Abstract1‐Acyl and 1,2‐diacyl 1,2,4‐triazolidine‐3,5‐diones proved to be potent anti‐inflammatory agents in rodents at 8 or 20 mg/kg. They were effective against induced edema, pleurisy, and septic shock. Furthermore, these agents were potent in blocking the writhing reflex suggesting that they should be effective against local pain generated by inflammatory processes. These compounds were not lysosomal hydrolytic enzyme or proteolytic enzyme inhibitors in mouse liver, macrophages or human leukocytes. AbstractTwenty 1,3,4‐thiadiazole‐2‐nitrosimines and two 1,2,4‐thiadiazole‐5‐nitrosimines were synthesized and assayed in theBorn‐test for their antiplatelet activity. AbstractThe absorption of D‐glucose by rat thymocytes is reduced to half of control by 30 μmol/L and decreased to 10 % by 100 μmol/L of RE 1492. This is backed by the fact that the absorption of 2‐deoxy‐D‐glucose is inhibited in the same extent. The more hydrophilic oligoamine RE 1888 had an analogous but smaller effect while spermine was ineffective. In a lipid peroxidation model RE 1492 or spermine in a concentration of 100 μmol/L nearly completely inhibited for formation of Fe3+ions when the phospholipid was mimicked by adenosine monophosphate. This suggests an interaction with negatively charged membrane phospholipids. RE 1888 had an equal but smaller effect. The effect of RE 1492 on lipid order and lipid motility was checked on ovine lymphocyte membranes by fluorescence polarization measurements. The steady state as well as the limiting anisotropy as an expression for lipid order is decreased by rising concentrations of RE 1492. The use of several anthroyloxy stearic acids as fluorescent probes also shows an increased lipid motility in several areas of the membrane bilayer. AbstractMichael‐analoge Reaktionen der Verbindungen vom Typ9wurden untersucht: Additionreaktionen mit Acrolein und Methylvinylketon führen in Abhängigkeit von den Reaktionsbedingungen zu den Pyridoindolin16oder14und/oder zu Tetrahydrocarbazolen vom Typ17oder15. Unter Triton B‐Katalyse zeigen die Verbindungen15retro‐Michael‐Eliminierung der Nitromethyl‐Gruppe, gefolgt von Hydrolyse/Decarboxylierung und Dehydrierung zu den zweifach substituierten Carbazolen20. AbstractAmine‐carboxyboranes are potent anti‐inflammatory agents reducing induced edema and pleural effusion at 8 mg/Kg,i.p.They protect against LPS (Salmonella) induced septic shock from 2–8 mg/Kg/day and are effective in blocking pain mediated both locally and centrally. The mode of action of these agents is by blocking release of cytokines from macrophages, thus reducing lysosomal hydrolytic and proteolytic enzyme activities of affected cells. AbstractNach i.p.‐Applikation (50–200 mg) an Ratten zeigten rac.1,S(+)‐1undR(−)‐1nur eine schwach ausgeprägte dosisunabhängige Verlängerung der Hexobarbitalschlafzeit ohne enantioselektive Wirkungsunterschiede. Rac.3verlängerte die Hexobarbitalschlafzeit überhaupt nicht. Die Untersuchung der Gewebeverteilung und der Konzentration im Serum ergab, daß1und3nach i.p.‐Applikation im Serum und Gehirn zwar nachweisbar, aber nicht quantifizierbar waren. Nach i.v. Applikation erwiesen sich dieS(+)‐Enantiomere von1und3überraschenderweise als konvulsiv wirksam. Sie sind in der Wirkungsintensität dem Pentetrazol um den Faktor 2.5‐5 überlegen.R(−)‐1undR(−)‐3verlängerten dagegen nach i.v. Gabe die Hexobarbitalschlafzeit, und zwarR(−)‐1dosisabhängig. Die durchS(+)‐1ausgelösten Krämpfe wurden durch Vorbehandlung mit Diazepam unterdrückt, nicht aber durch Flumazenil. Die EnantiomereR(−)‐1undR(−)‐3sind schwache Agonisten,S(+)‐1undS(+)‐3offensichtlich starke inverse Agonisten. Enantioselektive Unterschiede traten auch bei der Bindung der1‐Enantiomere an Hum
