Insulin-induced pituitary growth hormone (GH), adrenocorticotrophic hormone (ACTH) and adrenal Cortisol response were studied in 12 patients with prostatic carcinoma. 3 patients demonstrated significant abnormal GH release associated with concentrations which remained on a high plateau during the study. 5 patients showed lack of ACTH-cortisol response to insulin-induced hypoglycemia. Those patients subsequently presented clinically with rapidly progressing disease.

The polyamines spermidine and spermine and the diamines putrescine and cadaverine have been determined in their free and unbound state in the sera of patients with colorectal carcinoma. The obtained values were compared with normal control levels. Elevated levels of putrescine and spermidine were found in untreated patients and declined to the control range after surgical and chemotherapeutic treatment. Spermine levels remained unchanged throughout the observation at normal control levels.

An evaluation of the 5′-nucleotide phosphodiesterase isozyme V (5′-NPD-V) test for liver metastases in gastrointestinal (GI) cancer patients was undertaken with a coded serum panel provided by NCI Mayo Clinic. Upon decoding, 28 of 30 patients (93%) with known liver metastases and 11 of 32 patients (36%) with no known liver metastases were positive. In 20 patients with various benign GI diseases, there were 6 (30%) positive for 5’-NPD-V. 2 of these positive patients with benign GI diseases had identifiable liver disorders. After exposure to the mutagen, ethylmethanesulfonate (EMS), or the carcinogen, urethane (UR), anchorage independence was observed in cultures derived from normal skin biopsies from patients with the heritable cancer syndrome, adenomatosis of the colon and rectum (ACR). Such colonies from methylcellulose suspension cultures did not produce tumors in athymic mice. Dissociation of growth in suspension from tumori-genicity was considered to be consistent with the hypothesis that a series of somatic mutation-like events occur in carcinogenesis. Such ACR cells should prove to be useful in studies on the genetic predisposition to cancer.

49 patients with advanced lung cancer underwent a combined treatment including radiochemoimmunotherapy. Before and at the end of the combined treatment, the immune status of the patients has been evaluated by testing the immunoreactivity to PPD in vivo and the phytohemagglutinin lymphocyte response and the rosette-forming cells in vitro. While the in vitro tests were not significantly modified by immunostimula-tion, as regards the immunoreactivity to PPD, we observed a positive conversion at the end of the treatment in 11 out of 25 patients who showed a negative reaction before therapy. All the patients who converted from negative to positive showed also a good response to therapy with regression or no progression of the tumor. Survival curves confirmed the prognostic value of PPD reactivity.

C57BL/6 mice with progressive B-16 melanoma develop a generalized immunosuppression, as measured by their lack of response to SRBC in vivo and in vitro. The severity of immunosuppression increases with the progress of the tumor, and is due to the generation of an antigen-nonspecific suppressor cell. Suppressor cells were first demonstrable 15 days after the appearance of the tumor, and their appearance correlated with the induction of immunosuppression in vivo. The suppressor cell generated by the growth of B-16 melanoma was a T lymphocyte since it was nonadherent to plastic and nylon wool, unaffected by passage through Sephadex G-10, and sensitive to treatment with rabbit antimouse brain serum and complement.

The tumor-specific leukocyte adherence inhibition (LAI) response of lymphoid cells from C57B1/6 mice sensitized to a murine-sarcoma-virus (MuSV)-induced tumor was inhibited by adherent phagocytic spleen cells from mice with progressively growing RBL-5 tumors. Both the direct LAI response and the formation of the mediator of LAI were sensitive to the suppressive activity of the progressor spleen macrophages. Suppression of LAI was not restricted by H-2 differences between the sensitized lymphocytes and progressor macrophages. Suppression was also nonspecific since the antibody plaque-forming cell response of normal lymphocytes to triphenylated hemocyanin was lowered by the addition of progressor spleen macrophages. Removal of adherent suppressor macrophages during the late stages of tumor growth revealed greatly elevated LAI reactivity not detectable in an unfractionated population.

The growth rate of the transplantable murine plasmacytoma MOPC 315 is decreased by daily injections of 2 mg Cortisol. Estradiol injections (0.01 mg/day) have a slight stimulatory effect while testosterone has no detectable effect. Growth rate is unaffected by propylthiouracil, triiodothyronine, and growth hormone. MOPC 315 contains glucocorticoid binding proteins which bind 3H-dexamethasone with high affinity. In intact cells there are 3–4 x 104 dexamethasone binding sites per cell as judged by competition experiments in which cells were incubated with a saturating concentration of 3H-dexamethasone in the presence and absence of a 1,000-fold molar excess of unlabelled dexamethasone. Scatchard analysis of binding data reveals the presence of a class of sites which binds about 2 x 104 molecules of dexamethasone/cell with an equilibrium dissocation constant (Kd) of about 3 x 10-8 M. In intact cells, only half of the saturable dexamethasone binding sites have stereochemical specificity for dexamethasone. This was deduced from the observation that dexamethasone binding to about half of these sites was displaced by the addition of a 1,000-fold molar excess of estradiol. Nuclear binding sites for dexamethasone were detected at a level of about 1 x 104/cell. Administration of prostaglandins F2α(PGF2α) and E2(PGE2) to syngeneic mice transplanted with chemically induced squamous cell carcinoma from the same species (allograft) markedly enhanced the trans-plantability and cellular atypicality of these tumors. Large and invasive tumors with an atypical cellular pattern, anaplastic squamous cell carcinoma, grade IV (or fibrosarcoma), occurred in approximately 87% of mice transplanted with squamous cell carcinoma, grade I, and treated with PGF2α. Similar transplanted tumors in syngeneic nontreated mice grew to a much lesser extent and remained squamous cell carcinoma, grade I. Light microscopic autoradiography and radioactivity measurements revealed a significant increase of 3H-thymidine incorporation in tumor tissue of transplanted and PG-treated mice as compared to control epidermis or to that of tumor transplanted only. Electron microscopic examination revealed that the cellular evolution in transplanted tumors treated with PGF2α is shifted towards atypical fibroblasts (fibrosarcomas) which originate from the squamous cells. Neoplasms developed in 18 of 98 toads, Bufo regularis, subjected to enforced feeding with bracken fern. They comprise 7 cases of adenocarcinoma in the ileum, 16 cases of hepatomas in the liver and 6 cases of neoplasms in the kidney due to metastases from the hepatomas. The results demonstrate that the Egyptian toad can be considered as an advantageous model for detecting the carcinogenicity of bracken fern, since the lesions occur faster than in other experimental animals.
