Plasma and sera from various sources induced in vitro lysis of human chorionic syncytial trophoblast. In almost all instances the lytic activity was heat labile. Its optimal pH range was between 6.8 and 8.0. Once the trophoblast was exposed to plasma the continued presence of the plasma was not necessary for lysis to occur. The lysis could be inhibited by a variety of substances which could be added hours after the incubation was started and still be effective. The evidence showed that the lytic system required calcium, possessed a thiol group and that a protease was involved. Further, it suggested that two stages were necessary. The first, the initiator of events, was dependent upon a plasma-derived substance, likely complement; the second, the mediator of the lysis, was dependent upon substances inside the syncytial trophoblast, likely lysosomal enzymes.

The postpubertal sex differences in rats, for the hepatic mixed function oxidation of many steroids and drugs, are gonad-dependent in males. Hepatic microsomes of males castrated as neonates had DHA 16α- and 7α-hydroxylase activities in adult hood that were similar to those of adult females, but castration after the’critical period’ does not alter normalenzymic differentiation.

Plasma levels of androstenedione (A) and dihydrotestosterone (DHT) were measured in 7 patients with trophoblastic disease with varying degrees of ovarian enlargement before and after chemotherapy and in 2 patients after oophorectomy. The data presented show that plasma A and DHT levels were elevated in patients with trophoblastic disease with enlarged ovaries and decreased to normal levels after chemotherapy or oophorectomy. Elevated serum levels of both human chorionic gonadotrophic hormone (HCG) and testosterone in these same patients have been reported previously. These investigations suggest that HCG produced by trophoblastic disease is involved in these steroid changes.

Measurements have been made of in vitro glucose metabolism as a function of glucose concentration with all other parameters held as close as possible to normal values. These measurements show that the glucose utilization rate increases in proportion to glucose concentration up to about 150 mg% and then becomes constant at approximately 1.27 g/kg/h. The glucose utilization rates at normal concentrations agree with those observed by others. The transfer of glucose does not seem to be affected by the saturation of the glucose utilization rate, i.e., the permeability of the placental membrane to glucose seemed to remain unchanged at high concentrations.

Morphology of human trophoblast culture has been studied in relation to certain conditions such as duration of the pregnancy, age of the culture, and strength of the trypsinization. ‘Multinucleated giant cells’ appear to be actually fortuitous transversal sections of chorionic buds. Their presence and aspects depend on the degree of trypsinization. Origin of epithelioid polygonal cells and of fibroblast cells is discussed.

Human chorionic gonadotropin can inhibit the blast cell transformation of human lymphocytes, both Phytohemagglutinin-induced and in mixed-lymphocyte reactions. This immunosuppression is noncytotoxic, and specific as shown by the contrasting response of lymphocytes to human serum albumin in similar culture systems. These findings and the high rate of production of this hormone at the surface of the implanting trophoblast support the hypothesis that human chorionic gonadotropin may play an important immunosuppressive role in vivo to prevent the rejection of the fetal allograft by the maternal host.

The average blood flow to the ovaries, fallopian tubes, uterine horns and vagina of estrus rabbits was 1.40 ± 0.14, 1.44 ± 0.13, 0.75 ± 0.06, 0.30 ± 0.04 ml/min/g of tissue, respectively. There was no differences between the right and left organs. Ligation of the ovarian artery resulted in 80–90% decrease in blood flow to the ovary and oviduct, but recovery occurred within 1 and 6 days for the ovary and oviduct, respectively. There was no effect on the uterus and vagina. Ovulatory response to HCG occurred within 1 day and pregnancy after 6 days following ligation.

An important control mechanism in the regulation of fetal cardiac output and tissue blood flow may be the rate of CO2 production. By affecting blood osmotic pressure through changes in bicarbonate ion concentration, fetal CO2 may affect the rate of maternal to fetal water exchange, and, in turn, fetal blood volume, cardiac output and tissue blood flow.

Placing normally pregnant women at bed rest during the third trimester significantly increased the amount of total estrogen excreted into the urine to 122% of the control value, but the concentration of total serum estriol was not significantly altered. Estriol biosynthesis, secretion, conjugation, metabolism, and excretion are regulated by many poorly understood factors, but for purposes of clinical evaluation of the well-being of the infant, bed rest would appear to be another circumstance under which more reliable information can be obtained by blood instead of urine analysis.
