Plasma samples from a group of patients with severe essential hypertension were formerly shown to have a cardiotonic action on the isolated frog heart. This effect is significantly greater than that observed with plasma from normal persons or those with other diseases. The material in plasma responsible for this action is a system of 3 proteins. These proteins have been distinguished by fractionation with sodium sulfate, and are found among the serum globulins. The protein, the concentration of which determines the activity of the whole system, is called component L. When applied to the frog heart it becomes strongly bound. Two other proteins are needed, one precipitated between 0 and 17 per cent sodium sulfate, the other between 22 and 30 per cent sodium sulfate. The 0 to 17 fraction is thought to require calcium for its activity, which is bound to the protein. Calcium can be released by the addition of component L in a reaction which is dependent on temperature, reversal being possible in high calcium concentrations in the cold. The protein system described causes contracture of frog heart muscle in the absence of ionic calcium, in contrast to other well-known contracture-causing agents which require the presence of free calcium for activity.

Studies of the simultaneous rates of transfer of various tracers and stable isotopes from the pericardium of normal dogs under pentobarbital anesthesia indicated that the rate of transfer of Rb86was greater than that of Na24and Cl36, which were approximately equal. These observations show the pericardium to be a very active serous membrane concerned with electrolyte transfer phenomena as well as an excellent experimental model for the study of the kinetics of exchange of electrolytes across a normal living membrane.

Control of lipid metabolism in the surviving bovine aorta by chemical agents has been demonstrated. For this demonstration an internally controlled technic was developed. This teehnic is briefly described as well as the results assaying several hormones and 2 vitamins.

The effect of phenylhydrazine-produced anemia upon the hearts of rats was studied with observations upon the accompanying hypertrophy. Results indicated that the mechanism of hypertrophy under these conditions may be one of myocardial injury rather than mechanical.

Deep hypothermia in the rat was studied with respect to the sequential changes in body temperature, heart rate, and electrocardiogram. In vitro assays of myocardial metabolic activity were made at temperatures ranging from 5 to 35 C. These assays strongly suggested that the in vivo cardiac dysfunctions observed at low temperatures were attributable to shifts in temperature kinetics of enzyme systems at 20 to 21 C.

This study is the first attempt to correlate myocardial oxygen availability by the polaro-graphic method with direct electrocardiographic leads and cinematographic records of muscle contraction during experimental acute coronary branch occlusion and narrowing. The comparative insensitivity and slowness of the epicardial electrocardiogram as an index of acute regional ischemia is demonstrated. Special attention is given to the rates of myocardial oxygen change immediately following attainment of a significant degree of coronary obstruction, the effects of pure oxygen inhalation on the experimental situations, and alterations in coronary vein color following release of arterial occlusion.

The effects of treatment of rats with biphenylylbutyric acid were assessed by measuring the amount of acetate-1-C14incorporated into the cholesterol and phospholipid of several tissues. The drug inhibited the inclusion of the label into the plasma and hepatic lipid fractions. The treatment had no significant effect upon the lipid metabolism of either the testis or the adrenal. In the aorta, although the incorporation of the labeled acetate into cholesterol was unchanged, the amount of label found in the phospholipids was strikingly enhanced by the treatment. The unique nature of aortic lipid metabolism as evidenced by this work and previous reports is discussed.

The influence of glucose, given orally and intravenously, on the acceleration of plasma clotting and appearance of lipemia following the feeding of peanut oil was determined in dogs. Oral glucose blocked or masked the appearance of plasma lactescence and clotting accelerator activity, whereas intravenous administration tended to delay their appearance. In other experiments heparin abolished or reduced plasma lactescence without modifying clotting accelerator activity induced by fat feeding.

Cr31labeled red cells and I131human serum albumin were injected intravenously into 8 mongrel dogs. After 1 hour kidneys were removed and frozen rapidly with dry ice. Sections of outer cortex, inner cortex, outer medulla, inner medulla, outer papilla and inner papilla were removed and analyzed for Cr51and I131radioactivity. Tissue radioactivity was compared with that of arterial blood and concentrations of labeled red cells and albumin were calculated. No significant difference was found in relative red cell and albumin content between outer and inner cortex. The magnitude of the I131albumin content in the cortex is interpreted to indicate a sizeableextnivascularexchangeable albumin pool. The renal papillae were extraordinarily deficient in red cells, but contained per 100 Gm., albumin equivalent to that of 39 ml. of plasma.
