AbstractCarvedilol shows a highly enantioselective first‐pass extraction after therapeuticp.o. doses with preferential extraction of theS‐enantiomer. To investigate, whether the enantioselective first‐pass metabolism is saturable, male Sprague‐Dawley rats were administered increasing single doses ofR/S‐carvedilol (p.o., 5‐30 mg/kg;i.v., 5 and 10 mg/kg), and the individual stereopharmacokinetics were studied. ‐ Like in humans the plasma concentration ofR‐carvedilol exceeded always those ofS‐carvedilol. As expected, a dose‐dependent reduction in oral clearance was observed, while the total clearance after thei.v. doses was constant. Beyond 20 mg/kg an increased plasma half‐life was found for both enantiomers, which is due to a red

AbstractA new type of H1‐agonists resulted from the combination of the essential histamine structure with parts of H2‐antagonists. 2,4‐Disubstituted imidazole derivatives were synthesized by reaction of imidic acid methyl esters with 1,3‐dihydroxypropanone, 1,4‐dihydroxybutanone or 2‐oxo‐4‐phthalimido‐1‐butylacetate in liquid NH3. The imidazole intermediates were converted into histamine analogues by simple deprotection,Gabrielsynthesis followed by deprotection, or by side‐chain elongationviathe nitriles and final hydrogenation. AbstractReaction of 3‐phenyl‐10H‐1,2,4‐triazolo[4′,3′:2,3][1,2,4]triazino[5,6‐b]indole (4) with ethyl chloroacetate gave 10‐carbethoxymethyl‐3‐phenyl‐1,2,4‐triazolo[4′,3′:2,3][1,2,4]triazino[5,6‐b]indole (3). Condensation of3with hydrazine hydrate gave (3‐phenyl‐1,2,4‐triazolo[4′,3′:2,3][1,2,4]triazino[5,6‐b]indol‐10‐yl)acetylhydrazine (5). Reactions of5with a number of aromatic aldehydes, acetophenone, cyclohexanone and D‐galactose gave the corresponding hydrazones6‐12. Condensation of5with acetylacetone gave the pyrazole15. Cyclization of5with CS2gave (3‐phenyl‐1,2,4‐triazolo[4′,3′:2,3][1,2,4]triazino[5,6‐b]indol‐10‐yl)(2‐thiol‐1,3,4‐oxadiazol‐5‐yl)methane (16). AbstractA series of 19N‐alkyl‐diarylpyrrolyl‐acetic and ‐propionic acid derivatives was synthesized and tested. Using bovine blood as enzyme source the inhibition of cyclooxygenase and 5‐lipoxygenase, respectively, was applied to determine the antiinflammatory activity. AbstractFourteen 4,4′‐m‐phenylene‐ and two 4,4′‐p‐phenylene‐bis‐sydnone imine hydrochlorides have been synthesized. All compounds exhibited good solubility in water. AbstractAddition des β‐Alanin‐Derivats6an das Homophthalaldehydmonoacetal5und anschließende LiAlH4‐Reduktion führten zu dem Dihydroxyacetal8, das sich mit Säure zu den 5‐Aminomethyl‐2,6‐epoxy‐3‐benzoxocinen2aund2bcyclisieren ließ. Die Umsetzung von5mit dem Anion des β‐Lactams9lieferte zwei trennbare Diastereomere:10amit u,l‐Konfiguration und10bmit 1,1‐Konfiguration. Das β‐Lactam‐Addukt10a(10b) wurde über das 2‐Benzopyran11a(11b), den Aminoalkohol12a(12b) und das sekundäre Amin13a(13b) in das 5‐(α‐Dimethylaminobenzyl)‐2,6‐epoxy‐3‐benzoxocin3d(3b) überführt. Die relativen Konfigurationen aller Produkte dieser „β‐Lactam‐Route”︁ (10‐13, 3dund3b) wurden durch Auswertung der1H‐NMR‐Spektren aufgeklärt.
