AbstractEvidence has recently been obtained suggesting that the accumulation of specific endogenous proteins by the nucleus is due both to facilitated transport across the envelope and intranuclear binding. Using recombinant DNA methodology, polypeptide domains containing the signals required for nuclear accumulation have been identified in several karyophilic proteins.

AbstractCertain steroids can react with proteins to form stable, covalent addition products. 16α‐hydroxyestrone and the glucocorticoids, for example, contain ketol moieties which permit them to react non‐enzymatically with proteins by forming Schiffbase rearrangement products with lysine residues. The oral contraceptive agent, 17α‐ethinylestradiol, can add to proteins after being enzymatically activated by microsomal oxidation. Elevated plasma levels of these steroids result in an increased amount of protein modification in vivo. AbstractClonal expansion of T‐lymphocyte populations results from interactions of antigenic structures presented in combination with accessory cells (macrophages) and antibody recognition sites on the surface of T cells. The resulting activation of a membrane phospholipase C plays a crucial role in lymphocyte responses by releasing diglyceride and PIP3. The released diglyceride activates a cellular protein kinase C while PIP3stimulates Ca2+influx. Arachidonic acid released by the action of diglyceride lipase serves as substrate for the synthesis of bioactive eicosanoids (prostaglandins and leukotrienes) which in turn modulate cellular adenyl and guanyl cyclases. AbstractMultidrug resistance (MDR) is developed in a population of somatic mammalian cells in vivo or in vitro when they are selected for resistance to each of a large group of drugs (colchicine, adriamycin, actinomycin D, etc.). It is produced by the amplification of some unknown gene(s) whose product(s) evidently change(s) the plasma membrane permeability of a cell to a selective agent and to other unrelated compounds. A large specific genomic region (150–250 kbp) undergoes amplification in MDR cells selected for the resistance to different drugs. Amplified DNA is located in homogeneously staining regions of chromosomes, double minute chromosomes and small chromatin bodies – episome‐like structures that replicate out of the S‐phase of a cell cycle.
