Commercially available transplantable adenocarcinoma (T1703) in Jackson Laboratory mice (DBA/1J) were studied using T2B2 to carry 105B for 105B thermal neutron capture therapy. The UCLA thermal neutron fluence was 2.7 × 101210n/cm2 and exposure was total body. There were no signs of typical radiation damage to skin, gastrointestinal tract, or hematopoietic system. The results indicated a significant (p= < 0.01) increase in mean survival of 15.85 days between T2B2 and Tc controls. Retardation in neoplastic growth was observed, and apparent permanent regression was achieved in 22.7% of the T2B2-treated animals

A wide variety of non-metastatic neurological syndromes occur in association with malignant lymphoma. 20 patients with Hodgkin’s disease and 20 matched healthy subjects underwent motor nerve conduction velocity testing of the right ulnar and left median nerves. Interferon may be induced in cervical carcinoma in situ by inoculation of either live virus measles vaccine or the synthetic polynucleotide, Poly I: C. Changes were observed in tumor cells with no changes in normal cells and tissues. Tumor cell changes were characterized by ‘punched-out’ holes in nuclei, vacuoles in the cytoplasm and elimination of nuclear DNA in altered cells. A high local concentration of Poly I: C in the region of the localized in situ lesions, it is believed, leads to interferon production by cells in or adjacent to the cancer producing antiviral influence presumably affecting viral components in tumor cells. Serial white cell and differential counts in 114 patients receiving 131 courses of cobalt-60 radiotherapy revealed the following findings: (1) Large field cobalt-60 radiation was associated with eosinophilia in 30% of all such courses. (2) Eosinophilia was most frequent in patients receiving pelvic radiation unrelated to the type of disease treated. (3) Typically the eosinophilia increased progressively to peak values in the 4th week of pelvic radiation. The possibility of antigen-antibody reactions explaining radiation-related eosinophilia (RRE) was investigated by serial observations of serum immunoglobulins.

Australia antigen (Au Ag) has been found in a higher frequency in patients with acute hepatitis, leukemia or Hodgkin’s disease as compared to normal subjects, in this study, 759 sera from 453 patients with advanced cancer were tested for Au Ag by the double radical immunodiffusion of Ouchterlony and by counter-electrophoresis. In addition, 40 patients with different non-neoplastic disease, 45 with malignancy of reticuloendothelial (RE) tissues, and 15 with viral hepatitis were tested for the presence of Au Ag. The Au Ag was found in 5 (1.1%) of the cancer patients, and in 3 (60%) of 5 patients with serum hepatitis. No Au Ag was found in any other sera tested. None of the 12 patients with hepatoma were found to have Au AG, while α-1-fetoprotein was found in the sera in 8 of the cas

Results of chemotherapy are presented in detail for 603 courses of therapy with 30 drugs in 400 consecutive patients with 22 types of solid tumors. An overall response rate of 20% was observed. The length of response and length of survival was surprisingly short, even in patients who were treated on an adjuvant basis following potentially curative surgery for tumors with poor prognosis. Although chemotherapy provides encouragement and temporary improvement for many patients, and long-term objective benefit for a few, substantial improvement in drugs and methods is needed to provide useful, long-term benefit for the majority of patients with solid tumors.

Limb regeneration in the newt, Notophthalmus viridescens, was retarded by treatment with acetylcholine or imidazole, whereas mitotic activity and regenerative growth were stimulated by cyclic AMP, its dibutyryl derivative, and theophylline. It is proposed that cyclic nucleotides are implicated in amphibian limb regeneration and may be related to the neural factor postulated by others.

Mishell-Dutton’s spleen cell model of immunity development was used to study the effects of common detergents on production of plaque forming cells (PFC). At 100 ppm the anionic detergent Tide XK and sodium dodecyl sulfate (SDS) inhibit PFC development by approximately 50%. Nitrilotriacetic acid (NTA) and Haemo-Sol are non-inhibitive. Nonionic Triton X-100 completely inhibited PFC production at 10 ppm. Cationic cetyltrimethylammonium bromide (CTM) added on explantation day gives 80% inhibition at 0.01 ppm. Cytotoxicity of CTM appeared at lower concentrations than PFC inhibition. Though less favorable to undesired algal growth, cationic and nonionic detergents inhibit antibody development in vitro at low concentrations and may have a teratogenic potential in vivo

N-methyl-N'-nitro-N-nitrosoguanidine (NG) in drinking water (83 mg/l) results in a high incidence of gastric adenocarcinoma in random-bred Wistar rats. When NG was discontinued after 25 weeks, 61% of animals developed cancer and after 52 weeks the incidence was 73%. In a Wistar-Mai-Furth group (39th generation) invasive adenocarcinoma was found in 12% of rats ingesting NG for 25 weeks and in 10% exposed for 52 weeks. No adenocarcinomas and only two sarcomas were found in 50 Buffalo-Mai rats (56th generation) receiving NG for 52 weeks. Varying the dose and frequency of NG administration did not overcome host resistance. Host resistance to gastric adenocarcinogenesis induced by NG may represent a new advantage of this animal model.

A new concept of a basic mechanism involved in cell proliferation is presented. It is suggested that cells are normally restrained from proliferating by the highly viscous nature of the intercellular glycosaminoglycans. In order to proliferate, cells must escape from this restraint by depolymerizing the glycosaminoglycans in their immediate environment. This process is accomplished by the release of the enzyme hyaluronidase and is kept in check by physiological hyaluronidase inhibitor. There is some evidence that physiological hyaluronidase inhibitor is an oligoglycosaminoglycan that requires ascorbic acid for its synthesis, and perhaps incorporates residues of ascorbic acid. This hypothesis provides an explanation for the pathogenesis of scurvy. It explains the increased requirement for ascorbic acid that occurs in many cell proliferative diseases, including cancer. It indicates the existence of a basic underlying mechanism in many pathological states and suggests a common pattern of treatment. We conclude that ascorbic acid may have much greater therapeutic value than has been generally assigned to it.
