and non-sulfated mucopolysaccharides and glycogen in the urodele forelimb is described. The process, a combination of the alcian and PAS techniques, has proven valuable in following histogenic events during forelimb regeneration. Non-sulfated mucopolysaccharides are present in apical cap epithelium, sarcoplasm, and bone matrix. Both alcian blue and PAS responses are apparent in dedifferentiating cartilage. Glycogen was apparent in late blastema cells, mature and to a lesser extent in maturing muscle, and wound epithelium.
