Immunoperoxidase methods permit the demonstration of a variety of antigens in routinely processed tissues. The morphological detail obtained by this procedure is comparable to orthodox haematoxylin and eosin stained sections, and so offers the diagnostic pathologist the opportunity of simultaneously identifying normal and neoplastic cells both by their morphological characteristics and by their antigenic constitution. The application of this method to a study of malignant lymphomas has contributed to current re-appraisal of classification criteria for this group of neoplasms. It is anticipated that, as specific antisera against a range of different tissue antigens become available, immunoperoxidase methods will have a similar impact upon the re-definition of morphologic criteria related to other neoplasms.

The isoenzymes of human prostatic acid phosphatase have been studied by an isoelectric focusing technique. Purified acid phosphatase from malignant prostates contained eight isoenzymes with pI 4.4–5.3. The sera from patients with prostate cancer were shown to have similar acid phosphatase isoenzyme patterns at pI 4.0–5.5; as the serum enzyme activities increased, the pI of isoenzymes shifted to more acidic pH. These isoenzyme patterns of sera from patients with prostate cancer were different from those of patients with Gaucher’s disease or from acid phosphatase of human erythrocytes, both of which exhibited only one enzyme band around pI 5.0 and 6.0, respectively. Treatment of serum sample of prostate cancer with neuraminidase did not result in a single enzyme band but alter the pI of isoenzymes, which shifted to a higher pH region. The extracts of human prostatic tissue specimens contain oncornavirus-like reverse transcriptase activity. This activity was isolated by banding the tissue extract in an equilibrium sucrose density gradient followed by phosphocellulose chromatography of the lysate of the material banding at a density of 1.14–1.20 g/cc. 74 patients with untreated cervical cancer (FIGO II and III) were skintested with a battery of recall antigens, and also sensitised and challenged with DNCB. A significant reduction of reaction to Tuberkulin, Varidase and also to DNCB was found in patients with stage III in comparison to healthy females of the same age group. Significant changes in immunoglobulin levels, increase of IgA and decrease of IgG were ovserved in the cancer patients. Serum lysozyme values were the same as in the control group.

We have performed B2 microglobulin determination in 18 non-cancer and in 38 cancer cases. We found a higher value in the control group in 11% (2 cases) and a pathologic value in 43.5% (17 cases) in the group of patients with primary bronchial cancer.

The effect of Levamisole was studied in an animal model of Wilms’ tumor. No tumoridical effect of Levamisole could be documented in this tumor model, and no effect was shown on prevention of tumor, when Levamisole was given before tumor implantation. Three preparations of BCG, currently in use for clinical trials in cancer, were tested for relative potency as an immunological adjuvant. Using a lymphocyte trapping assay, Pasteur Institute BCG was shown to be a more effective adjuvant than the Tice strain or the Glaxo preparation of BCG.

Serum haptoglobin (Hp) levels were determined periodically in mice treated with the carcinogen 7,12-dimethylbenz[a]anthracene (DMBA). The magnitude and duration of the Hp response to tumors induced by DMBA were dependent on the tumor type; lymphocytic lymphomas elicited a minimal response, whereas mice bearing mammary carcinomas and stomach squamous cell carcinomas had high Hp levels which remained elevated throughout most of the period of tumor development. In the majority of mice with mammary carcinomas, the initial rise in serum Hp coincided fairly closely with the first appearance of palpable masses. Some variation in the magnitude ot the Hp response was observed between individual mice bearing chemically-induced tumors of similar histological types.

The effect of chronic ethanol intake on the growth and spread of some murine tumors has been investigated. The treatment had no effect on the B 16 melanoma but tended to decrease the number of Ehrlich ascites cells. In the case of the Lewis lung carcinoma, administration of ethanol for two weeks tended to lower the number of metastases to the lung without significantly affecting the primary tumor size, whereas more prolonged ethanol intake decreased the weight of the primary tumor in addition to decreasing its dissemination.
