AbstractA series of 1,2,3,4‐tetrahydro‐6‐methyl‐4‐(substituted phenyl)‐2‐thioxo‐5‐pyrimidinecarboxylic acid methyl esters were synthesized by condensing thiourea with methyl acetoacetate and nonsubstituted or differently substituted benzaldehydes in absol. ethanol using HCl as a catalyst according to theBiginellireaction. AbstractAusgehend vom 5‐Aminoisoxazol1wird das entspr. 4,5‐Diaminoderivat4erhalten, das mit Glyoxal zum Isoxazolo[4,5‐b]pyrazin5cyclisiert. Versuche zur partiellen Reduktion des Pyrazinteiles von5bzw. zur Einführung einesN‐Phenylrestes scheitern an der Labilität des Isoxazolringes. Das zugehörige Tetrahydroderivat6läßt sich aus4und Ethylenglykolditosylat synthetisieren. Das 3‐Benzoylisoxazolin‐5‐on9dient als Edukt für die vierstufige Synthese des Tetrahydro‐4H‐isoxazolo[5,4‐e]1,4‐diazepins13; das Tetrahydro‐isoxazolo[5,4‐e]1,4‐diazepin‐4‐on 18 wird ausgehend von Benzhydroxamsäurechlorid und Cyanessigsäure‐N‐(β‐hydroxyethyl)‐amid dargestellt. Alle neuen Verbindungen werden anhand der spektroskopischen Daten charakterisiert, die Reaktionsabläufe werden diskutiert. AbstractThe combined effects of amino acid antagonists with proven or potential inhibitory activities on aminoacyl‐tRNA synthetases were investigated on the murine leukemic cell line P388 D1. As the best result a summation of the antiproliferative effects was observed. Combinations with established cytostatic agents like platinum complexes or other antitumor compounds also yielded partly additive effects. In experiments performed with asparaginase L‐aspartic acid‐β‐hydroxamate gave synergistic growth inhibition of P388 D1cells in vitro, which was reflected by additive effects against murine leukemia P388

AbstractThe invasion of the body by toxic agents, in addition to the specific effects, causes a stereotype non‐specific response ‐stress‐, and an adaptive phenomenon like drug metabolism. Reserpine or hydrocortisone, causing a typical stress response, produce increased plasma concentrations of corticosterone in rats. ‐ Here it is shown that corticosterone administration once, or for three days twice daily (200 μmoles/kg) increase body resistance to zoxazolamine, methyprylon, and tetraethylammonium bromide, reduce their plasma concentrations and augment zoxazolamine, hexobarbital, and anilinein vitrodrug metabolism (9000 g liver supernatant). AbstractDer Konzentrationsverlauf des Krebschemotherapeutikums Carboplatin wurde im Serum und in der Intraperitonealflüssigkeit (IPF) von sechs Patientinnen bestimmt. Die pharmakokinetische Auswertung zeigte eine rasche Verteilungsphase innerhalb der ersten 4 h nach Infusionsbeginn zwischen beiden Kompartimenten (t1/2Serum = 4.4 h, t1/2IPF = 3.3 h). Die terminale Elimination von Carboplatin aus dem Blutgefäßsystem lag mit t1/2el = 20.3 h deutlich höher als aus der IPF (t1/2el = 10.6 h). Die Verfügbarkeit (berechnet als AUC‐Werte) lag innerhalb von 48 h für die Intraperitonealflüssigkeit zumindest 6 mal höher als für das Serum, wobei 98 % der bestimmten Menge während dieser Zeit aus beiden Kompartimenten eliminiert wurden.
