The wilderness whence Jack Stimpfling's voice has been heard during the past 23 years is not the“hideous and desolate waste”feared by Nathaniel Morton, secretary of the Plymouth Colony; neither does it necessitate, as in the case of John the Baptist—as recorded by St. John from Patmos—a diet of“locusts and wild honey.”Instead, it is, in Justice Douglas' words,“the murmur of brooks and the chatter of squirrels—the refuge for automated man.”It is the wilderness of the Missouri Canyon, the“beautiful clifts”(sic)seen by Merriwether Lewis, apparent as one floats down the river during a warm August night, the moon rising above Eagle Rock, and deer watering at the river's edge. The river meanders northward, and enters the vast plains under the Big Sky. You see Square Butte to the left. Yet further to the West, past hillocks carrying clumps of scraggly juniper and clusters of black Angus cattle, you see Haystack Butte and the shining mountains of the Saw Tooth Range of the Rockies.

An angler dreams of a rising rainbow, chooses just the precise fly currently hatching in order to complete the dream of a mountain stream battle that would ensue. The fly must land and drift perfectly so that the trout will be fooled. A camper dreams of just the right location for a warm fire to rest his tent where he will be protected from the wind, yet be able to view the natural beauty surrounding him. A scientist hopes that by pursuing his love of truth and awe of the unknown, he will be contributing to the benefit of his fellow man. Once upon a time, just such a scientist began such a career on the island of Mount Desert. He not only fulfilled his scientific dreams, but also in later years would enjoy the tranquility of nature, open space and the big skies. This is a chronicle of one aspect of his scientific contribution to his fellow man.

The I region of the murine major histocompatibility complex (H-2) encodes two cell surface glycoprotein complexes (Aα:Aβand Eα:Eβ) which exert profound control over many immunologic reactions including humoral and cellular responses to a variety of antigens,i.e., Ir gene effects, stimulation in mixed lymphocyte reaction, graft versus host disease, antigen presentation to T cells by macrophages, collaboration between T and B cells and determination of T cell help or suppression [1-13]. In the past few years, several laboratories have reported that antibodies directed against I region gene products (anti-la) are effective in modulating immune responses bothin vitroandin vivo[14-32, 48-51]. The ability of anti-la antibodies to alter immune responsiveness was attributed to their interaction with macrophage la antigens, thus masking or sterically hindering la epitopes involved in antigen presentation [16,17, 33-35]. However, other reports have suggested that additional mechanisms may be involved [28, 36-39, 49-51]. Experiments with anti-DR (human equivalent of mouse la) and anti-la reagents indicate that these antibodies can induce depression of humoral and cellular responses via active suppression [28, 36-39]. Both T suppressor cells and macrophages have been implicated [28, 36-39]. Additionally, anti-la reagents may deplete antigen specific B cell and helper T cells [48, 50, 51].

Since the recognition twelve years ago that the Ss/Slp proteins of the mouse [1, 2], which had been mapped inside the H-2 gene complex [3], are structurally [5-7] and functionally [8, 9]related to the fourth component of complement, the significance of this map location has remained a mystery. Evidence that the C4 structural genes are closely associated with the MHC in man [10], in the guinea pig [11], and in other species [12, 13], and that structural genes for complement factors C2 and Bf are also MHC-associated, in the mouse [14, 46, 47], in man [10], and in other species [12], has established that this is not a peculiarity of the murine H-2 complex.
