Supernatants of established cultures of human neoplastic and normal cells have been shown to contain a number of different biological activities, including inhibition of DNA synthesis as measured by thymidine uptake. We have found that supernatants of melanoma cell lines contain an inhibitor of thymidine uptake which is heat labile, ultraviolet sensitive, non-filtrable (0.22 μ) and partially sedimentable at 20,000 × g. The mechanism of action of the inhibitor involves the degradation of 3H-thymidine to a non-utilisable form, which we detect by failure of uptake of 3H-thymidine by cultures of mitogen stimulated lymphocytes to which the inhibitor is added. While microbiological tests have failed to reveal mycoplasma contamination of inhibitor producing cultures, treatment of these cultures with kanamycin suppresses inhibitor production. Qualitative evaluation of DNA synthesis by the inhibitor producing cultures using autoradiography and fluorescent DNA staining has confirmed the presence of mycoplasma. With the widespread use of established cell lines in cancer research, it is imperative that screening for mycoplasma contamination go beyond routine microbiological assays. Detection of 3H-thymidine degradation by cell culture supernatants is an additional simple and sensitive indirect test which could be used for this purpos

Leukocyte adherence inhibition (LAI) tests tor the assessment of tumor immunity in vitro have been based on the assumption that the adherence of the leukocytes is inhibited by specific interaction with an antigenic tumor extract. However, loss of leukocyte adherence could depend on several non-immunological factors including disease state of the leukocyte donor and the presence of a variety of agents, proteins or tissue extracts. We have studied LAI, therefore, using different species and concentrations of serum protein or tissue extracts. Our results show that leukocytes from patients with a variety of diseases adhere to glass in a rather consistent fashion. High concentrations of serum protein or tissue extract inhibited leukocyte adherence. 39 breast cancer patients and 38 female controls were studied with a total of 23 solubilized extracts from breast carcinoma, other tumor and non-tumor tissues. Breast cancer patients showed selective LAI in the presence of 200 μg per test tube of several breast carcinoma extracts

Nineteen human tumors, mostly of sarcomatous nature, were cultured in vitro. Three cell lines were isolated and further characterized :MG-57 derived from a giant cell tumor, MG-63 derived from an osteosarcoma and MG-72 derived from a xanthohistiocytoma. The cell lines varied in morphology and growth pattern. An abnormal karyotype with marker chromosomes was present in MG-63 and MG-72. None of the cell lines spontaneously produced detectable C-type virus particles. Stimulation with IUDR and dexamethasone also failed to induce detectable particle release.

We have applied a triple combination of 5-fluorouracil (5FU), vitamin A and cobalt-60 radiation (FAR therapy) to clinical treatment of head and neck tumors since 1972. The treatment of 33 patients with cancer of larynx and 15 patients with hypopharyngeal or cervical esophageal cancer with FAR therapy during three years from 1972 to 1974 resulted in highly effective synergism in vivo, and the improved points of this therapy were presented.

In 35 children with acute lymphatic or undifferentiated leukemia, immunological properties of normal lymphocytes and lymphoblasts were investigated. In most cases the number of circulating normal lymphocytes with surface or B markers and delayed type hypersensitivity was depressed. On the other hand, the response to mitogenic stimulation with PHA as well as the antibody and immunoglobulin serum concentrations were found to be normal. Antigens present in leukemic sera and organ extracts, but absent from non-leukemic sera and tissues, were detected by examining their reactions with anti-microbial antisera by means of counter-immunoelectrophoresis. Out of 110 different antisera used, 12 antisera, those produced against Micropolyspora, Coccidioides, Salmonella, Mycoplasma, Mycobacterium simiae, Absidia, pseudo-rabies virus and adenovirus, proved to react regularly with the leukemic sera. Absorption studies point to an immunological specificity of these cross-antigenicities. Interpretations and implications of these discoveries are discussed in the paper.

A patient with both chronic lymphatic leukemia and multiple myeloma is presented. Characterisation of the leukemic cells showed them to possess Fc receptors but to lack SmlG and C3 receptors. Intracellular immunoglobulin was not detected within the leukemic cells. The multiple myeloma presented as a soft tissue mass on the chest wall and produced an IgGK paraprotein. The possible relationship of the two disorders is discussed.

Radionuclide emission tomography and ultrasonography were obtained on a patient with recurrent epigastric pain. In the pancreatic region an ultrasonic scan revealed a conglomerated mass. Following administration of Se-75 selenomethionine, the multiplane longitudinal tomographic scan demonstrated marked concentration of activity in the region of the pancreas which corresponded to a large defect in the hilus of the liver observed on prior liver-spleen scan with Tc-99m sulfur colloid. Surgery revealed a poorly differentiated histiocytic lymphoma in the retroperitoneum. The radionuclide and echographic scans of the retroperitoneal lymphomas are briefly discussed, and their complementary role as a diagnostic tool stressed.

The application of standard radiographs (metastatic series) and radionuclide scanning in the serial evaluation of patients treated for metastatic osseous lesions is reviewed. False positive interpretations of bone scans are related to the inability to distinguish osteoblastic healing response from osteoblastic response to progressive tumor. False negative scans are uncommon and are related generally to symmetrical osteoblastic activity or to inadequate (subthreshold) osteoblastic response to tumor. Standard radiographs are preferable to bone scans in the serial evaluation of the effectiveness of therapy for osseous metastases in that anatomic quantitation of lesions as well as the dynamic evolution of lytic lesions to ‘blastic’ healing may be appreciated. Differentiated (papillary) thyroid cancer was detected 17 years following radioiodide 131I treatment for toxic multinodular goiter. 21 cases of thyroid cancers with previous 131I therapy for hyperthyroidism were summarised. This combination is rare compared to the incidence of thyroid cancers following external irradiation. This may be due to higher absorbed dose to thyroid in 131I treatment.
