Immunoperoxidase staining was used to examine carcinoembryonic antigen (CEA) expression in 167 breast cancer cases. Patients with histological evidence of regional or localized breast cancer who lived less than 3 years or greater than 10 years were assessed. Overall expression of CEA was 65%. There was no significant correlation in CEA expression and survival in either regional or localized breast cancer cases. There was no association between CEA expression and number of lymph nodes involved, size of tumor, parity, gravidity, blood type, or menopausal status of the patients in either group. When the lymph nodes of cases with regional breast cancer were examined, there was a statistically significant number of short survivors whose primary tumor was negative for CEA, but whose metastatic tumor expressed the marker when compared to long survivors with regional lymph node involvement.

A comparative study was made on the ectopic production of chorionic gonadotropin (hCG) and chorionic somatotropin (hCS) by carcinoma of the uterine cervix. Incidence of hCG in the sera of cancer patients was much higher than hCS. hCS has a better prospect in early detection of cancer cervix than hCG. All hCS-producing tumors and the majority of hCG-producing ones were histologically diagnosed as squamous cell carcinoma. The source of hCG and hCS were found to be the tumor cells. A fall or disappearance of detectable hCG and hCS was noted following surgery or radiotherapy.

33 consecutive patients with metastatic or locally recurrent soft tissue sarcoma were treated with a modification of the CYVADIC (cyclophosphamide, vincristine, Adriamycin, dacarbazine) protocol. In an attempt to enhance the response rate without increasing the toxicity, the dose of Adriamycin was raised to 60 mg/m2 and the dose of cyclophosphamide to 600 mg/m2 in each cycle. The dose of dacarbazine was decreased to 150 mg/m2/day for 5 days. Of 31 evaluable patients there were 4 complete responses (13%) and 9 partial responses for a total response rate of 39%. 11 patients (33%) had minimal response or stable disease. The response achieved with our dose modification was no better than that achieved in several series with conventionally dosed CYVADIC.

Polyclonal immunoglobulins (Ig) were measured at diagnosis and/or following chemotherapy in 226 patients with a malignant plasma cell dyscrasia (PCD), including 11 patients with solitary myeloma (SM) and 215 patients with multiple myeloma (MM). At diagnosis, Ig synthesis suppression was observed in 80.7% of patients with MM but never in case of SM (p < 0.001). In patients with MM, there was a clear correlation between IgA or IgM levels (but not IgG) and the total body burden of myeloma cells (p 1-year plateau period. We concluded that polyclonal Ig evaluation appeared to be of diagnostic and prognostic values in the management of malignant PCD.

The diagnostic accuracy of measurement of carcinoembryonic antigen (CEA) and of analysis of the cytological characteristics of pericardial fluid was assessed in 10 patients with benign pericarditis and 7 patients with malignant pericarditis. The mean CEA levels of pericardial fluid specimens from patients with malignant pericarditis were significantly higher than those from patients with benign pericarditis. However, a high CEA level was not seen in a specimen of pericardial fluid which contained nonepithelial tumor cells. Positive cytological findings were obtained in specimens of pericardial fluid from 85.7% of the patients with malignant pericarditis. Correct diagnosis of malignancy was made by CEA assay alone or by cytological examination alone in 85.7% of the patients examined, while a combination of these methods raised the diagnostic rate to 100%.

To define the in vitro response of nonparenchymal liver cells to chemical carcinogens, an endothelial-like cell line (RLC-1) derived from adult rat hepatic tissue was established in long-term culture. The RLC-1 line exhibited morphologic, ultrastructural and cytoskeletal characteristics consistent with derivation from mesenchymal vascular elements within the liver. Using various criteria to monitor changes in growth properties, RLC-1 cells were determined to be resistant to transformation by diethylnitrosamine, partially transformed by benzo[a]pyrene, and completely transformed by methylazoxymethanol acetate and N-methyl-N’-nitro-N-nitrosoguanidine. The effect of the differentiation-inducing agent dimethylsulfoxide (DMSO) on the protein composition of hepatic tumor cells was evaluated using the BW77–1 line of murine hepatoma cells previously determined to be DMSO-inducible for enhanced liver function. DMSO-mediated suppression of BW77–1 proliferation was accompanied by a stimulated uptake of [35S]-methionine and incorporation of the isotope into acid-insoluble product. DMSO produced a quantitative increase in the major cellular and secreted protein species per 106 cells although qualitatively the pattern of major peptides among control and DMSO-treated populations was basically similar; increases in two minor peptides of 90,000 and 110,000 daltons was noted, however, in the secreted protein fraction of DMSO-treated cells relative to controls. The data collectively suggest that the enhanced levels of differentiated liver cell gene products previously observed as a consequence of DMSO exposure reflects a stimulation in the synthesis, accumulation and secretion of at least all of the major proteins normally produced by BW77–1 hepatoma

The structural features and lipid solubility of four different classes of antifolate compounds were compared for their inhibition of dihydrofolate reductase (DHFR) and growth in a normal and methotrexate (MTX)-resistant 3T6 mouse cell line. All of the compounds have been shown previously to have antifolate activity. The resistant cell line has a 7-fold increase in DHFR activity with normal transport, but an altered affinity for MTX. All the antifolates were equally effective in inhibiting DHFR and growth in the parent cell line. Inhibition of partially purified DHFR from the resistant cells increased with changes in lipid solubility and structure of the compounds, compared to the parent DHFR. These data demonstrate that the resistant cells may be more sensitive to the structurally dissimilar antifolates than to MTX and lend importance to further development of this type of antifolate. These results suggest that these compunds may be useful in circumventing antifolate resistance due to alterations in target enzyme concentration and drug-enzyme affinity, as well as drug transport.

The hypothesis was put foreward that ‘immortalisation’ of an established nontransformed cell line might represent one type of ‘initiation’ in the process of malignant transformation. Therefore, a nontransformed rat fibroblast cell line, FR3T3, was investigated as to whether or not it could be irreversibly transformed by exposure to the tumor promoter 12–0-tetradecanoylphorbol-13-acetate (TPA). TPA was applied in different treatment schedules at varying growth phases of the cells (logarithmic or stationary growth). The cells’ acquisition of transformation-associated properties was studied by means of various transformation assays (saturation densities and formation of dense foci both at low and high serum concentrations, as well as growth in agarose and production of plasminogen activator, PA). We found that a phase of stationary growth plus TPA treatment was decisive for the induction of transformed cell clones. Growth in 0.5% serum was used as the selection procedure. Several isolated foci manifested, in vitro, highly transformed phenotypes. However, only 1 of 12 clones produced PA. All cell lines derived from these clones, proved to be tumorigenic in syngeneic animals. Since the nontransformed, but ‘immortal’ rat cell line is susceptible (at certain growth conditions) to the tumor promoting activity of TPA, we conclude that induction of’immortalisation’ may, from an operational point of view, correspond to ‘initiation’, at least in th
