The growth of a syngeneic Moloney lymphoma in CBA mice was inhibited by the injection of lymph node cells from normal CBA donors. This cytotherapeutic effect was also seen after chemotherapy with cyclophosphamide (CY) or dimethylmyleran (DMM). Two other murine tumours, the Meth A sarcoma in Balb/c mice and the L1210 leukemia in BDF1 mice, did not respond to treatment with normal syngeneic cells. In the L1210 system, tumour-promoting effects of fetal liver cells were demonstrated. The tumour inhibitory effects of normal lymph node cells against the Moloney lymphoma may be a virus-dependent phenomenon mediated through interferon and ‘natural killer’ cells. 200 mg/m2 methotrexate given intravenously in a running drip for 6 h has been used as an initial adjuvant therapy in 38 patients with advanced head and neck cancer. The response rate is as high as 80%, with 21% achieving complete remission. Histologically, specimens were tumor free in 3 patients. Toxicity in 38 patients included leukopenia (4), mucositis (6) and diarrhea (1). This particular dose of methotrexate appears to be safe and usually does not need leucovorin rescue. Also, when given as initial treatment, it is effective in reduction of tumor bulk. A prolonged randomized trial is essential to determine its role in improving long-term survival.

Macrophage and T lymphocyte content of sarcoma tumors in mice treated with Propionibacterium granulosum (PG) by different routes was investigated and compared with the effectiveness of the therapy. Two different methods for tumor macrophage detection, the Fc receptor analysis and phagocytic properties, were employed. Indirect fluorescent staining of theta-antigen on lymphocytes was used to evaluate the proportion of tumor T cells. Systemic treatment with PG appeared to be effective if given soon after tumor implantation; however, this route of administration failed to affect the tumor growth if given when the tumor was already advanced. The therapeutic efficiency of intralesional PG injection was almost as effective as early systemic treatment. We obtained evidence indicating that effectiveness of PG therapy was associated with an increase of the macrophage and T cell content of tumors.

Immunochemical analysis of various tumors, representing 12 different tissue sites of origin, revealed a γ-fetal antigen(γ-FA)-like component in 17 of 23 malignant neoplasms of the mouse. This antigen was identified in extracts of spontaneous and carcinogen-induced murine tumors as well as in extracts of hepatic tumors initiated in the rat with diethylnitrosamine. The occurrence of γ-FA in 75% of the malignant tumors examined and its presence in the sera of rodents bearing transplanted and primary cancers suggests an associative relationship of potential diagnostic value. The in vitro proliferation of herpesvirus-transformed marmoset lymphoblastoid cell lines (LCL) was inhibited by retinoic acid and retinyl acetate. Both Herpesvirus-ateles- and Herpesvirus-saimiri-transformedLCL with T-cell characteristics and Epstein-Barr-transformed LCL with B-cell markers were more sensitive to retinoic acid than to retinyl acetate. Inhibition of LCL proliferation was dependent on retinoid concentration and became apparent only after 3–4 days of exposure. In vitro assays for marmoset LCL sensitivity to retinoids may indicate the potential usefulness of these compounds in the chemoprevention of virus-induced lymphoma in vivo.

After oral administration of 100 mg Prednimustine® to patients with metastatic breast carcinoma, prednisolone levels and the area under the plasma concentration-time curve between 2 and 24 h are approximately half of those obtained after the administration of an equivalent dose of prednisolone. As previous studies have reported up to 55% of unchanged Prednimustine in feces, it is suggested that prednisolone plasma levels are as good a parameter of Prednimustine absorption and bioavailability as chlorambucil levels, Prednimustine itself having never been detected in plasma. Whether Prednimustine is hydrolyzed in the intestine before absorption or rapidly taken up by the tissues and hydrolyzed in situ after absorption is unclear at the moment. Myelosuppressive effects of Prednimustine correlated very well with maximal prednisolone levels, emphasizing the value of prednisolone level measurements

Inhibitory effects of two human interferon preparations, leukocyte interferon (Le-IF) and fibroblast interferon (F-IF), on granulocytic progenitor cells (CFU-C) from hematologically normal cancer patients and from patients with chronic myelogenous leukemia were evaluated. There was a wide variation in sensitivity of CFU-C to both Le-IF and F-IF. F-IF was more inhibitory against CFU-C than Le-IF. Normal CFU-C and chronic myelogenous Leukemia CFU-C were equally inhibited by both interferons. Effects of both interferons were neutralized by corresponding specific antisera but not by the other antisera. These observations confirmed that differences in immunogenicity of the interferons may attend their different origins.

In 2 patients with the malignant carcinoid syndrome, a Swan-Ganz balloon tip, flow-directed catheter was used to occlude hepatic arterial flow and perfuse hepatic metastases with 5-Fluorouracil. The first patient, symptomatic for 16 months, was rendered asymptomatic for 14 months after treatment. The second patient was relieved of severe diarrhea and generalized urticaria, but progression of the cardiac manifestations were unaffected. Symptoms of the carcinoid syndrome are usually the most debilitating aspect of this illness. When more conservative measures cannot alleviate their symptoms, hepatic artery occlusion with chemotherapeutic perfusion of metastases should be considered.

The plasma levels of 5α-dihydrotestosterone, testosterone, &karsi028;5-androstene-3β,17β-diol, &karsi028;4-andros-tenedione, dehydroepiandrosterone, estrone, estradiol, dehydroepiandrosterone sulfate, follicle stimulating hormone, luteinizing hormone and prolactin were measured in normal post-menopausal women and in post-menopausal women with proven breast cancer. The mean prolactin level was significantly higher (p < 0.01) and the mean 5α-dihydrotestosterone level lower (p < 0.05) for the women with breast cancer. There were no significant differences for any of the other hormones. A case of multiple myeloma associated with myelofibrosis and myeloid metaplasia is reported. The presentation of our patient and a detailed review of the current literature suggest that more than a chance relationship exists between the two disorders. The role of chemotherapy in the induction of bone marrow fibrosis appears to be unlikely.
