The tetracycline analogs (TA) have been modified to contain boron. The new family of compounds provides a tool to reevaluate the over 40-year-old theory of thermal neutron, capture therapy for malignant cells.This report presents some tissue culture observations of the boron (TA) treated cells concerning intracellular incorporation and distribution. The synthesis of the boron TAs will be presented at a later date.

Attempts to establish monolayer cell cultures from an X-irradiation-induced transplantable rat mammary carcinoma were successful only on a short-term basis (3 to 4 passages). However, suspension cell preparations of the same tissue were successfully cultured. The tumor cells stained more darkly than normal rat cells and tended to agglutinate in the presence of commercial horse serum. Examination of the low passage monolayer cells with the electron microscope revealed the presence of C-type particles. After 151 days in vitro the suspension cells also contained C-type virus in various stages of replication

The uptake of radiocalcium (45Ca) and radiophosphorus (32P) by 5 different types of experimental tumors have been studied. Through a chemical fractionation various fractions were obtained. Calcium is in corporated mainly in the acetic acid soluble fraction, while phosphorus in the HCl soluble fraction. The tumor necrosis shifts the calcium accumulation to the HCl soluble fraction. These findings indicated that the insoluble calcium is in a chemical form other than hydroxyapatite.

Survival experience of 398 patients with chronic lymphocytic leukemia (CLL) seen at the University of Kansas Medical Center between 1942 and 1968 was computed by a standard actuarial method. Median survival for all patients was 4.5 years. Of the 398 patients 40 had never received any treatment for their CLL. The untreated patients were older at diagnosis than the treated patients (67.0 vs 61.5 years) and lived longer (6.0 vs 4.5 years median survival). Thus, there is a distinct group of older patients with CLL whose disease is usually diagnosed accidentally, who do not require treatment for their CLL, and who tend to live longer than average treated CLL patients.

The patho-histological, diagnostical and therapeutical problems of the mixed tumours of the parotid gland are discussed. In the cases of the benign mixed tumours the extensive excision has mostly been combined with radium treatment, and less frequently with post-operative X-ray radiation treatment. The percentage of the 5 years’ relapses corresponded to 4.2% in the authors’ material. In the cases of malignant mixed tumours similarly combined treatment has been made. The 5-years’ tumour-free condition corresponded to 9% and the 5 years’ survival to 41%. On the basis of the data of the literature and his own experiences the augmented radicality of the surgical intervention and the simultaneous radiation treatment are emphasized by the

Examples are given of opinions, which were prevalent long ago, but were, after a period of apparent neglect, ascribed to newer authorities. Thus, although taken as 20th century views, yet it was known by 1842 that tumour cells exhibit poor cohesiveness, while the need for adjuvant chemotherapy after surgery for cancer was appreciated as far back as 1700. It is hoped that illustrations such as these could promote research into the important but neglected subject of cancer history.

Experiments designed to test the premis that variations in intracellular ionic concentrations accompanying different levels of the electrical transmembrane potential difference might serve as a mechanism for control of mitosis in somatic cells have yielded results supporting the validity of the premise Employing naturally synchronized CHO cells in vitro and test media designed to produce intracellular concentrations of Na+, K+, and CI- approximating those which would exist at various naturally-generated Em levels in vivo, it was found that mitosis was completely, but reversibly, blocked in both short and long-term tests by simulated Em levels of ––70 mV and greater. This blockage was furthermore found to result from complete, but reversible, prevention of DNA synthesis. These findings that simulated high Em levels introduce a reversible G1 mitotic block in somatic cells in vitro lend credence to the supposition that natural modulation of cellular Em levels in vivo may constitute a basic mechanism for mitosis control in vivo<

The part played by heredity in the genesis of childhood neoplasia was investigated by examining the relative frequency of leukemia and other childhood malignancies in a widely scattered ‘family’ of British descent. It was found that in Canada, England and Wales, and New Zealand, where there are slightly differing incidence rates of malignant disease the proportions of leukemia and the other common malignancies were constant. Virgin Drosophila melanogaster were mated in culture vials containing Vinblastine, dissolved in distilled water, at concentrations of 0, 0.16, 0.31, 0.63, 1.25, and 2.5 μg/ml. Progeny were examined daily from the 10th through the 17th days. The frequency of teratogenic effects increased with the concentration.
