Peripheral blood lymphocytes from 100 healthy subjects, 32 patients with active Hodgkin’s disease and 34 patients in clinical remission were studied by means of the rosette formation test. Rosette formation by the lymphocytes of patients in stages I, II and III of Hodgkin’s disease did not differ significantly from the normal controls. Patients in stage IV of the disease showed a significant decrease in the number of rosettes formed. The rosette formation from patients who had been in a clinical remission for more than 1 year did not differ significantly from the normal controls. The total white blood count, total lymphocyte count and percentage of rosette-forming cells were followed in six patients with Hodgkin’s disease receiving quadruple cytotoxic drug therapy. There was a significant fall in total white count during treatment and a slight but significant rise in the proportion of rosette-forming cells. It is concluded that a fall in the proportion of rosette-forming cells only occurred in advanced Hodgkin’s disease where the patient was generally ill. It was not a feature of early or of quite widespread disease. It seems unlikely that Hodgkin’s disease itself causes a specific depletion of rosette-form

A panel of immunologic tests were used to characterize the levels of immunocompetence in 30 patients with advanced lung and head and neck region cancer prior to their receiving chemotherapy. All cell-mediated immunity parameters tested were significantly depressed in these patients when compared to controls. Antibody-mediated immunity tests showed lower levels of isoantibody titers to blood group antigens, no difference from controls in titrations for established virus antibodies and significantly elevated serum IgA and complement levels. Primary vaccination with KLH revealed marked deficiencies in both the antibody and cell-mediated immune response. The degree of immunoincom-petence did not correlate with either response to chemotherapy or the diagnosis to study interval which emphasizes the severity of immunoincompetence in patients with advanced stages of disease.

Immunization with ribosomal complexes, prepared from experimental ependymomas (transplanted in C57B/6J female mice) significantly decreases tumor growth and prolongs survival of hosts transplanted with this particular tumor. Experimental brain tumors present an interesting model for immunological studies because of the ‘priviledged’ situation of the brain itself, the lack of local lymphatic apparatus and the functional peculiarity of the blood-brain barrier. We studied a tumor model originally induced by methyl-cholanthrene and subsequently transplanted by Zimmerman and Arnold since 1940 [1]. The tumors grow either intracranially or subcutaneously in female mice of the inbred strain C57B/6J; their histopathology is considered stable. A paper by v. Ardenne and Reitnauer [1974] is commented with the aim to refer to already known theoretical and experimental results of critical O2 supply in tumor tissue of DS-carcinosarcoma. This summarizing representation shows that the findings by v. Ardenne and Reitnauer [1974] essentially correspond with previous results. (1) The Po2, values in tumor tissue reveal a frequency distribution tilted to the left and dependent on the tumor age. (2) The mean Po2 in the tumor tissue depends on the tumor weight in a logarithmical regression. (3) The time constants of O2 responses after sudden changes of arterial Po2 depend on the initial Po2 values in the tumor tissue. (4) The critical O2 supply radii in tumor tissue can be calculated by diffusion laws, all boundary conditions taking into consideration. (5). The reaction kinetics of the respiratory enzymes at very low O2 pressures can be taken into account by means of an analogue computer. So as slightly improved O2 supply situation in capillary networks is evident.

Cells isolated from dimethylnitrosamine (DMN)-induced renal mesenchymal tumors and established in long-term culture retained various properties of malignant cells. In contrast to cells cultured from normal rat kidney cortex, tumor cells appeared to have an unlimited growth potential, were able to establish clones at low seeding rates and to produce colonies in carboxymethyl cellulose (CMC) gel and on chick embryo chorioallantoic membrane (CAM), and were agglutinable by concanavalin A. Cells of tumor lines were quantitatively smaller than normal cells, had a higher nucleus: cytoplasm ratio, a broader range of multiple nucleoli and a higher mitotic index.

A case of osteogenic sarcoma with multiple lung metastases is presented. 85Sr lung scans demonstrated abnormal uptake. There was no close correlation between lung metastases seen on chest X-rays and the accumulation of 85Sr. It is suggested that uptake of 85Sr occurs mainly in active tumor bone formation and less in ossified metastases.

Inhibition of hepatic drug metabolism in the Sprague-Dawley rat by coumarin pretreatment or pregnancy reduced significantly the incidence of mammary tumors induced by 7,12-dimethylbenz(α)anthracene (DMBA) alone in untreated or nonpregnant animals. When DMBA injections were followed by coumarin treatment the rats remained unprotected against the carcinogen. The authors present 5 cases of primary malignant melanoma of the vagina and have reviewed 74 additional cases from the literature. The prognosis is extremely poor for this disease as compared to the usual primary squamous carcinoma of the vagina. Radical surgery combined with postoperative radiotherapy to the pelvis and inguinal node regions is suggested as the treatment of choice.

Bone marrow cells derived from phenylhydrazine-treated rabbits were grown in vitro for 24 h. Marrow cultivated in the presence of 1 x 103m dibutyryl cAMP form large dense cell clusters while those from control cultures form smaller more discrete cell clusters. Actinomycin D (1 x 10––7m) prevents cluster formation. Erythroid cell maturation is inhibited in the presence of DBcAMP. However, cholinesterase synthesis is enhanced. Most of the cells containing newly synthesized membrane-bound cholinesterase are erythroid. On the basis of the data obtained, it is suggested that the cyclic nucleotide may stimulate membrane protein synthesis. The effect of hydrazine has been studied on the chick embryos cultivated in vitro by the method devised by New. Hydrazine treated embryos when grown in Pannet-Compton’s saline for 18–20 h at 37.5 ± 1°C showed abnormalities which are predominantly restricted to brain, neural tube, optic vesicles and somites. The differentiation of heart is also affected but to a lesser degree; when subsequently treated with different amino acids, these show a certain degree of specificity. Not all the amino acids which were used are equally effective in bringing about a reversal of the hydrazine-induced malformation. Predominantly affected are the amino acid conversions which depend on transmethylation reactions. Thus the hydrazine-induced abnormalities are effectively reversed by glycine but not by serine, by methionine but not by homocysteine, thus suggesting that the amino acid conversions which are mediated by folic acid are impaired in the hydrazine-treated embryos. From amongst the nucleic acid precursors which are used for the subsequent treatment of hydrazine-treated embryos, thymidine can effectively reverse the inhibitory effects of hydrazine while uridine cannot. Since other precursors such as orotic acid and adenylic acid are not effective in bringing about the reversal, the results obtained by thymidine and uridine can be explained on the basis of a block in normal methylation reactions brought about by hydrazine. Folic acid itself, as well as other related substances such as p-aminobenzoic acid, choline, vitamin B12 and vitamin C are also quite effective in reversing the inhibitory effects of hydrazine.
