The pathogenesis of the vascular lesions of polyarteritis nodosa found in young rats with experimental renal hypertension is described. The panarteritic lesion was a product of the combination of renal injury and feeding sodium chloride. It began with a fibrinoid necrosis of the intima and media which was followed by an extensive inflammatory reaction with subsequent fibrosis of all layers of the vessel wall.

By means of an intracellular capillary electrode the effects of barium chloride on the resting and action potentials of ventricular fibers of the frog were recorded in hearts with circulation intact and hearts isolated and perfused. The changes were essentially similar in both preparations. The major change observed was a statistically significant (p <0.01) increase in duration of the action potential. Changes were also observed in the magnitude of the resting and action potential, in the rate of depolarization, and in rhythm, but these did not occur invariably. By a series of exclusion experiments it was demonstrated that barium has a direct effect on the ventricular fibers of the frog's heart.

The administration of potassium to hypotensive, potassium-deficient rats results in a prompt restoration of their lowered blood pressure to normotensive levels. Similar repletion of the potassium deficit in adrenalectomized rats is only partially effective in restoring the blood pressure to normotensive levels. The spontaneous retention of potassium which occurs in potassium-deficient rats following adrenalectomy fails to restore lowered blood pressures to normotensive values.

Several measures of cardiovascular function were made in nine dogs before, during, and after the production of anemia by daily venesections. Stress is laid on the time sequence of the various changes in the circulation and their relation to each other. From the results obtained it was concluded that the elevated cardiac output observed in anemia was not the result of an increase in right atrial pressure or a decrease in tissue oxygen tension, per se. It is suggested that the increased cardiac output may be due to a slowly elaborated or slow-acting humoral agent produced as a result of tissue anoxia.

Intestinal loops brought up through the diaphragm and attached to the myocardium will cause the formation of connections between the vascular supplies of the heart and intestine. The vessels that develop between these two organs are large enough to transmit thick suspensions of barium sulfate. India ink injected in the mesenteric vessel can be readily found on all surfaces of the heart as well as deep within the myocardium.

When the femoral artery is occluded in the anesthetized dog, the pressure in the distal segment usually declines progressively and approaches a static limit asymptotically. This static limit is independent of the original Degrees of vasomotor tone except under extreme conditions. It is concluded that the rapid supervention of reactive hyperemia in the dog vitiates the static limit as an index of vasomotor tone, unless an instantaneous method is developed for its estimation.

In exploring the mechanism of action of vasodilating drugs an attempt has been made to study the enzyme systems of the blood vessels. A transphosphatase of the myokinase type has been found to be present in the coronary arteries of the steer. The activity has been studied under the optimal conditions for this enzyme system. Coronary arterial tissue may be maintained at the temperature of dry ice for a number of days without significant loss of enzyme activity. This will permit an investigation of the effects of drugs upon the activity of these enzymes.

Ground squirrels occasionally show extreme hyperlipemia. Triglycerides predominate, but high levels of cholesterol and phospholipids also occur. Evidence is presented indicating that the hyperlipemia results from periods of dietary caloric excess. During hibernation the triglyceride levels are low. Atheromata were found in the aorta and endocardium in those animals with the highest cholesterol levels.

The “equilibration” method introduced by Bradley to measure circulating “splanchnic blood volume” has been applied to the estimation of “pulmonary blood volume.” The theoretic and practical aspects of the method are analyzed. Values for circulating “pulmonary blood volume” determined in anesthetized dogs were not significantly different from values obtained by the “mean circulation time output” method and were significantly greater than those determined by the “slope output” method. When experimental conditions were varied, relatively parallel changes were recorded by the three methods. It is believed that the “equilibration” method is a valid one for estimating circulating “pulmonary blood volume.”
